    Notes: Globally coherent modes have been observed during formation in the S-1 Spheromak plasma [Plasma Physics and Controlled Nuclear Fusion 1984 (IAEA, Vienna, Austria, 1985), Vol. 2, p. 535] by analysis of magnetic field fluctuations measured from outside the plasma. The modes are of low n number (2≤n≤5), where n is defined by the functional dependence einφ of the fluctuation on toroidal angle φ. These modes are shown to be related to flux conversion and plasma relaxation toward a minimum-energy state during the spheromak formation. The modes are active while the q profile is rapidly changing, with q on axis, q0, rising to 0.7. A significant finding is the temporal progression through the n=5, 4, 3, 2; m=1 mode sequence as q rises through rational fractions m/n. During formation, peak amplitudes of the n=2, 3, 4 modes relative to the unperturbed field have been observed as high as 20%, while more typical amplitudes are below 5%.

    Notes: Experimental measurements of the equilibrium in the S-1 spheromak [M. Yamada, J. Sinnis, H. P. Furth, M. Okabayashi, G. Sheffield, T. H. Stix, and A. M. M. Todd, in Proceedings of the US-Japan Symposium on Compact Toruses and Energetic Particle Injection (Princeton Plasma Physics Laboratory, Princeton, NJ, 1979), p. 171] by use of magnetic probes inside the plasma show that the final magnetic equilibrium is one that has relaxed close to the Taylor (minimum-energy) state, even though the plasma is far from that state during formation. The comparison is made by calculating the two-dimensional μ profile of the plasma from the probe data, where μ is defined as μ0 j(parallel)/B. Measurements using a triple Langmuir probe proved evidence to support the conclusion that the pressure gradients in the relaxed state are confined to the edge region of the plasma.

    Notes: The histogenesis of hidrocystomas was examined by the use of immunostaining for S–100 protein. In normal sweat glands, S–100 protein was found exclusively in the secretory cells of eccrine glands, whereas this protein was not present in the other parts of eccrine glands or at any levels of the structure of apocrine glands. On the bases of this immunostaining pattern in normal sweat glands, we attempted to correlate the origin of 8 cases of hidrocystoma to the presence of S–100 protein-positive cells. S–100 protein was detected in the cells of one solitary eccrine hidrocystoma, but not in those of 2 cases of “classic”, multiple-lesion type of eccrine hidrocystoma. This indicated that the former arose from the secretory portion of the eccrine gland and the latter from the eccrine ductal cells. Two of the 5 cases of apocrine hidrocystoma showed positive staining in a part of the lining cells of the cyst wall, while the other 3 cases were negative to this protein. This finding suggests that some of the tumors diagnosed morphologically as apocrine hidrocystoma differentiate in the direction of eccrine secretory cells. In addition to S–100 protein, we also surveyed for the presence of carcinoembryonic antigen (CEA), and all cases examined were consistently positive to this substance. The detection of S–100 protein was considered to be more helpful in classifying hidrocystomas than that of CEA.

    Notes: The nucleotide sequence of a Salmonella typhimurium DNA segment of 549 base pairs which encompasses the operator-promoter of the pts operon, the entirety of the ptsH gene, encoding HPr of the phosphotransferase system (PTS), the first 29 nucleotides of the ptsl gene, encoding Enzyme I of the PTS, and the intercistronic region between the ptsH and ptsl genes was determined and compared with the corresponding sequence from Escherichia coli (De Reuse et al., 1985). The two sequences showed 91 % overall identity, with some regions showing sequence conservation and others exhibiting relative divergence. Two open reading frames were identified in both species: one encoded HPr on the ‘sense’ strand (255 nucleotides; 12 nucleotide differences, no amino acid differences); the other, on the anti-sense strand, consisted of 291 nucleotides (13 nucleotide differences, 13 amino acid differences). While HPr bears a net negative charge, the putative protein encoded by the open reading frame on the anti-sense strand is strongly basic.Computer analyses of HPr proteins from five different bacterial genera revealed four regions which show strong sequence identity and therefore are presumed to be critical for maintenance of biological activity. Two of these regions were specific to Gram-positive bacteria. Proposed functions for each of these regions are discussed. Relative evolutionary distances between the HPr proteins were also computed.

