    Notes: Abstract Human Recombinant IL-1 was investigated for its ability to increase non-specific resistance toStaphylococcus aureus in neutropenic mice. Mice, rendered neutropenic with cyclophosphamide and then administered IL-1 intraperitoneally, demonstrated enhanced resistance to subsequent challenge withS. aureus as measured by increased survival and bacterial clearance. No protective effects were observed with heat inactivated IL-1. Efficacy was observed only when both IL-1 and the subsequent bacterial challenge were administered into the same site. Despite the observed protective effects, animals treated with IL-1 did not have increased numbers of blood leukocytes or peritoneal phagocytes prior to infection or at the times coincident with bacterial clearance. Based upon these observations, enhanced activity of resident macrophages may be responsible for the protective effects observed in IL-1 treated mice.

    Notes: Abstract Smooth muscle cells were cultured from guinea-pig aorta and labelled with45Ca++ and32Pi to investigate the possible effect of cicletanine, a new antihypertensive drug, on the release of intracellular Ca++ and the metabolism of phosphoinositide induced by histamine. In45Ca++ labelled cells, histamine increased in a dose-dependent manner the45Ca++ efflux in the first two minutes. Stimulation of45Ca++ release was observed with H1-agonist [2-pyridylethylamine dihydrochloride (2-PEA)] but not with H2-agonist (dimaprit). In addition, histamine- or 2-PEA- induced45Ca++ efflux was inhibited by the H1-antagonists (mepyramine and terfenadine) whereas the H2-antagonist (cimetidine) was without effect. Similar results were obtained in32Pi labelled cells; both H1-agonists (Histamine and 2-PEA) increased the labelling of phosphoinositides. This effect was completely blocked by mepyramine. These results demonstrate that the histamine-induced stimulation of45Ca++ efflux and phosphoinositide metabolism are mediated through H1-receptors. In the above systems, cicletanine was as effective as the H1-antagonist (mepyramine) with an IC50 of 10−6 M for both45Ca++ efflux and phosphoinositide metabolism. Blockade of these systems by cicletanine may be part of the mechanism by which this drug produces relaxation of blood vessels and may account for itsin vivo antihypertensive action.

    Notes: Summary We propose a fast Monte-Carlo algorithm for calculating reliable estimates of the trace of the influence matrixA τ involved in regularization of linear equations or data smoothing problems, where τ is the regularization or smoothing parameter. This general algorithm is simply as follows: i) generaten pseudo-random valuesw 1, ...,w n , from the standard normal distribution (wheren is the number of data points) and letw=(w 1, ...,w n ) T , ii) compute the residual vectorw−A τ w, iii) take the ‘normalized” inner-product (w T (w−A τ w))/(w T w) as an approximation to (1/n)tr(I−A τ). We show, both by theoretical bounds and by numerical simulations on some typical problems, that the expected relative precision of these estimates is very good whenn is large enough, and that they can be used in practice for the minimization with respect to τ of the well known Generalized Cross-Validation (GCV) function. This permits the use of the GCV method for choosing τ in any particular large-scale application, with only a similar amount of work as the standard residual method. Numerical applications of this procedure to optimal spline smoothing in one or two dimensions show its efficiency.

    Notes: Summary An increase in free cytosolic calcium content has been reported in essential hypertension. Since within the membrane, the phosphoinositides participate in the control of cell calcium homeostasis, we investigated whether impaired phosphoinositide metabolism could account for the calcium handling abnormality observed in hypertensives. In erythrocyte membranes of hypertensives the activity of kinases involved in polyphosphoinositide formation appears to be impaired and could be related to the alteration in calcium binding capacity and ATP-dependent calcium transport. In platelets of hypertensives, the hyperactivity of phospholipase C (observed even in the absence of calcium in the external medium) is likely to be responsible for the hypersensitivity of cells to various agonists. These observations are consistent with the hypothesis that in cells from hypertensives, a membrane defect linked to phosphoinositide metabolism is involved in the overall calcium handling defect.

    Notes: The Raman spectra of ferroelectric [NH(CH3)3]3Sb2Cl9 single crystals were studied at temperatures ranging from 90 to 390 K. An assignment of most of the observed bands is proposed. A second-order phase transition around 364 K leading to a high-temperature paraelectric phase manifested itself by spliting of the antisymmetric CH3 stretching band (3035 cm-1) below the transition temperature. A temperature dependence study revealed the existence of disorder in the crystal. A systematic measurement of the CH3 torsional vibration Raman band (305 cm-1) was performed and the activation energy (Ea = 2.39 kcal mol-1) of an unidentified organic group HN(CH3)3+ motion was extracted from the half-width versus temperature curve.

    Notes: Polarized Raman and IR spectra of lattice vibrations (10-260 cm-1) in the m-aminophenol crystal have been recorded at various temperatures in the range 20-300 K. LO-TO splittings of about 2 cm-1 have been observed for two modes of A1 symmetry. Lattice dynamic calculations of frequencies and calculations of Raman intensities based on the generalized oriented gas model have been performed. A discussion of the influence of hydrogen bonding on the lattice dynamics is presented.
