    Notes: Abstract: A combination of posttranslational modifications contributes to the high heterogeneity of brain tubulin in mammals. In this report, the structures of the detyrosinated carboxy-terminal peptides of α-tubulin from newborn and adult mouse brain were compared. The heterogeneity of these carboxy-terminal peptides was observed to increase from newborn to adult brain tubulin. The major part of this increased heterogeneity is due to the posttranslational excision of Glu450, which makes α-tubulin nontyrosinatable (Δ-2 tubulin). The structures of the polyglutamyl side chain of the bi- and triglutamylated peptides were analyzed in this work. In polyglutamylation of α-tubulin, the first glutamyl residue can only be amide-linked to the γ-carboxyl group of Glu445, but the additional residues may be linked either to the γ- or to the α-carboxyl groups of the preceding one. By optimized reverse-phase separations and comparison with synthetic peptides corresponding to all possible linkages for the biglutamylated (γ1α2, γ1γ2) and triglutamylated (γ1α2α3, γ1γ2γ3, γ1α2γ3, γ1γ2α3, γ1γ2α2) tubulin peptides, it was possible to conclude that the mode of linkage connecting the second and third additional glutamyl residues corresponds mostly to α-bond structures, for both newborn and adult mice.

    Notes: Abstract The purpose of this study was to examine the health patterns of cardiac surgical patients in the home health care population and their relationships to outcomes and duration of home health care using Gordon's Functional Health Pattern framework. Home health care records of 96 cardiac surgical clients were reviewed. Admission health pattern data, reasons for admission, duration and outcomes of home care services, characteristics of hospital experience, and demographic data were analyzed. Dysfunctional health patterns were primarily in the area of activity/exercise. The most common reasons for admission were monitoring of cardiopulmonary status, wound care, and instruction on diet, medications, and cardiac regimen. The mean duration of home care was 28.8 days. Thirty percent of the sample were readmitted to the hospital. Duration of home care was shorter for those who were married and for those who reported weakness, tiredness, or fatigue as a chief complaint. Readmission to the hospital was more likely for those who had complications during their initial hospital stay and those who required at least partial assistance with bathing, dressing, feeding, or toileting. Implications for practice and research are discussed.

    Notes: The major mechanism by which bacteria acquire free or haemoglobin-bound haem involves direct binding to specific outer membrane receptors. Serratia marcescens also secretes a haem-binding protein, HasA, which functions as a haemophore that catches haem and shuttles it to a cell surface specific outer membrane receptor, HasR. We report the isolation and characterization of hasAp, a gene from Pseudomonas aeruginosa. HasAp is an iron-regulated extracellular haem-binding protein that shares about 50% identity with HasA. HasAp is required for P. aeruginosa utilization of haemoglobin iron. It can replace HasA for HasR-dependent haemoblobin acquisition in a system reconstituted in Escherichia coli. HasAp, like HasA, lacks a signal peptide and is secreted by an ABC transporter. These findings show that haemophore-dependent haem acquisition is not unique to S. marcescens.

    Notes: Abstract Membrane electroporation, vesicle shape deformation and aggregation of small, NaCl-filled lipid vesicles (of radius a = 50 nm) in DC electric fields was characterized using conductometric and turbidimetrical data. At pulse durations tE≤ 55 ± 5 ms the increase in the conductivity of the vesicle suspension is due to the field-induced efflux of electrolyte through membrane electropores. Membrane electroporation and Maxwell stress on the vesicle membrane lead to vesicle elongation concomitant with small volume reduction (up to 0.6% in an electric field of E = 1 MV m–1). At tE 〉 55 ± 5 ms, further increases in the conductivity and the optical density suggest electroaggregation and electrofusion of vesicles. The conductivity changes after the electric pulse termination reflect salt ion efflux through slowly resealing electropores. The analysis of the volume reduction kinetics yields the bending rigidity κ = (4.1 ± 0.3) ⋅ 10–20 J of the vesicle membrane. If the flow of Na+ and Cl– ions from the vesicle interior is treated in terms of Hagen-Poiseuille's equation, the number of permeable electropores is N = 39 per vesicle with mean pore radius rp = 0.85 ± 0.05 nm at E = 1 MVm–1 and tE≤ 55 ± 5 ms. The turbidimetric and conductometric data suggest that small lipid vesicles (a ≤ 50 nm) are not associated with extensive membrane thermal undulations or superstructures. In particular with respect to membrane curvature, the vesicle results are suggestive for the design and optimization of electroporative delivery of drugs and genes to cell tissue at small field strengths (≤1 MVm–1) and large pulse durations (≤100 ms).

