    Notes: Abstract  DNA methylation plays an important part in the regulation of gene expression. Alterations in DNA methylation in tumours have been reported and have been used to generate hypotheses about mutagenesis and silencing of tumour suppressor genes. However, the underlying mechanism is still poorly understood, and conflicting data on the levels of overexpression of 5′-cytosine DNA methyltransferase in sporadic colon carcinoma have been published. We used a competitive RT-PCR assay for quantification of mRNA of 5′-cytosine DNA methyltransferase in colon biopsies obtained from patients with hereditary colon carcinoma syndromes and compared the results with those obtained in a control group. No significant difference was found between the flat mucosa of FAP patients and the mucosa of the control group. In FAP and HNPCC patients, the 5′-cytosine DNA methyltransferase mRNA levels of adenomas were significantly higher (P〈0.05) than of flat mucosa in the same group, but both showed great variability from patient to patient. Our findings suggest that the mRNA levels of methyltransferase cannot be used as predictive marker for screening in families affected by hereditary colon carcinoma.

    Notes: Abstract  A new polymorphic form of glutathione S-transferase (GST), metabolising monohalogenated methanes, ethylene oxide and dichloromethane, has been purified from human erythrocytes and characterized. Several characteristics, such as similar elution patterns on different chromatographic matrices, KM-values and activity towards antibodies, confirm a previous assumption that this novel GST is a class θ enzyme. Although the presence or absence of the enzyme activity in human red blood cells is parallel with the polymorphism of the human GST T1 gene, the new GST θ in red blood cells may differ from the known GST T1-1 enzyme from other tissues in terms of substrate specificity, since established GST T1-1 substrates [1,2-epoxy-3-(p-nitro-phenoxy)propane and p-nitrobenzyl chloride] are not metabolized. The substrate specifity of the new enzyme in erythrocytes resembles more closely that of GST T2-2, most likely due to a common N-terminal modification which modifies substrate binding. The new polymorphic GST-isoform in human red blood cells therefore may be considered to represent an N-terminally modified isoform of GST T1-1.

    Notes: Abstract H-Tx rats produce congenitally hydrocephalic offspring with varying severity of the condition. We used moderately hydrocephalic rats without evident clinical signs of hydrocephalus and normal controls from the same stock when they were at least 1.5 years old. Macroscopic anatomy was studied by MRI and in fixed brain slices and the ultrastructure of the ependyma, with REM. Apart from markedly stretched areas, where the ependyma was totally destroyed and subependymal structures directly exposed to the CSF, the density of ependymal microvilli and of tufts of cilia was reduced in proportion to the ventricular distension of a given area. A supraependymal “network”– never seen before in acute hydrocephalus – was found, whose purpose is probably to prevent further ventricular enlargement. We conclude that even in arrested hydrocephalus the ependymal sequelae of hydrocephalus are similar to those of the acute stage, illustrating the extremely limited potenctial for recovery, but the organism seems nevertheless to react with an internal stabilization of the ventricular system.

    Notes: Abstract. The development of the central nervous α2-adrenergic system in the duck was studied by semiquantitative autoradiography at the ontogenetic stages embryonic days 20 (E20) and 27 (E27) and postnatal days 3 (P3) and 14 (P14) by using the monoradioiodinated α2-agonist clonidine ([125I]CLO) as radioligand. All structures endowed with α2-adrenoceptors in the adult animal were specifically labeled with [125I]CLO by E20. A detailed analysis of the binding capacity for [125I]CLO was performed for parts of several functional systems: hypothalamic structures (nucleus inferior hypothalami, nucleus magnocellularis preopticus, nucleus paraventricularis), limbic system (habenula, nucleus septalis lateralis, nucleus striae terminalis), circumventricular organs (organum pineale, organum subfornicale, plexus choroidei ventriculi tertii and ventriculi lateralis), visual system (hyperstriatum accessorium, nucleus reticularis superior, tectum opticum), and associative cortex (hyperstriatum ventrale). Except for the nucleus inferior hypothalami and the plexus choroideus ventriculi lateralis, all structures showed a perinatal (E27–P3) maximum of α2-adrenoceptor-binding capacity with a subsequent decline to values of prehatching stages. This uniform expression pattern of α2-adrenoceptors indicates that the days around hatching are a critical period for the development of the adrenergic system in the brain of the duck.

    Notes: The Helmholtz energy F is written as the sum F = FH + FA + FPol, where FH is the hard-body contribution, FA the attractive dispersion force contribution, and FPol either the quadrupolar or dipolar contribution. Here, a new expression for FA is constructed by a simultaneous correlation of experimental data of methane, oxygen, and ethane. The resulting equations for F with only three or four substance-specific parameters are tested for several nonpolar, quadrupolar, and dipolar fluids. In the correlation of large data sets good results are obtained. The significance of the approach, however, is in the good to excellent prediction of all thermodynamic properties in the whole fluid region based on a parameter fit to only four experimental data - two vapor pressures and two saturated liquid densities.

    Notes: Copper-catalyzed decomposition of alkyl diazoacetates in the presence of semicyclic enaminones 1a-o leads primarily to enamino esters 2 which constitute formal products of C-C insertion of the carbene unit. In the case of N-methyl (1a-e) and N-benzyl (1f-i) enaminones, 2a-i are accompanied by 2,3,5,6-tetrahydroindoles 3a-i, in which two carbene moieties are incorporated. At 250 °C, compounds 2b-e, which could not be isolated in pure form, undergo cyclocondensation to form 1,2,3,5-tetrahydrocyclopenta-[b]pyrroles 4b-e. In contrast, 2f-i can be isolated as Z,E mixtures and are transformed thermally into 4f-i only in the presence of silica gel. Carbene transfer to N-phenyl enaminones 1j-m and N-methyl enamino esters 1n,o leads only to the 1:1 adducts, enamino esters 2j-o, which do not undergo the cyclocondensation reaction under the previous conditions. Dehydrogenation of tetrahydroindoles 3c-e with tetrachloro-p-benzoquinone can be controlled to give either 1,2-dihydroindole-6,7-dicarboxylates 5 or indole-6,7-dicarboxylates 6.