    Notes: The nucleotide sequence of the known portions of the mannitol operon in Escherichia coli (mtlOPAD) has been determined. Both the operator-promoter region and the intercistronic region between the mtlA and mtID genes (encoding the mannitol-specific Enzyme II of the phosphotransferase system and mannitol–1- phosphate dehydrogenase, respectively) show parallels with corresponding regions of the glucitol (gut) operon, but neither the mtlA nor the mtlD gene products show obvious homology with the corresponding gene products of the glucitol operon. Five potential cyclic AMP receptor protein binding sites were identified in the mtlOP region, all showing near identity with the consensus sequence. Four regions of dyad symmetry (four to seven bases in length), serving as potential repressor binding sites, overlap with the potential cyclic AMP receptor protein binding sites. Repetitive extragenic pallndromic (REP) sequences, forming stem-loop structures in the intercistronic region between mtlA and mtlD and following the mtlD gene were identified. Probable terminator sequences were not found in any of these three regulatory regions. Mannitol–1-phosphate dehydrogenase exhibits two overlapping, potential NAD+ binding sites near the W-terminus of the protein. Computer techniques were used to analyse the mtlD gene and its product.

    Notes: In Shigella flexneri, in addition to several well-recognized plasmid-borne virulence loci, at least three genetic loci implicated in pathogenesis have been recognized on the chromosome. To understand more about the pathogenesis of bacillary dysentery at a molecular level, the genetically recognized but previously unidentified KcpA region (one of the chromosomal regions near purE) was cloned and sequenced. A single translatable open reading frame encoding a 12310 Dalton protein corresponding to the minicell product was found. Immunofluorescence microscopy, as well as optical and electron microscopic comparison of tissue-cultured cells and guinea-pigs’eyes infected with wild-type or kcpA mutant bacteria, revealed that the kcpA product is required by invading bacteria for spread into adjacent cells.

    Notes: Measurements of cell cycle kinetics using an immunoperoxidase method employing monoclonal anti-bromodeoxyuridinc (BrdU) antibody were compared with an autoradiographic method using [3H]-thymidine. The methods were applied to epithelial cells grown from explants of normal skin and basal cell carcinoma (BCC), and to fibroblasts from normal skin, hypertrophic scar and keloid.In normal keratinocytes the number of peroxidase-positive cells was higher than the number incorporating [3H]-thymidine, because of the presence of labelled cells in the centre of the explants in the former. The epithelial cells from BCC gave a mean (± SD) of 4.9 ± 1.2% peroxidase-positive cells, while no cells were labelled with [3H]-thymidine. In dermal fibroblasts from normal skin and hypertrophic scar the percentages of peroxidasc-positive cells did not differ significantly from the [3H]-thymidine labelling indices.The immunological method has advantages over [3H]-thymidine autoradiography in that it avoids radioactive material and the proportion of cells labelled by the two methods is the same.

    Notes: Alterations of binding capacity of surface IgG-Fc and complement receptors were demonstrated in polymorphonuclear neutrophils (PMNs) obtained from the pustular lesions of psoriasis. A marked decrease of C3 receptors, but not of IgG-Fc, was found in PMNs from the lesions of palmo-plantar pustulosis (PPP) and bacterial pustules. PMNs from pustular lesions of psoriasis exhibited only a slight decrease in the number of C3 receptors. No significant decrease in membrane receptors was noted in circulating PMNs from psoriatic patients. We suggest that mechanisms of formation of aseptic subcorneal pustules, mediated by PMN membrane receptors for C3 fragments, are different in pustular psoriasis and PPP.
