Description / Table of Contents: Summary Three cases of acute myocardial infarction with a.v.-block and shock improved immediately and survived after insertion of an artificial pacemaker via venous catheterisation. Restoration of normal rhythm took place after several days. Pathophysiology and indication of this therapy are discussed.

Notes: Summary Tunicamycin inhibited the production of infectious Borna disease virus (BDV) and glycosidase treatment eliminated the infectivity of cell-free virus. A glycoprotein of approximately 17 kDa, found in association with infectious virus, was identified by Concanavalin A binding.

Notes: [Auszug] Fading of compound I occurs rapidly at 25 C in the dark in the presence of simple primary, secondary or tertiary aliphatic amines in nominally anhydrous acetone, ethanol, acetonitrile or dioxan despite free access of air. Notes: Summary The yeast Saccharomyces cerevisiae has been engineered to synthesize and secrete desulfato-hirudin (hirudin), a thrombin inhibitor from the leech Hirudo medicinalis. The synthetic gene coding for hirudin was expressed constitutively under the control of four size-variants of the yeast glyceraldehyde-3-phosphate dehydrogenase promoter (GAP) and cloned into a 2 μ based multicopy yeast vector. The constitutive action of the four promoter variants was confirmed by demonstrating that the expression and secretion of hirudin is growth-related. The different efficiencies of the promoter variants not only affected hirudin expression but also led to changes in several cellular parameters, such as cell growth, average plasmid copy number and plasmid stability. The observed changes show that yeast cells establish a specific equilibrium for each promoter variant. We conclude, that the adjustment of cellular parameters in response to the expression levels of a heterologous protein is regulated by two counteracting selective forces: (1) the need for complementation of the auxotrophic host marker by the plasmid-encoded selection gene which, in the case of dLEU2, requires several plasmid copies; and (2) a selective advantage of cells with a lower copy number enabling them to escape the burden of heterologous protein production.

Description / Table of Contents: Summary The sideromycin antibiotic A 22,765 is taken up from the medium by Staphylococcus aureus SG 511 against a concentration gradient. In contrast to this, A 22,765-resistant cells display an extremely low degree of permeability to the molecule. Ferrioxamine B penetrates the cells in only very minute quantities, but nevertheless antagonises permeation by A 22,765. This permeation-antagonising effect of ferrioxamine B is discussed.

Description / Table of Contents: Summary In a cell-free system obtained from Staphylococcus aureus SG 511, the sideromycin antibiotics danomycin and A 22,765 inhibit the poly U-directed incorporation of phenylalanine; albomycin has no influence on this incorporation, whereas ferrimycin A1 actually enhances it. In the case of the antibiotic A 22,765 it has been demonstrated that S-30 extracts from A 22,765-sensitive cells of S. aureus behave in the same way as those from A 22,765-resistant cells; in no instance, however, was ferrioxamine B able to counteract the inhibitory effect of A 22,765. The sideromycins, which are regarded as a homogeneous group on the basis of their chemical and biological characteristics, possibly have differing mechanisms of action. The possibility that sideromycin permeation may be susceptible to antagonism by sideramines is discussed.
