Notes: Abstract The effect of (+)-5-oxo-D-prolinepiperidinamide monohydrate (NS-105), a novel cognition enhancer, on adenylate cyclase activity was investigated in cultured neurons of the mouse cerebral cortex. NS-105 (10–7 and 10–6 M) inhibited forskolin-stimulated cyclic AMP formation, an action that was dependent on pertussis toxin-sensitive G proteins. Conversely, in pertussis toxin-pretreated neurons, NS-105 (10–7 –10–5 M) significantly enhanced the forskolin-stimulated cyclic AMP formation, and this action was completely reversed by cholera toxin. A metabotropic glutamate receptor agonist (1S, 3R)-1-aminocyclopentane-1, 3-dicarboxylic acid (1S, 3R-ACPD) produced similar bi-directional actions on the cyclic AMP formation. Both of these inhibitory and facilitatory actions of NS-105 and 1S, 3R-ACPD were blocked by L(+)-2-amino-3-phosphopropinoic acid (L-AP3). NS-105 (10–6 M) and 1S, 3R-ACPD (10–4 M) significantly enhanced isoproterenol- and adenosine-stimulated cyclic AMP formation. The enhancement of such Gs-coupled receptor agonists-stimulated cyclic AMP formation was also produced by quisqualate but not by L(+)-2-amino-4-phosphonobutanoate (L-AP4). The phosphoinositides hydrolysis was enhanced by 1S, 3R-ACPD (10–4 M) but not by NS-105 (10–6 M), however, 1S, 3R-ACPD-induced increase in phosphoinositides turnover was attenuated by NS-105. These findings suggest that NS-105 stimulates metabotropic glutamate receptor subclasses that are coupled both negatively and positively to adenylate cyclase, but it acts as an antagonist at the receptor subclasses that are linked to phosphoinositides hydrolysis.

Notes: Abstract An unusual ultrastructure for neurofibrillary tangles, which has not been described so far, is presented in a case of Alzheimer’s disease. This profile consists of parallelly arranged paired helical filaments and criss-cross tubular profiles that are arranged at regular interval of 300–500 nm, resembling rattan bamboo blind or Japanese sudare-like profiles. Coexistence of Hirano bodies in the same neuron is infrequently encountered.

Notes: Abstract The positron lifetime of undoped Liquid-Encapsulated Czochralski (LEC)-GaAs and Si-doped (1.3×1018 cm−3) LEC-GaAs was measured before and after irradiation with protons (dose 1×1015/cm2, 15 MeV). In Si-doped GaAs, the decrease of positron lifetime at temperatures between 10 and 300 K are due to the decrease of the positron-diffusion length and the increase of the effective shallow traps such as antisite GaAs. The annealing stage of the proton-irradiation-induced defects which show the different behavior from that of electron-irradiation-induced defects suggests that proton irradiation creates more complicated defect complexes, containing vacancies rather than isolated vacancy-type defects or simple complexes which have been observed during electron-irradiation processes. Above 700 K, proton-irradiation-induced defects such as vacancy-type defects and simple vacancy complexes are almost annealed out, while Si-induced defects such as SiGa-VGa complexes cannot be annealed out above 973 K.

Notes: Poly(methacrylic acid) (PMA) and poly(acrylamide) (PAAm) with a small mole fraction of naphthol (NpOH) residues were prepared by polymerization of methacrylic acid or acrylamide with three position isomers, methacryloylaminonaphthol. The substituent effect on the apparent pK values (pKobsd), determined by spectroscopic pH titrations in aqueous solution, was observed for the NpOH-labeled PAAm. The electrostatic surface potential (ψ) for PMA was calculated from the difference in the pKobsd values for the labeled PMA and PAAm. The obtained ψ value decreased as the spacing between the OH moiety and the polymer main chain increased. This clearly indicates that the ψ value is a sensitive function of the distance from the polyion surface.
