    Notizen: Genetic abnormalities in human oral squamous cell carcinomas (OSCC) were examined using restriction landmark genomic scanning (RLGS), a method of two-dimensional gel analysis allowing detection of amplifications and other aberrations in genomic DNA. DNAs from 11 oral tumours as well as from contiguous normal squamous epithelium, were cleaved with the restriction enzyme Not I, [32P] end-labeled and electrophoretically size-fractionated. Following a second digestion employing Hinf I, the further fragmented DNA was again electrophoretically separated. Five fragments/spots were found amplified in at least 64% (7/11) (chromosome nos. 4, 9–12 or 22) of carcinomas, with one of these spots amplified in 100% (chromosome no. 4) of tumour samples. In addition, six other spots were frequently reduced in at least 55% (chromosome nos. 15 or 9–12) of tumour tissues. Further characterization of these common changes is needed to determine if they represent important alterations in OSCC.

    Notizen: Bacteroides gingivalis produces large amounts of proteolytic enzymes which may play a role in its virulence. These enzymes may participate in the tissue destruction of the inflammatory process. In this study, the characteristics of two such enzymes, N-CBz-glycyl-glycyl-arginyl peptidase (N-CBz-Gly-Gly-Arg peptidase) and glycyl-prolyl peptidase (Gly-Pro peptidase) were investigated. The enzymes eluted in different peaks from an anion exchange column. N-CBz-Gly-Gly-Arg peptidase was associated with cells up to 48 h in culture. If cultured longer, it also released in the supernatant. It exhibited optimal activity between pH of 7.0 and 7.5 and was readily inactivated by heat treatment (45°C for 15 min). The enzyme activity was inhibited by p-chloromercuribenzoic acid (PCMB), leupeptin and antipain, suggesting that it is a thiol protease. The B. gingivalis N-CBz-Gly-Gly-Arg peptidase was different from the serum enzyme that digests the same substrate. The serum enzyme was more resistant to heat treatment and was inhibited by diisopro-pylfluorophosphate (DFP). B. gingivalis also produced Gly-Pro peptidase that is released in the supernatant. The enzyme has an optimal pH range between 7.5 and 8.0. The B. gingivalis Gly-Pro peptidase was inhibited by DFP, suggesting that it represents a serine protease. The serum Gly-Pro peptidase did not differ from the bacterial enzyme with respect to its sensitivity to inhibitors; however, they were markedly different in heat sensitivity. The bacterial enzyme was completely inactivated at 60°C for 30 min, whereas the serum enzyme was not inactivated even at 1 h at 60°C.

    Notizen: The need to control and manage the effects of urban, industrial and agricultural development on Lake Biwa, Japan's largest lake, led to the formation of the Lake Biwa Comprehensive Development Project (LBCDP). This article describes the nature and effects of the development, how the LBCDP was conceptualized, implemented and managed, and comments on its effectiveness. Although the basic objective of the project was to promote development of the Keihanshin region by providing additional water, other important objectives included the conservation of the natural environment, the promotion of public welfare, and the restoration of water quality. To achieve objectives on a sustainable basis, extensive political, legislative, social, technological and environmental matters needed comprehensive consideration. This article considers such matters in detail.

    Notizen: Abstract A magnetodynamic model to deal with the acceleration and collimation of jets as a part of the global process of gravitational contraction of the magnetized gas to the central gravitator is discussed. We first review its application to the star forming jet case with several observational supporting evidence. Then, a justification will be given for the extension of this to the AGN jet case, despite of the difference in orders of magnitudes in the scales as well as in the physical parameters. The results of actual application of this magnetodynamic mechanism to the AGN case will be presented together with the discussions for the origin of the radio lobes and hotspots, and we show that this mechanism can explain the enhancement of accretion and the formation of the jet + lobe system consistently with the genetic consideration of the system.

    Notizen: Summary Cyclosporin A (CYA), when administered to CD-1 mice treated with a subdiabetogenic dose of Streptozocin (STZ), exacerbated the STZ-induced insulitis and elevated the plasma glucose levels, parallel to a reduction of the insulin content of the pancreas. The possible mechanisms of CYA-mediated aggravation of STZ-induced diabetes are discussed.

To identify individual chromosomes of a frog karyotype by their fluorescence banding patterns, chromosomes were stained with actinomycin D and 4,6-diamidino-2-phenylindole (DAPI) after incorporation of BrdU during the late S-phase. The chromosomes of three Rana species which were selected for this study (R. ridibunda, R. lessonae and R. japonica) showed well-defined late replication bands. The fluorescence patterns obtained were the reverse of those produced by a 4Na-EDTA Giemsa-staining technique. Fluorescence patterns of the two water frog species (R. ridibunda and R. lessonae) were similar to each other, except for the different fluorescence of the centromeric heterochromatin, which gave extremely bright signals in R. ridibunda but no signal in R. lessonae. Experiments also showed differences between the fluorescence patterns of R. lessonae chromosome 13 in the Italian and Luxembourgian populations. These results sho w that the fluorescence replication banding using actinomycin D and DAPI is very effective in identifying individual frog chromosomes and detecting their structural changes.
