    Description / Table of Contents: Abstract A total of 523 strains representing 35 species related to food fermentation organisms of practical importance were investigated for their potential for formation of biogenic amines (BA). The investigation was performed with resting cells in phosphate buffer (pH 5.5) and the formation of the following BAs was followed: putrescine, cadaverine, histamine, tyramine and 2-phenylethylamine. No potential was observed in species ofLactococcus, Leuconostoc, Pediococcus, Streptococcus and severalLactobacillus spp., such asL. pentosus andL. sake. A remarkable potential to form BA was observed in strains of carnobacteria,Lactobacillus buchneri, L. curvatus, L. reuteri, Staphylococcus carnosus and, to a lesser extent, inL. alimentarius, L. brevis, L. bavaricus, L. delbrueckii ssp.lactis, Micrococcus spp. andS. piscifermentans. In well known species with a practical function in the fermentation of dairy products, wine or cabbage a potential was observed for few strains only. In view of their role as starters in food fermentation, or their potential use in protective cultures and as probiotics, BA formation by the organisms has to be taken into consideration by selecting appropriate strains.
    Notes: Zusammenfassung Das Potential zur Bildung von biogenen Aminen (BA) wurde bei 523 Stämmen, die 35 Spezies aus dem Verwandtschaftsbereich der Lebensmittelfermentationsorganismen zugehörten, untersucht. Zu diesem Zweck wurden ruhende Zellen in Phosphatpuffer (pH 5,5) verwendet und die Bildung der folgenden BA verfolgt: Putrescin, Cadaverin, Histamin, Tyramin, 2-Phenylethylamin. Bei Spezies der GeneraLactococcus, Leuconostoc, Pediococcus, Streptococcus und einigenLactobacillus spp., darunterL. pentosus undL. sake fehlte dieses Potential. Eine deutliche Bildung von BA wurde bei Stämmen von Carnobakterien,Lactobacillus buchneri, L. curvatus, L. reuten undStaphylococcus carnosus beobachtet. Weniger ausgeprägt war die Aminbildung beiLactobacillus alimentarius, L. brevis, L. bavaricus, L. delbrueckii ssp.lactis, Micrococcus spp. undStaphylococcus piscifermentans. Ein Potential bei Spezies, die bei der Fermentation von Milch, Wein und Sauerkraut bekanntermaßen erwünscht sind, wurde nur bei wenigen Stämmen gefunden. In Anbetracht der Bedeutung bestimmter Spezies als Starterkulturen bei Lebensmittelfermentationen und der möglichen Anwendung als Schutzkulturen oder Probiotika ist die mögliche Bildung von BA durch Auswahl geeigneter Stämme ausreichend zu berücksichtigen.

    Notes: Abstract The purpose of the study was to investigate the cytokine gene expression patterns and immunohistochemical characteristics of genitoanal warts in order to obtain a clue as to the immunological mechanisms possibly relevant for wart regression or persistence. We analysed surgically removed warts from 11 patients, 2 of whom were immunosuppressed. Lesions of five of the nine otherwise healthy individuals were additionally treated with intralesional interferon-γ (IFNγ) prior to surgery. Invasion of CD4 + T cells into the papillomas and HLA-DR and ICAM-1 expression on keratinocytes were found in two otherwise healthy patients and were intensified by intralesional IFNγ in four of five patients. The mRNA expression patterns in seven of eight nonrecurrent warts were compatible with a predominant TH1 or mixed TH1/TH2 cytokine profile. In contrast, in recalcitrant warts of three patients (one healthy, two immunocompromised) histological signs of immunoreactivity and TH1-like cytokine mRNA expression were not detected. In recurrent warts of a renal transplant patient, IL-4 and IL-5 mRNA expression was repeatedly found suggesting a predominant TH2 response. In conclusion, immunoreactivity to genitoanal warts such as T-cell infiltration, HLA-DR and ICAM-1 expression was associated with a predominant TH1 or mixed TH1/ TH2 cytokine mRNA expression profile.

