    Notes: We have isolated a hybrid gene, composed of the first 455 nucleotides of hisP and nucieotides 275–1107 of malK, the genes coding for the nucleotide-binding components of the high-affinity transport systems for histidine and maltose in Salmonella typhimurium, respectively. The fusion had occurred by recombination within 11 homologous base pairs located between the two DNA fragments. In the chimeric protein peptidic motifs A and B, proposed to be part of the nucleotide-binding fold, originate from HisP and MalK, respectively. Plasmid pES42-39, harbouring the hybrid gene, was shown to complement only a malK mutation but failed to complement a hisP deletion mutation. The chimeric protein was identified by immunoblotting as a protein with an apparent molecular mass of 49kDa. Removal of the C-terminal 77 amino acid residues from the chimeric protein resulted in the loss of function in transport. In contrast, 51 amino acid residues could be removed from the C-terminus of wild-type MalK without any effect. Upon overproduction the chimeric protein, as wild-type MalK, inhibited expression of the malB regulon. However, both truncated proteins, when overproduced, did not exhibit this activity. Based on these results, a tentative model of the functional domains of MalK is presented.

    Notes: In the framework of the European project aimed at the sequencing of the Bacillus subtilis genome the DNA region located between gerB (314°) and sacXV (333°) was assigned to the Institut Pasteur. In this paper we describe the cloning and sequencing of a segment of 97 kb of contiguous DNA. Ninety-two open reading frames were predicted to encode putative proteins among which only forty-two were found to display significant similarities to known proteins present in databanks, e.g. amino acid permeases, proteins involved in cell wall or antibiotic biosynthesis, various regulatory proteins, proteins of several dehydrogenase families and enzymes II of the phosphotransferase system involved in sugar transport. Additional experiments led to the identification of the products of new B. subtilis genes, e.g. galactokinase and an operon involved in thiamine biosynthesis.

    Notes: Abstract One of the primary obstacles to be overcome for the development of economical fuel cells is the high cost of the membrane electrolyte. The currently favoured polymer electrolytes consist of poly(tetrafluoroethylene) backbone structures and poly(perfluorosulphonic acid) side chains. In an effort to find lower cost membranes, some radiation-grafted copolymer membranes were investigated. All the membranes contained poly(styrenesulphonic acid) side chains. Three different backbone polymer structures were studied: low-density poly(styrene), poly(tetrafluoroethylene)/poly(perfluoropropylene), and poly(tetrafluoroethylene). The results indicate that the membrane consisting of a poly(tetrafluoroethylene)/poly(styrenesulphonic acid) copolymer is a promising candidate as a fuel-cell electrolyte.

    Notes: Summary The influence of transforming growth factor-β (TGF-β) on hematopoiesis has been evaluated by adding blocking antibodies against TGF-β to colony forming assays (CFU-c). When optimum concentrations of recombinant growth factors, granulocyte-macrophage colony stimulating factor (GM-CSF), and interleukin-3 (IL-3) were added to stem cells from the peripheral blood of healthy individuals and certain patients with tumors or HIV infection, the anti-TGF-β capable of blocking 5 ng/ml of active TGF-β had no significant influence on erythroid or myeloid colony formation. However, in certain immunosuppressed individuals, anti-TGF-β resulted in a significant decrease of erythroid colony formation and slight suppression of myeloid colony formation. The significant inhibition of hematopoiesis by plasma of HIV patients could be due to the presence of active forms of TGF-β. The results of the blocking experiments are consistent with the concept that TGF-β in low concentrations is essential for erythropoiesis and myelopoiesis but that higher levels of TGF-β primarily inhibit erythropoiesis in vitro. TGF-β serves as a coordinating factor when efficient recruitment of granulocytes and monocytes is more essential than erythropoiesis and stem cell growth.

The majority of antibodies identify an 80-kDa molecule, which is not detected on healthy tissues except on certain cultured monocytes. One antibody recognize the CD2 ligand MIC2 and 2 antibodies (numbers 13 and 16) define a higher-molecular-mass antigen. Antibody 16 is also expressed on mesenchymal fibroblasts of bone marrow or fetal origin. Tumorspecific antigen expression is potentially linked to the chromosome 22 abnormality decribed in Ewing's sarcoma.

    Notes: Zusammenfassung 1. Es wurde die Umlagerungsfähigkeit der Carotine als der Vorstufen zum Vitamin A unter verschiedenen Bedingungen der Leberschädigung verfolgt. 2. Im Stadium der vollen avitaminotischen Störung kann aus dem Provitamin das Vitamin A nicht mehr gebildet werden, Vitamin A selbst dagegen wird gut verwertet und gespeichert. 3. Die thyreogene Leberschädigung geht mit Vitamin A-Verlust einher. Dieser Verlust beruht auf einem stark erhöhten Vitamin A-Umsatz und -Verbrauch, dagegen nicht auf einer mangelnden Umlagerungsfähigkeit. 4. Im Zustand der thyreogenen Leberschädigung ist die Verwertungs- und Speicherungsfähigkeit des Vitamin A achtmal höher als die seiner Vorstufen, der Carotine. 5. Der Vitamin A-Umsatz ist an den Glykogenumsatz der Leber gekoppelt.
