Objectives: The biotransformation of caffeine has been studied in vitro using human cytochrome P-450 isoenzymes (CYPs) expressed in human B-lymphoblastoid cell lines, namely CYP1A1, 1A2, 2A6, 2B6, 2D6-Val, 2E1 and 3A4, and microsomal epoxide hydroxylase (EH). In addition, CYP 2D6-Met was also studied, in which a valine in the wild type (CYP2D6-Val) has been replaced by a methionine due to a G to A mutation in position 112. Results: At caffeine 3 mmol ⋅l−1, five CYPs (1A1, 1A2, 2D6-Met, 2E1 and 3A4) catalysed the biotransformation of caffeine. Among the enzymes studied, CYP1A2, which predominantly catalysed paraxanthine formation, had the highest intrinsic clearance (160 l . h−1 ⋅ mmol−1 CYP). Together with its high abundance in liver, it should be considered, therefore, to be the most important isoenzyme in caffeine metabolism. The affinity of caffeine for CYP1A1 was comparable to that of its homologue 1A2. CYP2D6-Met, which catalysed caffeine metabolism by demethylation and 8-hydroxylation, also had a relatively high intrinsic clearance (3.0 l . h−1 mmol−1 CYP), in particular for theophylline and paraxanthine formation, with kM values between 9–16 mmol ⋅l−1. In contrast, the wild type, CYP2D6-Val, had no detectable activity. In comparison, CYP2E1 played a less important role in in vitro caffeine metabolism. CYP3A4 predominantly catalysed 8-hydroxylation with a kM value of 46 mmol ⋅l−1 and an intrinsic clearance of 0.60 l . h−1 ⋅ mmol −1 CYP. Due to its high abundance in human liver, the latter CYP may contribute significantly to the in vivo formation of TMU. Conclusion: The findings of this study indicate that i) microsomes from transfected human B-lymphoblastoid cell lines give results close to those obtained with microsomes isolated from human liver, ii) at least four CYP isoforms are involved in caffeine metabolism, iii) at a substrate concentration  〈 0.1 mmol ⋅l−1, CYP1A2 and 1A1 are the most important isoenzymes, iv) at higher concentrations the participation of other isoenzymes, in particular CYP3A4, 2E1 and possibly also CYP2D6-Met, are important in caffeine metabolism, and v) the nucleotide composition at position 1120 of CYP2D6 determines the activity of this isoenzyme in caffeine metabolism.

    Notes: The interfacial tensions of immiscible binary blends of poly(dimethyl siloxane) (PDMS) and polybutadiene (PBD) have been determined as a function of molecular weight and temperature. The technique employed for these measurements takes advantage of recent advances in the determination and analysis of pendant fluid drop profiles. The experimental data are compared to the predictions of square gradient theories and theories based on the diffusion equation approach as developed by Helfand and co-workers. Qualitative agreement is obtained with both types of theory when the Flory-Huggins interaction parameter is taken to be comprised of two terms: a temperature independent term of entropic origin; and an enthalpic term that is inversely dependent upon the temperature.

    Notes: Instrumented impact testing and analysis were performed on an interlaminartoughened carbon fiber epoxy composite (Toray 3900-2/T800H) and a conventional (untoughened) epoxy composite (Fiberite 934/T300). Severity of impact was varied by using a wide range of impact energy. The effect of difference in the tup of the impact instrument was discussed. The residual compressive strengths after compact (CAI's) were measured by using a universal mechanical tester. The impact damage and delamination resistance of the toughened and conventional composites were quantitatively compared. At the same impact energy, the extent of damage was much less severe and the CAI was higher for the toughened composite. Relationships between the CAI's, the state of impact damage, and impact energy were investigated. Impact damage mechanisms for these two composites were not exactly the same. The impact damage of the conventional composite exhibited much greater extents of delamination. By contrast, interlaminar delamination in the toughened composite was found to be significantly suppressed, and the impact damage was more dominated by controlled matrix cracking/fiber damage, instead of catastrophic delamination. As a result, the CAI's of the toughened composite exhibited a monotonous decrease with increase in the impact energy and in the linear dimension of extents of damage.

    Notes: An application of wavelets and multiresolution analysis to density estimation and process monitoring is presented. Wavelet-based density-estimation techniques are developed as an alternative and superior method to other common density-estimation techniques. Also shown is the effectiveness of wavelet estimators when the observations are dependent. The resulting density estimators are then used in defining a normal operating region for the process under study so that any abnormal behavior by the process can be monitored. Results of applying these techniques to a typical multivariate chemical process are also presented.

    Notes: Case histories of 65 incidents in runaway reactions and emergency relief in Taiwan were analyzed and classified into several categories according to their causes, materials involved, equipment types, reaction types, and ignition sources. The cases in reactors and storage tanks were examined in more detail owing to the higher probability or larger potential hazard in these two types of equipments. The most common consequence of the incidents are explosions, fires, and atmospheric release of toxic chemicals. The most severe case was a thermal explosion from an organic peroxide storage area which caused the death of 33 persons. Popping and direct releasing of process chemicals to the atmosphere from relieving devices cause the greatest environmental concerns to the community close to the plants. Runaway reactions in batch type reactors occur frequently due to various operational mistakes. Heat of reaction is the most frequent ignition source of runaway reactions and emergency relief.

    Notes: A model for local heat transfer between a gas-fluidzed bed and a submerged tube is proposed based on combined dense-phase and lean-phase transport. The heat transfer process during dense-phase contact at the tube surface is modeled by packet renewal mechanism and the transfer process during lean-phase contact by fluid convection mechanism. The model predictions show good agreement with experimental local and average heat transfer coefficient data for horizontal tubes.