    Notes: Summary Acetylcholine receptor, isolated in Triton X-100 on a cobra α-neurotoxin affinity column was incorporated into unilamellar phospholipid vesicles by a detergent depletion method using Amberlite XAD-2. Vesicles of an average diameter of 25 nm were formed, as verified by freeze-fracture electron microscopy and gel filtration. 85 to 95% of the α-bungarotoxin binding sites of the reconstituted acetylcholine receptor were oriented towards the outside of the vesicles. In the reconstituted receptor one molecule of residual Triton X-100 per 2.5 α-bungarotoxin binding sites on the receptor molecule could be assessed. The reconstituted protein was not accessible to papain digestion, whereas the pure acetylcholine receptor, solubilized by Triton X-100 was split into smaller polypeptides under the same condition. Reconstituted acetylcholine receptor and receptor-rich membranes did not exhibit the same behavior as measured by use of a potentiometric dye. This is interpreted as an irreversible alteration of at least 95% of the receptors purified in the presence of Triton X-100. Furthermore, it could be shown that the fluorescence intensity changes induced by carbamylcholine in receptor-rich membranes did not reflect ion fluxes, but conformational changes of the protein or a displacement of the dye from the protein.

    Notes: Abstract The aim of the present study was to assess a possible role of the tumour suppressor genep53 in neuroendocrine (Merkel cell) carcinoma of the skin with regard to tumour development and tumour progression. p53 was investigated in a series of routinely processed Merkel cell carcinomas, with application of four different p53 antibodies (CM-l, PAb1801, D07, and PAb240) to 25 carcinomas and screening forp53 mutations of exons 4–8 by single-strand conformation polymorphism (SSCP) analysis in 9 cases. All 25 tumours in the present series showed the characteristic microscopic and immunohistochemical features of Merkel cell carcinoma of the skin. In 5 of the 25 Merkel cell carcinomas investigated 5–10% of tumour cell nuclei showed a positive p53 reaction with at least one anti-p53 antibody. A few scattered p53 positive nuclei were found in an additional 9 cases. The remaining 11 cases completely lacked p53 immunostaining. SSCP analysis of exons 4–8 revealed no significant alterations in the mobility shift of the single strand DNAs in the five cases with 5–10% p53-immunoreactive tumour nuclei or in five cases lacking p53 accumulation significant. Our results suggest that alterations of the p53 gene play only a minor part in the development or progression of Merkel cell carcinoma of the skin.

    Notes: Abstract  An extranodal high-grade B-cell lymphoma, centroblastic type, with exclusive involvement of the heart, stomach and small bowel was detected at post-mortem examination following the death of an 80-year-old man. Autopsy revealed massive cardiomegaly with a total heart weight of 1800 g owing to an intramyocardial tumour involving the right ventricle, and multiple mucosal tumour plaques and nodules in the stomach and small bowel. The case highlights the difficulties of diagnosing cardiac lymphoma clinically even in the presence of a large tumour mass.

    Notes: A new technique has been employed for producing Model 11 Biomer bladders with integrally textured blood-contacting surfaces. For the past year, we have been routinely fabricating integrally textured polymeric surfaces from negative-replica silicone molds, which are a high-fidelity facsimile of conventionally flocked bladders. The objectives of this investigation are threefold: (1) To develop a blood-contacting surface that promotes the formation of a stable pseudoneointimal lining; (2) to investigate means of accelerating replacement of initial fibrin coagulum by a collagenous lining; and (3) to characterize the resultant pseudoneointimal lining biochemically, histologically, and ultrastructurally. Systematic in vivo studies were undertaken to evaluate the new surface in terms of its ability to form a thin, stable, biologic lining. Control experiments were carried out to compare results with previous experience utilizing polyester fibril flocked surfaces. Additional studies were conducted to define the effects of cell seeding of integrally textured surfaces. A total of 16 experiments have been conducted to date; eight bladders were seeded with allogeneic fetal cells prior to implantation and eight bladders were unseeded, to serve as controls. In vivo trials of seeded surfaces have resulted in the formation of strong, cellular, and highly collagenous linings, with collagen contents varying between 31% and 65%. These linings contain viable cells and thus exhibit high cellular activity, as evidenced by postexplant cell culture techniques. This indicates that a living, viable pseudoneointima has been generated. On the other hand, control (nonseeded) linings do not produce significant amounts of collagen in their linings.
