    Notes: Background The two main features of asthtna are bronchial hyperresponsiveness and inflammation. The inflammatory response in asthma consists of infiltration and activation of a variety of inflammatory cells including neutrophils. Our previous studies have shown that stimuhited neutrophil supernatants cause hyperresponsiveness of human bronchial tissue in vitro.Objective To investigate the effect of the sensitization status of the tissue and the albumin concentration used to prepare supernatants on the response of human bronchial tissue to stimulated neutrophil supernatants.Methods Neutrophil supernatants were prepared from human isolated blood in the presence of varying concentrations of albumin (0%, 0.1% and 4%), Neutrophil supernatants were added to sensitized and non-sensitized human isolated bronchial tissue which was stimulated with electrical field stimulation (EFS) (20 s every 4min). Receptor antagonists specific for the prostaglandin and thromboxane (10−7M GR3219I), platelet activating factor (10−6M WEB 2086), leukotriene D4 (10−6M MK-679) and neurokinin A (10−7M SR48968) receptors were used to identify neutrophil products responsible for the effects observed in the bronchial tissue. Results In non-sensitized human bronchial tissue, stimulated neutrophil supernatants induced a direct contraction in the presence of 0% and 0.1 % but not 4% albumin. This contraction was due to leukotriene D4 as MK-679 completely inhibited the contraction. In contrast, stimulated neutrophil supernatants increased responsiveness of sensitized human bronchial tissue to EFS. The increased responsiveness was observed only in the presence of 0.1% albumin, with the site of modulation likely to be prejunctional on the parasympathetic nerve. The increased responsiveness was not inhibited by any of the antagonists tested.Conclusion Sensitization status of the tissue and albumin concentration effect the responsiveness of human bronchial tissue to stimulated neutrophil supernatant. Our results suggest a possible role for neutrophils in hyperresponsiveness.

    Notes: FK506 (tacrolimus) is a natural product now approved in the US and Japan for organ transplantation. FK506, in complex with its 12 kDa cytosolic receptor (FKBP12), is a potent agonist of immunosuppression through the inhibition of the phosphatase activity of calcineurin. Rapamycin (sirolimus), which is itself an immunosuppressant by a different mechanism, completes with FK506 for binding to FKBP12 and thereby acts as an antagonist of calcineurin inhibition. We have solved the X-ray structure of unliganded FKBP12 and of FKBP12 in complex with FK506 and with rapamycin; these structures show localized differences in conformation and mobility in those regions of the protein that are known, by site-directed mutagenesis, to be involved in calcineurin inhibition. A comparison of 16 additional X-ray structures of FKBP12 in complex with FKBP12-binding ligands, where those structures were determined from different crystal forms with distinct packing arrangements, lends significance to the observed structural variability and suggests that it represents an intrinsic functional characteristic of the protein. Similar differences have been observed for FKBP12 before, but were considered artifacts of crystal-packing interactions. We suggest that immunosuppressive ligands express their differential effects in part by modulating the conformation of FKBP12, in agreement with mutagenesis experiments on the protein, and not simply through differences in the ligand structures themselves.

    Notes: Abstract This study was designed to compare the effectiveness of using two measurement devices: the WATSMART system and the WILDCAT theodolite system for robot metrology. The former device is less expensive, and runs automatically. The theodolite system has a better resolution but is more expensive, and runs manually. The objective was to determine the agreement in their measurement accuracy, variation, and the resulting best performance condition for the robot (i.e. optimal factor–level combination) between the two kinds of measurement equipment. The idea was that even if the accuracy and variation of the equipment differed, if the practical aspects (i.e. measurement performance and the purpose of measurement) were acceptable, the less expensive equipment would provide cost and automation benefits. The experiment was conducted on a PRO-ARM RS-2200, five axis, open loop, joint-coordinate type, educational robot. A Taguchi L27 orthogonal array was selected for the experimentation. Seven controllable factors (load, speed, distance moved, orientation of the robot arm, direction of travel, height of travel, and the starting point), along with three interactions, were studied. The accuracy of the robot and the best factor–level combination (FLC) were computed for each set of observations. The mean, standard deviation and maximum values for the two pieces of equipment were also computed. Based on this, the agreement between the two equipment was deduced. Although some similarity existed, the results were not good enough to convince the authors that the two measurement systems were equally effective. Recommendations are made for the appropriate situations for using each of the measurement devices.

    Notes: Abstract Near-infrared images of comet Hale-Bopp (C/1995 O1) were obtained from NICMOS/HST on UT August 27–28, 1997, when the comet emerged from the 50 degree solar elongation limit at 2.99 AU from Earth. Diffraction-limited images were obtained with camera 2 filters centered at 1.87, 1.90, 2.04 and 2.22 μm with ∼0.2″ resolution (0.076″/pixel; 165 km/pixel). Over the 1.7-hour baseline of observation, a recent (〈7 hours) outburst is seen in the form of an expanding spiral arm with a projected expansion velocity of ∼80 m/s. Other asymmetric features include a jet emanating from the nucleus and several static linear features. Comparisons of the flux distribution in the 2.04 and 2.22 μm filters indicate that the region near the nucleus exhibits a slight, ∼3%, water ice absorption.

    Notes: Summary Human herpesvirus 7 (HHV-7) is a close relative of human herpesvirus 6A (HHV-6A) and human herpesvirus 6B (HHV-6B) based on limited biologic and genetic data. In this work we describe physical and genetic maps for HHV-7 strain SB [HHV-7(SB)], which was obtained from the saliva of a healthy adult. The HHV-7(SB) genome length is approximately 144 kb by clamped homogeneous electric field gel electrophoresis and approximately 135 kb by summation of restriction endonuclease fragments. We constructed plasmid clones and PCR amplimers that span the HHV-7 genome, except for the genomic termini, and determined the maps of the restriction endonuclease cleavage sites forBamHI,PstI, andSacI. The HHV-7(SB) genome is composed of a single unique region of approximately 122 kb bounded at each end by a 6 kb direct repeat. Homologs to thirty-five herpesvirus genes were identified. The highest similarity was with the HHV-6 genes, with an average amino acid identity of 50%, followed by the human cytomegalovirus counterpart. The genomic and genetic maps indicated that the HHV-7 and HHV-6 genomes are colinear. There was no sequence variation in a segment of the gene encoding the DNA polymerase-associated factor homolog among six HHV-7 isolates, while the corresponding segment of the HHV-6A and HHV-6B counterparts differed by 4.6%. These data support previous observations that the closest genetic relatives of HHV-7 are betaherpesviruses.
