    Notes: Using surface micromachined samples, we demonstrate the accurate measurement of cantilever beam adhesion by using test structures which are adhered over long attachment lengths. We show that this configuration has a deep energy well, such that a fracture equilibrium is easily reached. When compared to the commonly used method of determining the shortest attached beam, the present method is much less sensitive to variations in surface topography or to details of capillary drying. © 1999 American Institute of Physics.

    Notes: The concept that activation of MHC class I-restricted CD8+ cells entirely depends on help from MHC class II-restricted CD4+ T cells has recently been supplemented with an alternative model in which CD8+ cells can directly be activated by MHC class I-expressing professional antigen-presenting cells (APC), which are able to deliver an accessory signal. The authors analysed the role of CD28-mediated costimulation for T helper cell-independent activation of purified human CD8+ T cells in two different in vitro models. Freshly isolated CD8+ cells could be activated (proliferation, IL-2 production and cytotoxic activity) by anti-CD3-presenting FcγR+ mouse cells transfected with the human CD28 ligand, CD80, as the only accessory signal. On the other hand, activation of CD8+ cells by allogeneic MHC class I on EBV-transformed B cells, which express two different CD28 ligands, CD80 and CD86, also proceeded very efficiently (proliferation, cytotoxic activity and CD25 expression), but was either not, or only partially, blocked by anti-CD80 and anti-CD86 MoAb or CTLA-4Ig. This indicates that other costimulatory signals are also effective, and that CD28 triggering is not absolutely required for initial T-cell activation. CsA and CD80/CD86-blocking agents were synergistic in completely inhibiting activation of CD8+ cells in the MLR with allogeneic B-cell lines. This combination also induced non-responsiveness of CD8+ cells upon restimulation in the absence of blocking agents. Therefore, although professional APC can apparently provide multiple costimulatory signals for direct activation of CD8+ T cells, the signal derived from CD80/CD86 is unique in providing CsA-resistance.

    Notes: summary The purpose of the study was to evaluate the effect of early orthodontic treatment of functional unilateral posterior crossbite (FUPC) and to evaluate temporomandibular function in the short and long term. Orthodontic treatment consisted of slow expansion of the maxillary dental arch by means of a removable expansion plate, with flat coverage of the occlusal surfaces of the left and right posterior teeth. Evaluation of the occlusion showed a strong correlation between the crossbite side and the direction of the RCP-ICP slide and with the side of first occlusal contact in RCP. In 26 of 27 children that were treated (one withdrew), the average time required for correction of the crossbite was 7 months, followed by a retention period, on average, for 6 months after completion of treatment. Early orthodontic treatment resulted in an elimination of occlusal disturbances, and the crossbite remained stable on follow-up during an average of 8 years after the retention period, except in two children with a class III tendency. In nine other children an orthodontic anomaly had developed requiring further treatment (two children showed crowding and seven children showed a class II malocclusion). This study showed that FUPC can be treated adequately by early orthodontic intervention; however, its correction does not guarantee the absence of functional disturbances at a later age. Therefore, FUPC should be treated early in order to achieve normal growth and development rather than to prevent temporomandibular disorders.

    Notes: Abstract In food chain models the lowest trophic level is often assumed to grow logistically. Anomalous behaviour of the solution of the logistic equation and problems with the introduction of mortality have recently been reported. As predation on the lowest trophic level is a kind of mortality, one expects problems with these food chain models. In this paper we compare two formulations for the lowest trophic level: the logistic growth formulation and the mass balance formulation with resources modelled explicitly. We examine the effects of both models on the dynamic behaviour of a tri-trophic microbial food chain in a chemostat. For this purpose bifurcation diagrams, which give the existence and stability of the equilibria of the nonlinear dynamic system, are used. It turns out that the dynamic behaviours differ in a rather large region of the control parameter space spanned by the dilution rate and the concentration of the resources in the reservoir. We urge that mass balance equations should be used in modelling food chains in chemostats as well as in ecosystems.

    Notes: Abstract We present preliminary results from deep optical searches of small (≤∼5 arcmin2) GRB error boxes determined using the Third Interplanetary Network (IPN3). Two of these fields also have been found to have historical OT events located within the IPN3 error boxes. We compare the preliminary results of these searches to those reported for the larger IPN1 error boxes. The small size of the IPN3 error boxes should allow a test of the hypothesis suggested by the IPN1 study that there are excess QSOs associated with the GRB fields.

    Notes: Abstract The ability of dexamethasone and prednisolone (corticosteroids), FK506 and cyclosporin A (T cell immunosuppressants), and of nitraquazone and rolipram (phosphodiesterase IV inhibitors) to inhibit cytokine production by stimulated human blood was investigated. Heparinized human blood obtained from normal healthy volunteers was stimulated with phytohemagglutinin (PHA) in the presence or absence of drug. After different incubation times, supernatant levels of interleukin (IL)-2, IL-5, granulocyte-macrophage colony stimulating factor (GM-CSF) and interferonγ (IFN-γ) were quantified by ELISA. Dexamethasone strongly inhibited the production of IL-5 (IC50 = 0.004 µM), was less potent against IL-2 and IFN-γ (IC50 = 0.02–0.05 µM) and showed a relatively weak effect against GM-CSF (IC50 = 0.6 µM). Similarly prednisolone potently suppressed IL-5 generation (IC50=0.05 µM), displayed a more modest activity on IL-2 and IFN-γ (IC50 = 0.2–0.3 µM) and exerted only partial effects (43% inhibition at 1 µM) on GM-CSF. FK506 strongly suppressed the production of IL-2 (IC50 = 0.01 µM) and GM-CSF (IC50 = 0.03 µM), but was inactive (〈30% inhibition at 1 µM) against IL-5 and IFN-γ. Similarly, cyclosporin A reduced the generation of IL-2 (IC50 = 0.4 µM) and GM-CSF (IC50 = 0.6 µM) while barely affecting the other two cytokines. Nitraquazone and rolipram were most active in reducing the production of IL-5 (IC50 = 0.8 and 1.3 µM, respectively), while their potency against IL-2, GM-CSF and IFN-γ was 3–6 times lower, with IC50's between 2.4 and 8.0 µM. These data indicate that corticosteroids, T cell immunosuppressants and phosphodiesterase IV inhibitors affect cytokine production by PHA-stimulated human blood cells in a differential and “pharmacotypical” manner.

    Notes: Abstract We investigated the potential of cross-over to the serotonin receptor (5HT3) antagonist ondansetron after protection failure with tropisetron. Several cases of complete protection were observed. These limited data suggest that there is an indication for retreatment with a different 5HT3 antagonist after an initial failure to another and also stress the need and relevance for comparative studies between 5HT3 antagonists.
