    Notes: Abstract The effect of recombinant human erythropoietin (rhEPO) and interleukin 3 (IL3) on circulating haematopoietic progenitors consisting mainly of immature burst-forming-units-erythrocytes (BFU-E), was investigated in ten paediatric patients treated by regular haemodialysis. During a 30-week study rhEPO treatment resulted in a rise of median haemoglobin levels from 6.7 g/dl to 〉10 g/dl in all patients. Before initiating rhEPO treatment the number of circulating BFU-E in chronic renal failure patients responded to grading doses of rhEPO in vitro similar to that in control children; however, the dose-response curves were not predictive for the in vivo response to rhEPO. After an initial rise in five patients BFU-E numbers declined by week 30 of rhEPO treatment. BFU-E numbers decreased to 35% of pretreatment values. The number of granulocyte-macrophage colony forming cells (GM-CFC) also decreased during rhEPO treatment. Addition of IL3 to the culture medium containing saturating concentrations of granulocyte-macrophage colony stimulating factor did not stimulate BFU-E numbers of patients before rhEPO treatment or those of controls. However, 2 weeks after start of rhEPO treatment IL3 increased the growth of patient's BFU-E in vitro to 220% of pretreatment levels, followed by a gradual decrease of stimulation until the end of observation. These findings indicate that: (1) long-term recruitment of circulating haematopoietic progenitors during rhEPO treatment is low in children with renal anaemia; (2) rhEPO sensitivity of circulating BFU-E is not predictive for the in vivo response; (3) rhEPO treatment results in enhanced sensitivity of BFU-E to IL3.

    Notes: Abstract The effect of structural modifications on the arotinoid molecule, a new class of retinoids, on their teratogenicity in mice was studied. Animals were treated on days 8 and 9 of gestation, the most susceptible stages to retinoid-induced malformations in rodents. The teratogenic potency of the 13 arotinoids tested varied over a dose range of more than five orders of magnitude. Next, we tested whether the quantitative differences in the teratogenicity of these arotinoids correlates with their activity in high density (micromass) cultures of rat embryonic limb bud and midbrain cells. There was a good quantitative correlation between the in vivo teratogenicity and the in vitro activity in limb bud cells but no correlation was found in midbrain cells. Thus, the limb bud cell culture system may be useful for a preliminary testing to select non-teratogenic retinoids. For the risk assessment in humans, however, the in vitro results should be verified in animals studies.

    Notes: Soluble (Tetraethylphthalocyaninato)iron(II) and -cobalt(II) CompoundsThe (tetraethylphthalocyaninato)iron and -cobalt compounds Et4PcFe (9), Et4PcCo (8), and Et4PcH2 (7) have been synthesized starting from 3,4-dibromoacetophenone (1) via the corresponding dinitrile 5 or isoindolenine 6, respectively. Et4PcFe (9) reacts with tBuNC, p-diisocyanobenzene (dib), and me4dib to form the bisaxially coordinated compound Et4PcFe(tBuNc)2 (10) and the oligomers [Et4PcFe(dib)]n (11) and [Et4PcFe- (me4dib)]n (12). All of the synthesized compounds, except the oligomers with a chain length of more than 8 - 14 units, are soluble in organic solvents such as chloroform or toluene. The new compounds were characterized by 1H-, 13C-NMR, 13C-CP/NMR, UV/Vis, and IR spectra. In addition the redox properties of Et4PcCo (8) were studied by cyclic voltammetry and spectroelectrochemical methods. The Mößbauer spectra of the (phthalocyaninato)iron compounds 9, 11, and 12 demonstrate the bridged structure of [Et4PcFe(L)]n (L = dib, me4dib) (11, 12). The powder conductivity of [Et4PcFe(dib)]n (11) (σRT = 5·10-6 S/cm) is ca. 106 times higher than that of Et4PcFe (9) (σRT = 9·10-12 S/cm). After doping with iodine the oligomers 11 and 12 show an increase in conductivity of 105 and 104, respectively (σRT ≍ 10-2 S/cm for [Et4PcFe(dib)I0.85]n).

    Notes: Bisaxially Coordinated (Tetraphenylporphinato)iron(II) ComplexesThe coordination behavior of (tetraphenylporphinato)iron(II) (TPPFe) has been studied with the preparation of the monomeric complexes TPPFe(L)2 with L=py (pyridine), pyz (pyrazine), 2,6-dimepyz (2,6-dimethylpyrazine), bpy (4,4′-bipyridine), bpyac [1,2-bis(4-pyridyl)acetylene], dabco (1,4-diazabicyclo [2.2.2]octane), and BzINC (benzyl isocyanide). Pure TPPFe has been obtained by thermal decomposition of TPPFe(py)2. The monomeric complexes TPPFe(L)2 with L=pyz, bpy, bpyac, and dabco can be transformed into the bridged systems [TPPFe(L)]n by heating them in toluene or chlorobenzene. The bridged structure of the complexes [TPPFe(L)]n has been confirmed by IR and TG/DTA investigations and especially by Mößbauer spectroscopy. In comparison with the corresponding (phthalocyaninato)iron compounds the bridged TPPFe complexes exhibit lower semi conducting properties even after doping with iodine.