    Notes: Abstract Axonemal tubulin exhibits a high degree of heterogeneity mostly due to several posttranslational modifications (PTM). The aim of this work was to chemically characterize the different PTM occurring in the C-terminal tail of axonemal tubulin purified from sea urchin, Paracentrotus lividus, spermatozoa. After its purification, tubulin was enzymatically cleaved. The C-terminal peptides were chromatographically isolated, first by anion exchange and then by reverse-phase HPLC. Peptides were characterized by their sequence, determined by Edman degradation, and by their mass, determined by MALDI-TOF/MS. The two major conclusions are that the majority of the isolated C-terminal peptides were unmodified and that polyglycylation and polyglutamylation can occur simultaneously on one molecule of α-tubulin.

    Notes: The introduction of an aminoethyl side chain into cyclopentadienyl compounds of many d-block elements leads to interesting changes in structure and reactivity compared to the parent species: (i) A hemilabile bonding situation is observed in complexes with low-valent transition-metal centers; (ii) amino-group coordination to an adjacent metal or nonmetal center creates novel types of bimetallic compounds; (iii) complexes with a tethered ammoniumethyl group often show good solubility in protic solvents, relevant with regard to organometallic chemistry in water. These features together with others are described in this review, which informs about the synthesis and properties of this new type of transition-metal complexes.

    Notes: An analytical protocol suggested previously for the mass spectrometric sequence analysis of cyclosporins was extended by addition of methods for discrimination among isobaric amino acids differing either in the nature of the side-chain or N-substitution. The former goal was achieved by comparison of B/E linked-scan mass spectra of corresponding collisionally activated immonium ions with those of standard amino acids and the latter by comparison of B/E fragment ion spectra of [M + H]+ and [Md + 2H]+ ions. The use of the improved protocol is illustrated by the structure elucidation of a novel natural cyclosporin, [Leu9]CS.

    Notes: Mixed bent sandwich titanium (IV) complexes containing the donor-substituted [2-(diisopropylamino)ethyl]cyclopentadienyl (C5H4CH2CH2NiPr2 ≡ CpN) ligand are described. The highly moisture-sensitive metallocene dichloride (C5H4CH2CH2NNiPr2)(C5H4SiMe3)TiCl2 (1) was synthesized by reaction of CpNLi with CpSTiCl3 (CpS ≡ C5H4SiMe3). Complex 1 reacts with one equivalent of HCl with protonation of the amino group to give the air- and water-stable metallocene dichloride-hydrochloride [(C5H4CH2CH2-N(H)iPr2)(C5H4SiMe3)TiCl2]+Cl- (2). The structure of 2 was determined by a single-crystal X-ray diffraction study. Compounds 1 and 2 are precursors for ethylene polymerization catalysts. The reaction of 2 with one equivalent of Na+B[3,5-(CF3)2C6H3]4- afforded the borate [(C5H4CH2CH2N(H)iPr2)-(C5H4SiMe3)TiCl2]+B[3,5-(CF3)2C6H3]4- (3). The dimethyl compounds (C5H4CH2CH2NiPr2)(C5H4SiMe3)Time2 (4) was obtained by reaction of 1 with two equivalents of methyllithium. The diphenoxy complex (C5H4CH2CH2-NiPr2)(C5H4SiMe3)Ti(OPh)2 (5) was prepared analogously with a stoichiometric amount of lithium phenoxide. Compounds 4 and 5 show remarkable activity in the catalytic dehydrocoupling of phenylsilane.
