    Notizen: Abstract: A cell-free enzyme system from cultures of Fusarium culmorum catalyses the 12,13-epoxidation of semi-synthetic 9β, 10β-epoxytrichodiene to 9β, 10β;12,13-diepoxytrichodiene. This enzyme activity may be involved in the biosynthesis of trichothecene mycotoxins and since the 12,13-epoxide is known to be essential for toxicity, the enzyme activity probably confers the toxic properties associated with this group of mycotoxins. The epoxidase requires NADPH and molecular oxygen, is inhibited by carbon monoxide, and thus appears to be a cytochrome P-450-dependent mono-oxygenase. Whole cell cultures of the fungus carry out the same biotransformation, and in addition hydroxylate the diepoxide product at position 3, yielding 3α-hydroxy-9β,10β;12,13-diepoxytrichodiene.

    Notizen: Abstract Callus cultures have been established from root explants of aseptically-grown Podophyllum hexandrum seedlings. A fully defined medium based on Gamborg's B5 salts supplemented with 2/4-dichlorophenoxyacetic acid, gibberellic acid and 6-benzylaminopurine was effective for both initiation and sustained growth of callus tissue. Cultures produced anticancer lignans podophyllotoxin, 4′-demethylpodophyllotoxin and podophyllotoxin 4-O-glucoside at levels similar to those found in the expiant material as assayed by high performance liquid chromatography. The relative proportions of podophyllotoxin and 4′-demethyl-podophyllotoxin were markedly influenced by the presence of plant growth regulators. Particularly high levels of podophyllotoxin were associated with growth regulator induced tissue differentiation.

    Notizen: Abstract Fusarium sporotrichioides and Gibberella saubinetti O-demethylate 3,9-dimethoxypterocarpan to 3-methoxy-9-hydroxypterocarpan and then 3,9-dihydroxypterocarpan. F. anguioides Sherb., F. avenaceum and F. graminearum convert the same substrate to a mixture of 3-methoxy-9-hydroxypterocarpan and 3-hydroxy-9-methoxypterocarpan. Induction of pterocarpan degradation by pisatin in F. avenaceum leads to preferential conversion of 3,9-dimethoxypterocarpan to 3-hydroxy-9-methoxypterocarpan (medicarpin) and the isoflavan vestitol. The data are discussed with respect to phytoalexin degradation by phytopathogenic Fusarium fungi.
