    Notes: The destruction of Trichinella spiralis by low temperature treatment has been demonstrated to be an effective tool in combatting this parasite in fresh pork products. Current procedures, however, require a holding period under varying times and temperatures to accomplish this destruction. Freezing with cryogenic materials offers the opportunity to attain ultra low temperatures and, thus, eliminate post–freezing holding periods. Four trials were conducted using trichina-infected fresh pork patties approximately 9 mm thick and 88 mm square. In trials 1, 2 and 3 the patties were frozen with LN, using a modified Heath freezing tunnel to final equilibrated temperatures of –12C, –14°C, –20°C, –23°C, –25°C, –28°C, –29°C, –39°C and −47°C. In trial 4 the patties were frozen in a Certified Multideck tunnel using liquid CO2 as a refrigerant to −10°C, –17°C, –23°C, –29°C and –39°C. Patties were thawed immediately and checked for viable T. spiralis. No positive samples were found at temperatures of –29°C or below.

    Notes: SUMMARY– An official USDA dry-cure ham processing procedure was evaluated for efficacy in devitalizing trichinae. The procedure consisted basically of a 40-day salt cure followed by 10 days of drying at 95° F or above. Within the cure period, marked loss of viability was noted only for trichinae in muscle with brine concentrations approaching 8%. The drying temperature was the critical factor in devitalizing trichinae. 28 days' cure followed by 6 days of drying at 98° F destroyed viability of all parasites regardless of brine concentration. Other effective drying time-temperature combinations included: 125° F-4 days; 70° F-35 days and 40°F-190 days.

    Notes: Zusammenfassung Nach den vorliegenden Veränderungen der Lungenvolumina ergibt sich für die Lobektomierten und Bilobektomierten mit Lungencarcinom p.op. ein anderes Verhalten als für die Patienten, die wegen benigner Lungentumoren operiert wurden. In beiden Gruppen kommt es zu einer kompensatorischen Überblähung des Restparenchyms mit unterschiedlicher Dehnbarkeit beider Lungenseiten nach dem 1. Jahr. In der Gruppe der Carcinompatienten erfolgt die Überblähung fast ausschließlich zugunsten des Residualvolumens und schränkt die funktionelle Anpassung erheblich ein. Die unterschiedliche Dehnbarkeit führt zu einer Verteilungsstörung insbesondere bei Lobektomierten und Bilobektomierten wegen Bronchialcarcinom in Abhängigkeit von schon vorbestehenden Lungenveränderungen, insbesondere einem mehr oder weniger ausgeprägten Lungenemphysem. Daraus resultiert p.op. eine Hypoxie bereits in Ruhe, die sich bei mittelschwerer körperlicher Belastung bis zur akuten Globalinsuffizienz steigert. Bei den Patienten mit Resektionen wegen benigner Tumoren oder Bronchiektasen erfolgt die Kompensation eher im Sinne eines Emphysema pulmonum auctum und bildet sich nach einem Jahr wieder zurück. Bei den pneumonektomierten Patienten mit Lungencarcinom kommt es vorübergehend zu dem Einfluß der sog, Distension, die sich bis zum 3. Jahr sicher, nach dem 5. Jahr jedoch nicht mehr eindeutig nachweisen läßt. Bei den Bilobektomierten und Pneumonektomierten ist eine unter Belastung sich intensivierende Hypoxie auf eine Diffusionsstörung wegen reduzierter Capillaroberfläche und verkürzter Kontaktzeit bei Steigerung des Herzzeitvolumens zurückzuführen.

