    Notes: The expression of CD44 isoforms (CD44std, CD44v6, CD44v10) was investigated by an immuno-histochemical technique in 42 basal cell carcinomas (BCC) of the superficial and nodular variety. All BCCs studied displayed very low amounts of CD44std, a receptor for hyaluronic acid. Except for single CD44std-positive cells located preferentially in the central parts of the BCC nests, the bulk of the tumour formations were CD44std-negative. CD44v6 showed a heterogeneous distribution pattern accentuated in the peripheral palisading tumour cells. In superficial BCCs, the labelling intensity for CD44v6) increased with the size of the tumour nests. CD44v10 was not detectable in BCCsOur findings support the notion that CD44v6 is not linked to the metastatic proclivity of tumours originating from keratinocytes. We suggest that the very low expression of the receptor for hyaluronic acid (CD44std) may be one of the factors which block the formation of metastases from BCCs.

    Notes: Increasing evidence suggests that the retinmd-X receptors RXR(-α,-β-γ) play a crucial part in regulating the transcription a I activity of several steroid hormone receptors, including 1.25-dihydroxyvitamin D3 receptors (VDR) and retinoic acid receptors (RAR-α,-β,-γ). We developed a new technique for immunohistochemical in situ detection of RXR receptors, and investigated the localization of RXR-a in normal and psoriatic human skin using a recently raised corresponding specific antibody. RXR-α positive cells related to the skin were phenotyped by sequential sections and a double-labelling procedure for the simultaneous demonstration of this nuclear receptor and cell membrane antigens, as well as cytokeratin 10. HLA-DR and vimentin.Our findings indicate that: (i) RXR-m is strongly expressed in normal and psoriatic human skin: (ii) most of the cell types in normal human skin, including keratinocytes, melanocytes, fibroblasts andskin immune cells such as Langerhans cells, reveal strong nuclear immunoreactivity for RXR-a, with less cytoplasmic staining; (Hi) altered levels or distribution of RXR-n in the skin do not appear to be involved in the genesis of psoriasis vulgaris, but subepidermal and subcellular distribution suggest a function of RXR-α in the transition from proliferation to differentiation in epidermal keratinocytes: (iv) expression in the hair follicle points to a contribution from RXR-α to hair growth.

    Notes: Abstract Aims/hypothesis. Major histocompatibility complex class II molecules present antigenic peptides to T-cells and have an important role in T-cell thymic education. The mechanism by which major histocompatibility complex alleles confer a high genetic risk for autoimmune diabetes is not known. One hypothesis is that during positive thymic selection, the peripheral T-cell repertoire is modelled by major histocompatibility complex-restricted presentation of self major histocompatibility complex molecule-derived peptides, some of which mimic tissue autoantigens. The sequence similarity between a known T-cell epitope of glutamic acid decarboxylase-65, 509:VPPSLRTLED and the non-obese diabetic mouse class II major histocompatibility complex molecule I-Ag7 86:VPTSLRRLEQ is consistent with this. Methods. We measured spontaneous proliferation of peripheral T-cells from non-obese diabetic mice and other, non-diabetes-prone strains, to the I-Ag7 86–101 and glutamic acid decarboxylase-65509–524 peptides, binding of these peptides to intact I-Ag7 and assessed the effect of tolerance induction on diabetes development, by injecting young non-obese diabetic mice with high doses of peptide. Results. T-cells from non-obese diabetic, but not other mice strains, spontaneously proliferate to the I-Ag7 86–101 and glutamic acid decarboxylase-65509–524 peptides, but not control peptides. Both test peptides bind I-Ag7. Tolerance induction prolongs diabetes-free survival in non-obese diabetic mice when either the I-Ag7 86–101 or glutamic acid decarboxylase-65509–524 peptide, but not control peptide, is used. Conclusion/interpretation. A peptide from the unique class II major histocompatibility complex, diabetes-susceptibility molecule, I-Ag7, presented by I-Ag7 is a target of T-cell responses in diabetes-prone non- obese diabetic mice and tolerance induction against the peptide offers appreciable protection against the development of diabetes.     Description / Table of Contents: Abstract. We report on a 23-year-old man with hidrotic ectodermal dysplasia of the Clouston type showing a severe expression of his gene defect. As a prominent feature of this case, we describe the presence of verruciform keratoderma of hands and feet, and we visualize these skin changes by means of a new imprinting technique.
    Notes: Zusammenfassung. Ein 23jähriger Patient leidet seit Geburt an einer hidrotischen ektodermalen Dysplasie vom Typ Clouston in besonders schwerer Ausprägung. Als ein eindrucksvolles Symptom bestand bei diesem Patienten eine ausgeprägte palmoplantare Keratodermie mit Ausbildung warzenähnlicher Strukturen, die mittels einer neuen Abformtechnik durchlichtmikroskopisch dargestellt werden.

    Notes: Summary A 45-year-old man with end-stage renal disease underwent a cadaveric kidney transplantation. The allograft had to be removed 10 days after transplantation because of an acute vascular rejection. After explantation, the patient suffered from a life-threatening infection withStaphylococcus epidermidis involving lungs, eyes and liver for 11 months. Despite adequate therapy including vancomycin followed by teicoplanin, he developed spondylodiscitis requiring repeated surgical interventions. The definitive cure was achieved by a sequential therapy with chloramphenicol and quinupristin/dalfopristin.

    Notes: Abstract  The performance of a new commercial system for the identification of different groups of gram-positive cocci [BBL Crystal Gram-Positive (GP) Identification System; Becton Dickinson Microbiology Systems, Germany] was evaluated in comparison with two currently used commercial systems, the API Staph and the API Strep (bioMérieux Diagnostic, Germany). A total of 191 strains from seven different gram-positive genera comprising 32 different species were tested. For the BBL Crystal GP system, the correct identification rate without additional tests was 89.5% at the species level and 97.9% at the genus level. The findings suggest that the newly introduced BBL Crystal GP ID system provides an accurate method for the identification of gram-positive cocci, with an overall rate of correct species identification of about 90%, similar to that of the established API systems. Its major advantage is the extended spectrum of taxa included in a single test panel in contrast to the two different API test kits. Furthermore, the simplicity of use and the safe and rapid handling in a closed system conveniently accommodate existing laboratory workflow.
