    Notes: Extensive networks of metastable ions link the major peaks in the electron impact mass spectra of two crown ethers containing 2,6-pyrido units. High-resolution mass measurements and the metastable peaks allow the elucidation of the fragmentation pathways. The spectra are influenced more by the presence of aromatic substituents than by the 2,6-pyrido units.

    Notes: Summary The immunosuppressive activity of azathioprine (AZA) is unpredictable and depends on the formation of intracellular thiopurine ribonucleotides. However, the quantification of these active thiopurines presents difficult analytical problems. It has recently been postulated that plasma concentrations of 6-thiouric acid (6-TU) and 6-mercaptopurine (6-MP), metabolites of AZA, may provide more readily measurable indices of the pharmacologic activity of AZA. In order to evaluate the utility of 6-TU and 6-MP plasma concentrations in monitoring AZA therapy, we studied their pharmacokinetics in 6 renal transplant patients, and their in vitro immunosuppressive potency in a mixed lymphocyte proliferation assay. A peak plasma 6-TU concentration of 710.7 ng/ml was observed at 3.8 h after oral dosing. Good correlation was observed between the elimination t1/2 of 6-TU and serum creatinine, and between AUC over 24 h and serum creatinine. However, we did not observe a second peak in plasma 6-TU concentration that could be attributed to the degradation of active AZA metabolites. 6-MP plasma concentrations in the patients were low (mean peak concentration 36.0 ng/ml) and rapidly disappeared within 8 h. In vitro immunosuppressive activity could not be demonstrated for 6-TU over a concentration range of 1.25 ng/ml to 0.25 mg/ml. We conclude that 6-TU is pharmacologically inert and is primarily eliminated by the kidneys. Our findings currently do not support the use of plasma concentrations of 6-TU or 6-MP to monitor AZA therapy. In order to optimize AZA therapy, analytical techniques that are technically feasible and that can directly quantify the active intracellular thiopurines are being explored.

    Notes: Abstract The paper investigates quantitative differences in the signal characteristics of ventricular fibrillation (VF) and other cardiac arrhythmias. The analysis procedure comprises two steps: calculation of a short-term autocorrelation function (ACF) followed by a regression test on a plot of peak magnitudes of the ACF against lag values (the ACF/lag plot). We detect VF by testing the hypothesis that the ACF/lag plot of VF does not pass a linear regression test. Analysis of 31 separate episodes (of VF and other ventricular arrhythmias), each comprising three successive segments of 1·5s each produced the following results: (1) 100 per cent sensitivity (Se), 62 per cent specificity (Sp) and 74 per cent test efficiency (TE) after analysis of the first segment; (2) 100 per cent Se, 86 per cent Sp and 90 per cent TE after the second segment; and (3) 100 per cent Se, 100 per cent Sp and 100 per cent TE after the third segment. This method quantifies the notion that VF signals are nonperiodic with a random amplitude distribution, whereas ventricular tachycardia (VT) signals are usually periodic with more uniform amplitude distributions. Accurate discrimination and identification of VF can be very important in intensive-care settings, as well as in the design of automatic cardioverters and defibrillators.

    Notes: Negative-ion continuous-flow fast-atom bombardment mass spectroemtry was evaluated asa means for the quantitative analysis of N-acetylneuraminyl-galactosyl-glucosyl ceramide (NeuAc-GM3) and N-acetylgalactosaminyl-(N-acetylneuraminyl)galactosyl-glucosyl-ceramide (NeuAc-GM3). This study was carried out on a 7070-EQ mass spectrometer (VG Analytical, Manchester, UK) using a home-made continuous-flow fast-atom bombardment probe with a mixture of methanol +water+triethanolamine (70:27:3, v/v/v)as the mobile phase. Utilizing 100 ng of acetyl-lysogalactosyl-N-acetylgalactosaminyl-(N-acetylneuraminyl)galactosyl-glucosylceramide (acetyl-lysoGM1) as an internal standard curves for NeuAc-GM3 d18:1-16 0, NeuAc-GM3 d18:1-18:0 and Neuac-GM3 d18:1-18:0 were found to be linear over the range 5-250ng, with associated correlation coefficients of 0.990-0.997. The lower limit of detection was found to be 2.5ng. Satisfactory results could also de obtained when the calibration curves were derived from the deprotonated molecular ions of a mixture of the NeuAc-M2 and NeuAc-GM3 classes. Using this approach, quantitative determination of NeuAc-GM3 d18:1-16:0 from rat adrenal gland was performed sing N-acetylneuraminic acid assay as a test control. We found 278±36 ng of this species in 1 mg of tissue (three replicate experiments). The procedure represents a senstive method for the quantitation of mionosialogangliosides and its capability to give molecular species information.

    Notes: The polarized and depolarized Raman spectra of the ν2 (CH2 bend) and ν3 (C—Br stretch) vibrations in dibromomethane have been studied as a function of temperature throughout the liquid range. Reorientational correlation times derived from the two modes were simllar in both magnitude and dependence on temperature. This result, together with other structural data, suggests that the rotational dynamics of this molecule in the liquid phase are quasi symmetric-top in character. Vibrational dephasing times calculated from the two lineshapes were also of the same magnitude. However, modulation times determined via the Kubo lineshpae formalism varied markedly between the vibrations. This provides evidence that vibrational dephasing of the bending and stretching modes are influenced by different intermolecular forces in the liquid.