    Description / Table of Contents: Zusammenfassung Bei 86 Patienten, die zu mehr als 80% auf allgemeinen internen Stationen untergebracht waren, wurden Wiederbelebungsmaßnahmen durchgeführt. Sie waren bei 22 Patienten kurzfristig, bei 23 Patienten langfristig erfolgreich. 21 dieser 23 Patienten konnten die Klinik verlassen. Bei 63 Fällen handelte es sich um ein primäres Kammerflimmern. Bei 55 von ihnen und bei 15 von 22 Patienten mit primärer Asystolie ließen sich die elektrischen Erregungsvorgänge des Herzens wiederherstellen. Bei 35 Patienten mit primärem Kammerflimmern folgte der Defibrillation eine längeranhaltende Asystolie, die in 8 Fällen unbeeinflußbar blieb. Auch bei älteren Menschen sind Wiederbelebungsversuche bei gegebener Indikation erfolgversprechend, soweit es sich nicht um eine primäre Asystolie handelt. Bei der Mehrzahl der Patienten lag als Grundleiden eine coronarsklerotische Kardiopathie vor. Bei ihr sind die Aussichten auf eine erfolgreiche Wiederbelebung günstiger als bei anderen Herzerkrankungen und bei Lungenembolien. Bei langfristig Wiederbelebten wurden häufiger als bei erfolglosen oder kurzfristig erfolgreichen Rcanimationen normale Harnstoffwerte und eine Behandlung mit Antiarrhythmica oder Nebennierenrindensteroiden vor Eintritt des Herzstillstandes sowie eine Bchandlung mit Antiarrhythmica und Herzglykosiden nach erfolgreicher Wiederbelebung nachgewiesen.
    Notes: Summary Resuscitation methods were applied in 86 patients hospitalized in a medical in-patient service. A short-lasting therapeutic success was observed in 22 patients while 23 patients showed long-term survival. Two of these died in hospital.—Primary ventricular fibrillation was diagnosed in 63 cases. The normal electric excitability of the heart was restored in 55 cases of ventricular fibrillation and in 15 of 22 patients with primary asystolia. In 35 patients a long-lasting asystole was seen after defibrillation which remained uninfluenced in 8 cases.—Attempted resuscitation seems to be successful even in older patients provided they do not show primary asystolia.—Coronary artery disease was the main cause of the cardiac lesions. The chances for successful resuscitation are better in patients with coronary artery disease than in those with other cardiac pathology or in the presence of pulmonary embolism. In patients with a long-lasting effect aftes resuscitation, the following characteristics were more often noted than in those demonstrating short or unsuccessful effce's: a) normal blood urea levels. b) a treatment with antiarrhythmic agents or corticoids prior to cardiac standstill. c) a treatment with antiarrhythmic and digitalis after resuscitation.

    Notes: Summary In severe cerebral trauma oxygen pressure, acid-base balance, acid metabolites and electrolytes in C.S.F. are a sensitive gauge of celebral circulation. The difference between pO2 levels in blood and C.S.F. can inform about the extent of damage of the brain. The pH of C.S.F. and the difference between arterial blood and C.S.F. pCO2 levels provide information about changes in cerebral blood circulation. They seem to be related to pH in cerebral edema. A most significant sign of reuced and insufficient cerebral circulation is an increase of lactate and a decrease of bicarbonate in C.S.F. Increase of lactate to more than 45 mg% is not compatible with recovery of cerebral function.

    Notes: Summary The yeast Saccharomyces cerevisiae has been engineered to synthesize and secrete desulfato-hirudin (hirudin), a thrombin inhibitor from the leech Hirudo medicinalis. The synthetic gene coding for hirudin was expressed constitutively under the control of four size-variants of the yeast glyceraldehyde-3-phosphate dehydrogenase promoter (GAP) and cloned into a 2 μ based multicopy yeast vector. The constitutive action of the four promoter variants was confirmed by demonstrating that the expression and secretion of hirudin is growth-related. The different efficiencies of the promoter variants not only affected hirudin expression but also led to changes in several cellular parameters, such as cell growth, average plasmid copy number and plasmid stability. The observed changes show that yeast cells establish a specific equilibrium for each promoter variant. We conclude, that the adjustment of cellular parameters in response to the expression levels of a heterologous protein is regulated by two counteracting selective forces: (1) the need for complementation of the auxotrophic host marker by the plasmid-encoded selection gene which, in the case of dLEU2, requires several plasmid copies; and (2) a selective advantage of cells with a lower copy number enabling them to escape the burden of heterologous protein production.
