    Notes: Abstract A highly specific recognition system, capable of distinguishing between allogeneic (non-clonemates) and syngeneic (clonemates) conspecifics, exists in Anemonia sulcata (Coelenterata: Anthozoa). During a competitive interaction between allogeneic individuals, specialized structures (acrorhagi) are exposed and used to sting opponents. This complex behavior is called the acrorhagial response. In order to quantify this behavior we measured the acrorhagial response time (period from first contact with opponent to full expansion of acrorhagi). More than 320 reciprocal histoincompatibility responses were recorded. These histoincompatibility (H) markers are free components of the sea anemone mucus: they were characterized as glyco-proteins of a molecular weight 〈18 KD (Kilo-Dalton) containing terminal α-d-glucose and/or α-d-mannose. In “alloimmune memory” testing experiments, the “memory trace” in A. sulcata was found to persist for 5 d.

    Notes: Summary In two field experiments, plant materials labelled with 15N were buried separately within mesh bags in soil, which was subsequently sown with barley. In the first experiment, different parts of white clover (Trifolium repens), red clover (T. pratense), subterranean clover (T. subterraneum), field bean (Vicia faba), and timothy (Phleum pratense) were used, and in the second, parts of subterranean clover of different maturity. The plant materials were analysed for their initial concentrations of total N, 15N, C, ethanol-soluble compounds, starch, hemicellulose, cellulose, lignin, and ash. After the barley had been harvested, the bags were collected and analysed for their total N and 15N. In the first experiment the release of N was highest from white clover stems + petioles (86%) and lowest from field bean roots (20%). In stepwise regression analysis, the release of N was explained best by the initial concentrations of lignin, cellulose, hemicellulose, and N (listed according to decreasing partial correlations). Although the C/N ratio of the plant materials varied widely (11–46), statistically the release of N was not significantly correlated with this variable. The results of the second experiment using subterranean clover of different maturity confirmed those of the first experiment.

    Notes: Summary A technique of implantation of portal and hepatic venous catheters in miniature pigs is described in detail. To overcome early patency loss, Swan-Ganz balloon catheters were used. Portal as well as hepatic venous catheters could be held open to withdraw blood and to apply fluids over a period of at least 50 days. The collected hepatic venous blood represents mixed hepatic outflow. The method is recommended to study absorption of drugs and substrates from the gut and their metabolism in the liver of conscious unrestrained experimental animals.

    Notes: Summary For studies in physiology and biochemistry of the liver the measurement of hepatic blood flow is a fundamental precondition. Therefore, the least invasive method, the measurement of liver blood flow by indocyanine green (ICG) dye elimination, was re-evaluated for the miniature pig, an animal model of increasing interest. The accuracy of spectrophotometric determination of ICG was enhanced by addition of 5% of desoxycholic acid. It was shown that ICG was eliminated exclusively by the liver. An extraction rate of 30%–50% was found in mini-pigs, but an insufficient rate of 5%–10% in pigs of German landrace. A liver blood flow ranging from 70 to 160 ml/100g·min−1 was measured. These values are in good accordance with datas from simultaneous estimation of blood flow by133Xenon washout. As shown for a starvation/refeeding cycle, liver blood flow measurement by ICG combined with permanently implanted catheters is a practicable tool for studies of the metabolic and pharmacologic fate of substances resorbed from the gut.

    Notes: Summary The glucoregulatory function of glucagon was investigated in hypo-, eu- and hyperthyroid miniature pigs. Infusion glucagon, (3 ng x kg body weight−1 · min−1) transiently increased blood glucose (p〈0.01) and hepatic glucose production (p〈0.01) in euthyroidism, but was without effect in hyperthyroidism. Infusing glucagon plus somatostatin (2 ng x kg body weight−1 · min−1 and 0.2 μg x kg body weight−1 · min−1) transiently increased blood glucose (Δ 3.0 to 4.3 mmol/l) and hepatic glucose production (Δ 3.3 to 7.7 umol x kg body weight−1 · min−1) in all thyroid states, the effect was less pronounced in hyperthyroid pigs. By contrast, hypoglucagonaemia (74 to 107 pg/ml) at basal insulin (28 to 35 μU/ml) provoked hypoglycaemia (1.4 to 2.2 mmol/l) and a fall in glucose production (Δ 4.7 to 8.3 umol x kg body weight−1 · min−1), which was independent of the thyroid state; the effect was most pronounced in hyperthyroidism (p〈0.01). Hepatic glycogen content, arterial gluconeogenic precursor concentrations as well as the glycaemic response (Δ 0.60 mmol/l) to alanine infusion (23 umol x kg body weight−1 · min−1) were all unaffected by hyperthyroidism. We conclude that moderate experimental hyperthyroidism reduces glucagon action due to reduced glycogen mobilisation. This may in part result from increased insulin sensitivity.

    Notes: Summary Cyclosporin A toxicity on pancreatic B-cells and its prevention by rioprostil, a prostaglandin E1 analogue, were studied in the model of the isolated perfused pancreas of rats treated with both compounds for 8 days. At toxic doses of cyclosporin (10 and 20 mg/kg b.wt), the B-cells showed severe hydropic degeneration of the endoplasmatic reticulum and slight degranulation of the B-cells. Accordingly, the insulin secretion was markedly impaired. Administration of rioprostil ameliorated the insulin secretion significantly, but not the ultrastructural changes. At therapeutic levels of cyclosporin (5 mg/kg b.wt), the hydropic degeneration and the drop in insulin secretion were completely prevented by rioprostil. This observation might have therapeutic implications in the treatment of patients, in particular those undergoing pancreatic transplantation.
