    Notes: Summary The localization of sympathetic fibers on the floor of the cranium was studied in rats using amine fluorescence histochemistry, neuropeptide-Y (NPY) immunohistochemistry, and electron microscopy. The vast majority of amine fluorescent fibers joined the abducent nerve and were localized in the peripheral zone under the perineurium. After advancing along this nerve for some distance, the fibers diverged into many bundles that converged to form the cavernous plexus at a rostral end of the trigeminal ganglion. On the dorsal surface of the trigeminal ganglion, one or two medium-calibered fluorescent bundles ran inside or in close proximity to the trochlear nerve, while many small-calibered, brightly fluorescent bundles also extended longitudinally in the epidural connective tissue. In rats that had undergone nerve severance, NPY-immunoreactive fibers were detected at the cut ends of the abducent and trochlear nerve. The differing amounts of NPY accumulated at the rostral and the caudal stumps indicated the direction of the NPY-bcaring fibers. Electron microscopy confirmed the presence of unmyelinated fibers in both the abducent and trochlear nerves.

    Notes: Summary To date, there have been few immunohistochemical investigations of atrial natriuretic polypeptide (ANP) in human cardiac tissue, especially the ventricles. In this study, myocardial tissue was obtained from two sources: the bilateral atria and ventricles at autopsy; and biopsy tissues from the right auricle and left ventricle of a patient with myocardial infarction undergoing surgery. These tissues were examined by the avidin-biotin immunoperoxidase technique using three kinds of primary ANP-antibodies. ANP-immunoreactivity was observed in the perinuclear region of myocytes of all tissues examined. The intensity of the reaction was stronger in atrial tissue, weaker in ventricular tissue. In the later tissue, the positive-staining myocytes were not part of the pulse-conducting system. Although the tissues we studied were not obtained from normal hearts, our data demonstrates that ANP-reactivity can be detected in ventricular myocytes outside the pulse-conducting system.

    Notes: Summary The retrograde fluorescence tracer, True Blue (TB), was injected into the forebrain septal area of neonatal rats. After 3 to 6 days the brains of these animals were carefully removed and placed in ice-cold sterilized physiological saline containing 1% glucose. Under the surgical microscope, one or two pairs of mesencephalic tissue samples, each containing a dorsal raphe nucleus, were punched out and transplanted into the third ventricle of a 5,6-DHT-pretreated adult rat. One month after transplantation, all animals were perfused and their brains sectioned using a cryostat. The sections were examined using a fluorescence microscope, and then processed for serotonin immunohistochemistry. The grafts were found to be successfully implanted and connected with the middle portion of the third ventricle. Four types of neurons, i.e., TB-labeled, serotonin-labeled, both TB-and serotonin-labeled, and non-labeled neurons, were detected in the grafts. This double-labeling method is considered to be a useful technique in characterizing the neurons in grafts which consist of a heterogeneous cell population.

    Notes: Summary Masked indoleamine cells (MICS) in the area postrema and adjacent areas in the rat were immunohistochemically studied (the peroxidase-antiperoxidase method) using a serotonin antiserum. After pretreatment with nialamide (200–300 mg/kg), immunoreactive MICS could be observed. They were small cells (about 12 μm in diameter) with several processes and were distributed in nearly all parts of the area postrema and also in the nucleus tructus solitarii. Following a single intraventricular injection of 75 μg 5,6-dihydroxytryptamine, the immunoreactivity of these cells conspicuously decreased for several days. The submicroscopical structure of the cells was investigated using immunoelectron microscopy. Immunoreactive products were observed in the cytoplasm as particles with a diameter of 25–40 nm and high electron density, but these were not found in the nucleus or cell organelles.

    Notes: Summary Using the immunoperoxidase technique in conjunction with specific antisera to α-atrial natriuretic polypeptide (α-ANP), it was shown that immunoreactive cell bodies and varicose fibers are widely distributed throughout the rat brain. The highest concentrations of α-ANP-containing neuronal cell bodies and fibers were found in the hypothalamus and septum. This result confirms the radioimmunological determination of α-ANP immunoreactivity in the rat brain.

    Notes: Summary A newly modified and improved immunohistochemical method was devised for the demonstration of the serotonin neuron system in the central nervous system of the rabbit using serotonin antibody obtained by the immunization of rabbits, i.e., the biotinylation of serotonin antibodies, and the application of the two-step avidin-biotin-peroxidase complex (ABC) method. Using this technique, high background staining and nonspecific reactions were avoided, and extremely clear preparations were produced. The serotonin neurons of rabbit brain, which have a Golgi-like appearance, were followed to the fine terminals. This technique were also applied for electron microscopy, and satisfactory results concerning the submicroscopical distribution of serotonin were obtained.

