    Notes: Empirical pseudopotential calculations for the entire range of alloy concentrations of cubic Cd1−xMnxTe are presented. The atomic form factors have been deduced empirically by fitting the band structure to spectroscopic data available from the literature. The pseudopotential band structures indicate optical bowing may occur in the alloy Cd1−xMnxTe and have been used to determine the effective masses of the electron and light, and heavy holes, which for CdTe are in agreement with accepted values. The effective masses for Cd1−xMnxTe are given for the first time, and are expressed as first- and second-order polynomials in x. The implications of these results for spectroscopic experiments are discussed. © 1996 American Institute of Physics.

    Notes: A detailed comparison of the empirical pseudopotential method with single and multiple band calculations based on the envelope function and effective mass approximations are presented. It is shown that, in order to give agreement with the more rigorous microscopic approach of the pseudopotential method, structural dependent effective masses and Luttinger parameters must be invoked. The CdTe/Cd1−xMnxTe system has been employed as an example, and the first pseudopotential calculations of quantum wells and superlattices in this material are presented. It is shown that the electron, light- and heavy-hole effective masses tend towards twice their bulk values in the limit of narrow quantum wells. © 1997 American Institute of Physics.

    Notes: The trimethylamine N-oxide (TMAO) reductase of Escherichia coli is a molybdoenzyme that catalyses the reduction of the TMAO to trimethylamine (TMA) with a redox potential of + 130 mV. We have successfully substituted the molybdenum with tungsten and obtained an active tungsto-TMAO reductase. Kinetic studies revealed that the catalytic efficiency of the tungsto-substituted TMAO reductase (W-TorA) was increased significantly (twofold), although a decrease of about 50% in its kcat was found compared with the molybdo-TMAO reductase (Mo-TorA). W-TorA is more sensitive to high pH, is less sensitive to high NaCl concentration and is more heat resistant than Mo-TorA. Most importantly, the W-TorA becomes capable of reducing sulphoxides and supports the anaerobic growth of a bacterial host on these substrates. The evolutionary implication and mechanistic significance of the tungsten substitution are discussed.

    Notes: Abstract The fdhF gene of Escherichia coli, coding for at least one component of benzyl viologen-linked formate dehydrogenase (FDH-BV) activity, was isolated on a ColE1-fdhF hybrid plasmid from the Clarke and Carbon colony bank.Endonuclease restriction maps of this plasmid and its pBR322-subcloned derivative, pLW06, were constructed. Various hybrid plasmids were further obtained by deletion of endonuclease-cleaved fragments from pLW06 DNA. Their complementation pattern was analyzed after introduction into different fdhF mutant strains. The fdhF gene was shown to be located on a 5.5 kb BamHI-PvuII-DNA fragment, which restored FDH-BV activity to the wild-type level.

    Notes: Summary Two classes of mutants defective in benzyl viologen-linked formate dehydrogenase (FDH-BV) activity were isolated from Escherichia coli K12. Class I consisted of four mutants which were specifically devoid of FDH-BV activity. Their mutation mapped between the ssb and melA genes at 92 min on the genome, at a site recently designated fdhF by Pecher et al. (1985). The direction of transcription of gene fdhF was found to be counterclockwise on the E. coli chromosome in one Mudl(Apr lac) fusion mutant. Expression of the lac operon in this mutant was induced by formate and repressed by nitrate, nitrite or trimethylamine N-oxide. It was found to be dependent on the positive control exerted by the fdhA, B and C genes, possibly involved in selenium incorporation, and by an hydB gene affecting the formate hydrogenlyase pathway. Class II, represented by one Mudl(Apr lac) mutant, exhibited no FDH-BV activity and a reduced level of hydrogenase activity. The relevant fdv mutation was shown to be located at 58 min and to affect the expression of fdhF.
