    Notes: Summary 12-O-Tetradecanoylphorbol-13-acetate (TPA)-treated adult epidermis as well as untreated fetal and adult epidermis were investigated to elucidate the effect of TPA in terms of cell differentiation using techniques of ultrastructural stereology. Twenty-four hours after a single application of TPA, the treated epidermis was characterized by involutional changes, i. e., increased volume density of intercellular spaces and of mitochondria, vacuoles, and cytoplasmic ground substance in the basal layer. However, 48 h after application, the TPA-treated epidermis was very similar to fetal epidermis, i. e., high volume density of nuclei ribosomes, rough endoplasmic reticulum, membrane-coating granules, and keratohyalin granules, and low volume density of bundled filaments in the upper layers. These stereological data indicate that the changes observed 48 h after TPA treatment were related not only to increased cell proliferation but also to inhibition of cell differentiation expressed as a reversion of the adult differentiation patterns and the acquisition of fetal characteristics in all epidermal layers.

    Notes: Abstract The mean minimal capillary transit time was estimated in muscles of various animals using a combination of physiological and morphometric methods. Radioactive microspheres were injected intravascularly in various animals running on a treadmill at maximum oxygen consumption rate (VO2,max) to label blood flow to individual muscles. The muscles were then removed and preserved by standard methods for electron microscopy. The volume density of mitochondria was measured to assess muscle oxidative capacity. Capillary densities in muscle cross-sections, capillary diameters and tortuosities were incorporated into an estimate of capillary volume per unit muscle mass. Mean capillary transit time (t c) in the exercising muscles was estimated by dividing mass-specific capillary volume by mass-specific blood flow. Estimates of t c ranged from values near 1 s in horse heart and thigh muscles to 0.2 s in duck gastrocnemius. The relationship between muscle blood flow and t c was hyperbolic. The experimental data indicate a limiting value of 0.2 s for transit times at very high blood flows. There was no correlation between t c and body-mass-specific VO2,max.

    Notes: Abstract The relationship between maximal oxygen consumption rate ( $$\dot V{\text{O}}_{{\text{2max}}}$$ ) and mitochondrial content of skeletal muscles was examined in horses and steers (n=3 each). Samples of the heart left ventricle, diaphragm,m. vastus medialis, m. semitendinosus, m. cutaneous thoracicus andm. masseter, as well as samples of muscles collected in a whole-body sampling procedure, were analyzed by electron microscopy. $$\dot V{\text{O}}_{{\text{2max}}}$$ per kilogram body mass was 2.7× greater in horses than steers. This higher $$\dot V{\text{O}}_{{\text{2max}}}$$ was in proportion to the higher total volume of mitochondria in horse versus steer muscle when analyzed from the whole-body samples and from the locomotor muscle samples. In non-locomotor muscles, total mitochondrial volume was greater in horses than steers, but not in proportion to their differences in $$\dot V{\text{O}}_{{\text{2max}}}$$ . The $$\dot V{\text{O}}_{{\text{2max}}}$$ of the mitochondria was estimated to be close to 4.5 ml O2·ml−1 mitochondria in both species. It is concluded that in a comparison of a highly aerobic to a less aerobic mammalian species of similar body size, a higher oxidative potential may be found in all muscles of the more aerobic species. This greater oxidative potential is achieved by a greater total volume of skeletal muscle mitochondria.

    Notes: Four novel symmetric tri-n-alkyltin fluorides were prepared. In this homologous series, melting point decreases with increase in the carbon chain length while an x-ray diffraction study shows an increase in amorphousness. 1H-NMR spectra in cyclohexane, a nonpolar solvent in which these compounds form long polymeric chain, show a peak broadening effect due to high solution viscosity at very low solute concentration. Contrary to this, the spectra in CDCl3, a polar solvent in which these compounds are monomeric, show well resolved and narrow peaks. IR frequencies for Sn—C vibrations have been assigned.

    Notes: Copolymers of styrene-methyl methacrylate were irradiated in vacuo with γ-rays; viscosity changes and gas yields (moles gas per mole methyl methacrylate in copolymer) for H2, CO, CO2, CH4, HCOOCH3, and CH3OH were examined at doses of 1.0 Mrep to 10.0 Mrep. It was found that styrene molecules protect the copolymer from degrading while decreasing the gas yields up to a styrene concentration of about 0.5 mole fraction; above this concentration, these radiation effects are independent of composition. When styrene-methyl acrylate copolymers were irradiated, styrene offered protection aganist gelation but simultaneously increased the gas yields (gases liberated were identical with those from the first system).

    Notes: The distribution of Type I and Type II fibers, as determined from histochemical estimation of myofibrillar ATPase activity, was studied within and among the locomotory muscles of the forelimb, trunk, and hindlimb of three mongrel dogs. All Type II fibers had high oxidative capacities as estimated from the histochemical assay for reduced nicotinamide adenine dinucleotide tetrazolium reductase, so they were not further divided into subpopulations. Furthermore, Type I and Type II fibers had similar oxidative potentials as indicated by both histochemistry and biochemistry.Type I fiber populations ranged between 14% and 100% in the muscles sampled. The highest percentages of Type I fibers were found in deep muscles of physiological extensor groups in the arm and thigh that serve to resist gravity (antigravity muscles) when the dog is in the quadrupedal standing position. More superficial muscles in these same groups had fewer Type I fibers. The patterns of Type I fiber distribution among muscles in the antigravity groups of the forearm and leg were the opposite of those in the arm and thigh, with the more superficial muscles of the distal limb segments having more Type I fibers than the deeper muscles. In all limb segments, muscle groups that do not serve to resist gravity did not show as much intermuscular variation. Type I fiber populations in these muscles did not exceed 50%. A stratification of fiber types also existed within muscles, both in extensor and flexor groups, with the deeper portions of the muscles having more Type I fibers than the more superficial portions.

    Notes: Optical indicators of the cationic, cyanine and anionic oxonol classes were used to evaluate the plasma membrane potential of animal cells in suspension and in monolayer culture. The optical signals were calibrated by using diffusion potential either of K+ (in the presence of valinomycin) or of H+ (in the presence of carbonyl cyanide p-trifluoromethoxyphenylhydrazone; FCCP); both classes of dye gave similar values of plasma membrane potential, in the range -40 to -90 mV for different cell types. Addition of haemolytic Sendai virus or Staphylococcus aureus α-toxin depolarizes cells and causes them to leak monovalent cations; these effects are antagonized by extracellular Ca2+. Cells infected with vesicular stomatitis or Semliki Forest virus become depolarized during an infectious cycle; infection with other viruses was without affect on plasma membrane potential.
