    Notes: A tandem mass spectrometer consisting of a double-focusing magnetic-sector mass analyzer in series with a time-of-flight (TOF) mass analyzer has been designed and constructed. The TOF analyzer was a quadratic-field ion mirror. The method of ionization used was matrix-assisted laser desorption/ionization. Precursor ions were mass selected with the magnetic-sector analyzer, and time focused by ion bunching prior to fragmentation in a collision cell. The fragment ions were mass analyzed with the TOF analyzer, which possessed the property that residence times (i.e., times of flight) in the mirror were independent of ion velocity. The theoretical background to the instrumental design is presented. Experimental results are presented, showing resolutions of 4000 in fragment ion spectra and demonstrating effective high-energy collision-induced decomposition of peptide molecule ions. © 1998 American Institute of Physics.

    Notes: Abstract  Numerous techniques are used to maintain intraoperative heart viability. The studies presented here evaluated heart function and metabolism after various periods of preservation up to 4 hours with intermittent warm and cold blood perfusion. Using a het-erotopic heart model cooled to 10°C and maintained for 1, 2, 3, and 4 hours, various preservation techniques were compared. Changes in myocardial metabolism were determined from substrate uptakes and biopsy samples of the left ventricular muscle for high-energy phosphates. Preservation techniques included: (1) sustained hypothermia, (2) 1 or 2 hours of sustained warm blood perfusion with fibrillation, (3) intermittent cold blood perfusion during 2, 3, and 4 hours of preservation, (4) intermittent warm blood perfusion during 2, 3, and 4 hours of preservation and (5) a control group (no preservation). Normothermic fibrillation had no effect on postpreservation functional or metabolic parameters. Sustained hypothermia reduced functional recovery proportional to the length of ischemia. The cold intermittent procedures maintained function and metabolism better than sustained hypothermia, while warm intermittent preservation maintained function and metabolism at control levels throughout the recovery period for all preservation techniques. Changes in ATP mirrored the functional changes. Creatine phosphate (CP) was markedly reduced during heart isolation and preservation and exceeded the control by 100% during reperfusion. For operative procedures of 2 hours or less, functional and metabolic recovery was not affected by the various preservation methods applied. Warm intermittent perfusion during hypothermie preservation offered the best protection for the myocardium. The warming cycles during hypothermia may provide some degree of preconditioning and protect the myocardium during reperfusion.

    Notes: Abstract : The transcriptional mechanism underlying amyloid precursor protein (APP) regulation in primary neurons during development was investigated. We observed an approximately threefold elevation of APP mRNA levels in differentiating rat hippocampal neurons between day 1 and day 7 in culture and in rat brain hippocampi between embryonic day 18 and postnatal day 3. When an APP promoter construct extending to position -2,832 upstream from the main transcriptional start site was transfected into primary rat hippocampal neurons, promoter activity increased from day 1 until reaching a maximum on day 7 in culture. This increase in APP promoter activity was correlated more closely with the time course of expression of the synaptic vesicle protein synaptophysin, an indicator of synaptogenesis, than with neurofilament accumulation, an indicator of neuritogenesis. Transfection of 5′ APP promoter deletions and internal block mutations indicated that the CTCF binding domain designated APBβ was the primary contributor to the increase in APP promoter activity. Furthermore, the binding of transcription factor CTCF to the APBβ element increased approximately fivefold between day 1 and day 7, whereas the binding of USF to the APBα sequence increased only twofold. These results suggest that CTCF is pivotal for the up-regulation of APP expression during synaptogenesis in primary neurons.

    Notes: Abstract: Olfactory receptors are difficult to express functionallyin heterologous cells. We found that olfactory receptors traffic poorly to theplasma membrane even in cells with neuronal phenotypes, including cell linesderived from the olfactory epithelium. Other than mature olfactory receptorneurons, few cells appear able to traffic olfactory receptors to the plasmamembrane. In human embryonic kidney 293 cells and Xenopus fibroblasts, olfactory receptor immunoreactivity overlapped with a marker for the endoplasmic reticulum (ER) but not with markers for the Golgi apparatus or endosomes. Except for the ER, olfactory receptors were therefore absent from organelles normally involved in the plasma membrane trafficking of receptors. Olfactory receptors truncated prior to transmembrane domain VI were expressed in the plasma membrane, however. Co-expression of the missing C-terminal fragment with these truncated receptors prvented their expression in the plasma membrane. Intramolecular interactions between N- and C-terminal domains joined by the third cytoplasmic loop appear to be responsible for retention of olfactory receptors in the ER of heterologous cells. Our results are consistent with misfolding of the receptors but could also be explained by altered trafficking of the receptors.

    Notes: Pulsed laser ablation of aqueous medium irradiated under conditions of temporal confinement of thermal stress is described. Time-resolved measurements of laser-induced transient stress waves with simultaneous imaging of ablation process by laser-flash photography were performed. Stress transients induced in aqueous solution of K2CrO4 by ablative nanosecond laser pulses at 355 nm were studied by a broad-band lithium niobate acoustic transducer. Recoil momentum upon material ejection was measured from the temporal profiles of the acoustic transducer signal as a function of incident laser fluence. Cavitation bubbles produced in the irradiated volume during the tensile phase of thermoelastic stress were shown to drive material ejection at temperatures substantially below 100 °C. Experimental data are evident that nanosecond-pulse laser ablation of aqueous media (when temporal stress-confinement conditions are satisfied) include the following two main stages of material ejection: (1) ejection of water microdroplets due to expansion and rupture of subsurface cavitation bubbles; (2) ejection of liquid streams with substantial volume upon collapse of initial crater and large cavitation bubbles in the depth of irradiated volume (after coalescence of smaller bubbles). © 1995 American Institute of Physics.

    Notes: Three-dimensional Ge islands between 15 and 200 nm in size were found to grow only on Si(111) surface windows in ultrathin SiO2 film after Ge deposition and subsequent SiO2 decomposition. The size of Ge islands gradually decreased as the Ge thickness decreased. Pseudomorphic two-dimensional Ge layers with the 5×5 structure formed in surrounding areas of the windows. The windows were produced by selective thermal SiO2 decomposition induced by focused electron beam irradiation. These results suggest a new technique for nanometer-scale Ge island fabrication at given points on Si surfaces. © 1998 American Institute of Physics.

    Notes: An approach for calculating the interaction between a hard superconductor and a permanent magnet in the field-cooled case is proposed. The exact solutions were obtained for the point magnetic dipole over a flat ideally hard superconductor. We have shown that such an approach is adaptable to a wide practical range of melt-textured high-temperature superconductors' systems with magnetic levitation. In this case, the energy losses can be calculated from the alternating magnetic field distribution on the superconducting sample surface. © 1998 American Institute of Physics.