    Notes: Summary The sequence of the 3′-terminal 1223 nucleotides (nts) of a Japanese isolate of turnip mosaic virus (TuMV-Jap) RNA has been determined. The sequence reveals a single open reading frame (ORF) which terminates at a position 212 nts upstream of the 3′ poly(A)-tract. Determination of the N-terminal amino acids of TuMV-Jap coat protein (CP) mapped the CP cistron within this ORF and revealed a Glu-Ala dipeptide sequence as the putative cleavage site by which the CP is released from the viral polyprotein. The predicted amino acid sequence of the TuMV-Jap CP shows 97.2% identity with that of a Canadian isolate of TuMV (TuMV-Can) and 99% with a second, Chinese, isolate (TuMV-Chi). However, the 3′-terminal non-translated region (NTR) of TuMV-Jap RNA is significantly shorter (212 nts) than the 3′-NTR of TuMV-Can RNA (668 nts), but of equal length as the 3′-NTR of the TuMV-Chi isolate which also measures 212 nts. The 3′-NTRs of both the TuMV-Jap and TuMV-Chi RNAs show homology with the first 201 nucleotides of the TuMV-Can RNA 3′-NTR. A search in the EMBL nucleotide sequence database revealed that the 467 nt-long unique extension of the 3′-NTR of TuMV-Can RNA has 89.8% homology to a part of the chloroplast ribosomal protein 12 gene (rsp 12-gene). Irrespective of the origin of this extra sequence in the reported TuMV-Can sequence, which may have been introduced by a genuine RNA recombination event, it is concluded that the standard TuMV genome has a CP gene of 864 nts and an conserved 3′-NTR of approximately 212 nucleotides in length.

    Notes: Abstract The genetic model for hybrid sterility that an allelic interaction at the S 5 locus induces the abortion of megaspores has been proposed as partial clarification of the wide compatibility in Asian rice cultivars; this model predicts the presence of an egg-killer. The present study was carried out in order to confirm that the proposed S 5 i allele acts as an egg-killer against its counterpart, the S 5 j allele, in the Indica-Japonica hybrid. A conspicuous feature of an egg-killer is the high rate of its transmission into the progeny through the egg. Backcrossing experiments were conducted using the Indica-Japonica hybrid in which the S 5 i and S 5 j alleles were assumed to be involved. Although an egg-killer was easily identified by these backcross experiments, it was not detected in the Indica-Japonica hybrid, which suggests that the proposed genetic mechanism for hybrid sterility in Asian rice should be viewed with caution.

    Notes: Zusammenfassung Histologische Untersuchungen an der Hypophyse von Megalobatrachus japonicus ergaben: 1. In der Sella turcica liegt dorsal von der Hypophyse ein Gewebskomplex mit knochenmarkähnlichen Zellen. Es lassen sich sowohl Vorstufen und viele Formen von verschiedenen Granulozyten und Lymphozyten ausmachen, ferner Zellen, die sich mitotisch teilen. Die Diapedese von reifen Blutzellen durch die Wandung sinusartiger Kapillaren ist zu beobachten. 2. Das Stroma dieses Gewebes wird von argyrophilem Bindegewebe mit Retieulumzellen gebildet, das sich zwischen den Blutgefäßen ausspannt. Außerdem finden sich elastische Fasern und Bündel markloser Nervenfasern. 3. Dieses Gewebe wird mit dem myeloiden Meninxorgan über dem IV. Ventrikel bei Ganoiden vergleichend betrachtet, von dem es sich dadurch unterscheidet, daß es weder erythropoetische Funktion besitzt, noch in Verbindung zum Schädelskelet steht.

    Notes: Zusammenfassung Das Kaudalrückenmark von Thunfischen (Thunnus, Neothunnus, Parathunnus), Igelfisch (Canthigaster rivulatus) und Zenopsis nebulosa wurde makroskopisch und histologisch im Hinblick auf die Ausbildung einer Neurophysis spinalis caudalis untersucht. Bei allen untersuchten Spezies wurden außer der Neurophysis spinalis caudalis im Bereich des letzten Wirbels noch neurophysäre Anschwellungen im kaudalen Rückenmarksanteil beobachtet. Die Neurophysen vom Thunfisch treten in einigen kaudalen Rückenmarkssegmenten regelmäßig auf. Die Neurophyse der Teleostier ist nicht immer ein einziges Organ. Bei einigen Fischarten ist sie multipel ausgebildet.
