    Notizen: Summary Starting withadhC mutants ofEscherichia coli in which alcohol dehydrogenase (ADH) and acetaldehyde CoA dehydrogenase (ACDH) are expressed constitutively at high levels, we selected mutants with still higher levels of both enzymes. Selection for growth on ethanol in the presence of inhibitors of ADH gave several mutants that had from 2- to 10-fold increases in the levels of both enzymes. These mutations were found to map far from theadhC locus at around 90 min. SuchadhR mutants were unable to grow on acetate or ethanol in certain media unless supplemented with extramanganese. This growth disability was suppressed by secondary mutations, one of which,aceX, increased sensitivity to several toxic metals and may perhaps derepress Mn transport. When theadhR mutation expressing the highest ADH and ACDH levels was present together withfadR andatoC mutations (allowing efficient catabolism of acetoacetyl-CoA) and with anaceX mutation, the resulting strains became capable of usingn-butanol as sole carbon and energy source. The use of butanol byE. coli illustrates the artificial evolution of a new catabolic pathway, in this case by the selection of four successive regulatory mutations (fadR, adhC, atoC, andadhR) together with the poorly definedaceX mutation. Each stage in the acquisition of this nove pathway confers the ability to use a new growth substrate: decanoic acid (fadR), ethanol (adhC), butyric acid (atoC), and butanol (adhR, when present withaceX).

    Notizen: Abstract A map of summer total phosphorus in lakes has been compiled for Minnesota, Wisconsin, and Michigan to clarify regional patterns in attainable lake trophic state. Total phosphorus was used as a measure of lake trophic state because: (1) phosphorus plays a central role in controlling the overall fertility of most lakes, (2) total phosphorus values are available for a great number of lakes, and (3) phosphorus is measured in a consistent manner. The maps were compiled using patterns of total phosphorus data and observed associations between these data and geographic characteristics including physiography, land use, geology, and soils. Regions depicted on the map represent areas of similarity in phosphorus concentrations in lakes, or similarity in the mosaic of values, as compared to adjacent areas. Within each region, differences in total phosphorus can be compared to natural and anthropogenic factors to determine the types of lakes representative of each region, the factors associated with differences in quality, and the realistically attainable phosphorus levels for each type of lake.

    Notizen: Abstract Field assessments of impacted streams require a control or at least an unbiased estimate of attainable conditions. Control sites, such as upstream/downstream or wilderness sites, have proven inadequate for assessing attainable ecological conditions where the control streams differ naturally from the impacted streams to a considerable degree or where different disturbances exist than those being studied. Relatively undisturbed reference sites with watersheds in areas having the same land-surface form, soil, potential natural vegetation, and land use as are predominant in large, relatively homogeneous regions are suggested as alternative control sites. These areas are considered typical of the region and therefore the sites also are considered typical of the region because their watersheds exhibit all the terrestrial variables that make that region a region. The logical basis for developing regional reference sites lies in the ability to group watersheds and common stream types into regions by integrating available maps of terrestrial variables that influence streams. Relatively undisturbed reference sites can be selected from typical areas of the regions and from transition zones where one or two of the terrestrial variables are not the predominant one(s) of the region. These reference sites are useful for estimating attainable conditions, for evaluating temporal and spatial changes in ecological integrity, for classifying attainable uses of streams, and for setting biological and environmental criteria.

    Notizen: Summary Mutants of Escherichia coli resistant to chloroethanol or to chloroacetaldehyde were selected. Such mutants were found to lack the fermentative coenzyme A (CoA) linked acetaldehyde dehydrogenase activity. Most also lacked the associated fermentative enzyme alcohol dehydrogenase. Both types of mutants, those lacking acetaldehyde dehydrogenase alone or lacking both enzymes, mapped close to the regulatory adhC gene at 27 min on the E. coli genetic map. The previously described acd mutants which lack acetaldehyde dehydrogenase and which map at 63 min were shown to be pleiotropic, affecting respiration and growth on a variety of substrates. It therefore seems likely that the structural genes for both the acetaldehyde and alcohol dehydrogenases lie in the adhCE operon. This interpretation was confirmed by the isolation of temperature sensitive chloracetaldehyde-resistant mutants, some of which produced thermolabile acetaldehyde dehydrogenase and alcohol dehydrogenase and were also found to map at the adh locus. Reversion analysis indicated that mutants lacking one or both enzymes carried single mutations. The gene order in the adh region was determined by three point crosses to be trp - zch:: Tn10 - adh - galU- bglY - tyrT - chlC.

    Notizen: Abstract The mean life times of negative muons bound to actinide nuclei have been measured by detecting the time difference between a stopped muon and the arrival of fragments from delayed fission after muon capture. The deduced capture ratesΛ c are 1.392(4)·107/s for237Np, 1.290(7)·107/s for242Pu and 1.240(7)·107/s for244Pu. The results are compared with published data for the fission and the neutron decay channels and for the electron decay of the bound muon. Including a former measurement ofΛ c for239Pu, an isotopic dependence of the muon capture rates in the Pu isotopes is clearly observed.

    Notizen: Abstract The excitation function of the fission probability P E E x) for238U has been measured in the reaction238U(α, α′ f) at 480 MeV bombarding energy. The reaction mechanism of this reaction is discussed for excitation energies belowB nf , the threshold for second chance fission, and aboveB nf up toE x =37 MeV. In comparing with results from fission induced by photons and by particle transfer reactions the (α, α′f) reaction gives too low values for the fission probabilityP f at excitation energies well aboveB nE . The role of the quasi-elastic knock-out process in this reaction is discussed.
