    Notes: Abstract Several attempts to produce conducting polymer based all-solid-state reference electrodes are presented. Open circuit potential of conducting polymers is redox sensitive and Donan equilibrium dependent. Therefore, more sophisticated constructions are necessary. Most promising were bilayers composed of conducting polymers with different ion-exchanger properties.

    Description / Table of Contents: Abstract Vulvodynia is a complex syndrome of chronic vulvar pain. It is divided into several subtypes: 1. cyclic vulvovaginitis (pain occurs after coitus), 2. vulvar vestibulitis syndrome (pain mainly with intercourse), 3. dysaesthetic vulvodynia (psychosomatic; diagnosis of exclusion), 4. vulvar dermatoses (e.g. pemphigus vulgaris, contact dermatitis). Additional causes have been described in single cases. Though vulvodynia is often accompanied by psychological distress, somatic causes have to be considered in each case.
    Notes: Zusammenfassung Der Begriff Vulvodynie beschreibt chronische Schmerzzustände des äußeren weiblichen Genitales. Klinisch werden verschiedene Subtypen unterschieden: 1. Zyklische Vulvovaginitis mit postkoitalem Beschwerdemaximum und fraglicher Assoziation mit Candida albicans. 2. Vulvavestibulitis mit umschriebener Schmerzhaftigkeit des Introitus durch Entzündung der dortigen Drüsenausführungsgänge mit koitalem Beschwerdemaximum. 3. Essentielle Vulvodynie als funktionelles Beschwerdebild ohne erkennbare somatische Ursache. 4. Dermatosen der Vulva unterschiedlicher Genese, wobei Autoimmunerkrankungen (z.B. Pemphigus vulgaris) und Kontaktdermatitiden von besonderer Bedeutung sind. Zusätzlich kommen verschiedene meist kasuistisch dargestellte seltene Ursachen in Betracht. Somatoforme Störungen oder psychische Alterationen können die Differentialdiagnose einer somatischen Ursache der Vulvodynie (z.B. Kontaktdermatitis) verschleiern. Ein interdisziplinäres Vorgehen ist in jedem Fall sinnvoll.

    Notes: Abstract The effect of structural modifications on the arotinoid molecule, a new class of retinoids, on their teratogenicity in mice was studied. Animals were treated on days 8 and 9 of gestation, the most susceptible stages to retinoid-induced malformations in rodents. The teratogenic potency of the 13 arotinoids tested varied over a dose range of more than five orders of magnitude. Next, we tested whether the quantitative differences in the teratogenicity of these arotinoids correlates with their activity in high density (micromass) cultures of rat embryonic limb bud and midbrain cells. There was a good quantitative correlation between the in vivo teratogenicity and the in vitro activity in limb bud cells but no correlation was found in midbrain cells. Thus, the limb bud cell culture system may be useful for a preliminary testing to select non-teratogenic retinoids. For the risk assessment in humans, however, the in vitro results should be verified in animals studies.

    Notes: Abstract Fusarium sporotrichioides and Gibberella saubinetti O-demethylate 3,9-dimethoxypterocarpan to 3-methoxy-9-hydroxypterocarpan and then 3,9-dihydroxypterocarpan. F. anguioides Sherb., F. avenaceum and F. graminearum convert the same substrate to a mixture of 3-methoxy-9-hydroxypterocarpan and 3-hydroxy-9-methoxypterocarpan. Induction of pterocarpan degradation by pisatin in F. avenaceum leads to preferential conversion of 3,9-dimethoxypterocarpan to 3-hydroxy-9-methoxypterocarpan (medicarpin) and the isoflavan vestitol. The data are discussed with respect to phytoalexin degradation by phytopathogenic Fusarium fungi.

    Notes: Abstract The effect of recombinant human erythropoietin (rhEPO) and interleukin 3 (IL3) on circulating haematopoietic progenitors consisting mainly of immature burst-forming-units-erythrocytes (BFU-E), was investigated in ten paediatric patients treated by regular haemodialysis. During a 30-week study rhEPO treatment resulted in a rise of median haemoglobin levels from 6.7 g/dl to 〉10 g/dl in all patients. Before initiating rhEPO treatment the number of circulating BFU-E in chronic renal failure patients responded to grading doses of rhEPO in vitro similar to that in control children; however, the dose-response curves were not predictive for the in vivo response to rhEPO. After an initial rise in five patients BFU-E numbers declined by week 30 of rhEPO treatment. BFU-E numbers decreased to 35% of pretreatment values. The number of granulocyte-macrophage colony forming cells (GM-CFC) also decreased during rhEPO treatment. Addition of IL3 to the culture medium containing saturating concentrations of granulocyte-macrophage colony stimulating factor did not stimulate BFU-E numbers of patients before rhEPO treatment or those of controls. However, 2 weeks after start of rhEPO treatment IL3 increased the growth of patient's BFU-E in vitro to 220% of pretreatment levels, followed by a gradual decrease of stimulation until the end of observation. These findings indicate that: (1) long-term recruitment of circulating haematopoietic progenitors during rhEPO treatment is low in children with renal anaemia; (2) rhEPO sensitivity of circulating BFU-E is not predictive for the in vivo response; (3) rhEPO treatment results in enhanced sensitivity of BFU-E to IL3.

    Notes: Abstract Two genomic fragments capable of driving the expression of the hygromycin B resistance gene (hph) were isolated from the phytopathogenic ascomycete Gibberella pulicaris (anamorph Fusarium sambucinum) using a “promoter-probe library” strategy. Two libraries consisting of random, 0.5–2.0-kb fragments of genomic DNA inserted 5′ of a promoterless hph gene were constructed and used for transformation of G. pulicaris. Both libraries transformed G. pulicaris at a low frequency. Transformants tolerated up to 800 μg/ml of hygromycin B, while untransformed colonies were inhibited completely by 50 μg/ml of the antibiotic. Plasmids were re-isolated from transformants by simply digesting, the genomic DNA with KpnI, which cuts once in the polylinker 5′ to the insert, and transforming E. coli with the re-ligated DNA. The recovered plasmids transformed G. pulicaris with a frequency of up to 4.4 transformants/μg of DNA. Both promoter fragments were sequenced and found to contain TATA and CAAT boxes as well as CT-rich sequences. This method makes it possible to easily isolate many fragments with promoter activity from filamentous fungi, and should facilitate the investigation of the promoter structures necessary for the expression of fungal genes.

    Notes: Abstract ATP citrate lyase (ACL) catalyzes the formation of cytosolic acetyl-CoA, which is mainly used for the biosynthesis of fatty acids and sterols. In this paper, we show for the first time that in filamentous fungi two different subunits of ACL are encoded by two separate genes. This is in contrast to animals where ACL is encoded by a single gene. Data are presented on acl genes from the filamentous fungi Sordaria macrospora and Gibberella pulicaris. In S. macrospora, both genes, acl1 and acl2, are clustered within a region of 10 kb and are divergently transcribed.

    Notes: Seven eggs from four clutches of grey plovers (Pluvialis squatarola) breeding in the Lena Delta, Sakha Republic, Yakutia, in 1997 were analysed for persistent organochlorines and mercury. Contamination levels were low and within the same range as found in eggs from waders (Charadriiformes) breeding in north-west Europe. One exception was ?HCH, averaging 99.2 ng g−1 fresh egg mass, a level about ten times higher than in wader eggs from north-west Europe. β-HCH accounted for 98% of the total HCH concentration. There are indications that the pesticide levels may reflect former local use. ∑PCB level (57.0 ng g−1, 62 congeners) and mercury concentration (68.7 ng g−1) can be considered low.
