The fine structure of the paraxial rod of Phytomonas davidi and Herpetomonas megaseliae was analyzed in thin sections of Triton X-100-extracted, tannic acid-glutaraldehyde-fixed cells and in replicas of quick-frozen, freeze-fractured, deeply etched and rotary shadowed cells. The paraxial rod is formed by a complex array of filaments. Two regions, designated as proximal and distal, are formed by two and at least 11 plates, respectively, composed of an association of 25-nm and 7.0-nm-thick filaments which are oriented at an angle of-50° in relation to the major axis of the axoneme. The intermediate region is less dense and is formed by thin filaments. Short single and Y-shaped filaments connect the proximal plate to doublets numbers 4 and 7 of the axoneme. Based on the images obtained in stereopairs as well as in photographs obtained before and after inclination of the specimen, a three-dimensional model of the paraxial rod is proposed.

    Notes: The fine structure of trypanosomatids from the plants Euphorbia hyssopifolia, E. characias, E. pinea, and Manihot esculenta, cultivated under axenic conditions, was compared. All parasites had a structural organization characteristic of members of the Trypanosomatidae. The organization of the membranes of the endoplasmic reticulum (ER) varied according to the isolate. In Phytomonas francai (isolated from M. esculenta), the ER cisternae form parallel rows. In Phytomonas sp. from E. characias, a para-crystalline array of the ribosomes attached to the ER membranes was observed. The ER membranes found in Phytomonas sp. from E. pinea seemed to originate from the central portion of the protozoon, branching in all directions. The peroxisome-like organelle (glycosome) found in Phytomonas sp. from E. hyssopifolia and E. characias occasionally showed an organized array. Taken together, the morphological observations make it possible to distinguish the four isolates of Phytomonas.

    Notes: Summary The replication of Mayaro virus inAedes albopictus cells, was studied by electron microscopy at various times post-infection. In infected cells we observed the presence of cytoplasmic vesicles containing viral nucleocapsids and mature virus particles but at no time did we detect virus budding into such vacuoles. Budding of virus through plasma membrane was rarely observed. Our results are discussed considering the possibility of the release of virus particles to the extracellular space by exocytosis.

    Notes: Abstract Significant differences were found in the ability of resident mouse peritoneal macrophages to ingest amastigote and promastigote forms of Leishmania mexicana amazonensis. Differences in the association index of the parasites to the macrophages were also found between infective and non-infective promastigotes. Evidence was obtained suggesting that the macrophage receptor, which recognizes mannose-6-phosphate-containing units found in lysosomal enzymes, is involved in the association with the macrophage of promastigotes, but not of amastigotes. Addition of mannose-6-phosphate, its structural analogue fructose-1-phosphate, Hansenula holstii phosphomannan or the mannose-6-phosphate-containing lysosomal enzyme α-D-mannosidase to the interaction medium, markedly inhibits the association of the parasites with macrophages.

    Notes: Abstract The process of interaction of bloodstream trypomastigotes from the myotropic CL and Colombiana strains and the macrophagotropic Y strain ofTrypanosoma cruzi with mouse myoblasts and myotubes was analysed. After 24 h of parasite-host cell interaction, parasites from the CL and Colombiana strains appeared to be more infective to myoblasts than those from the Y strain. Parasites from the Colombiana strain were more infective for myotubes than those from the Y strain, while those from the CL strain showed very a low ability to infect the cells. For all strains the infectivity was low for short periods of interaction, increasing with time. Myoblasts infected with parasites from the Y strain fused with other infected and uninfected cells to form myotubes. However, the process of fusion was blocked when the myoblasts were infected with parasites from the CL and Colombiana strains. These data indicate a different behavior of muscle cells when in contact with myotropic or non-myotropic strains ofT. cruzi.

    Notes: Abstract Epimastigotes of Trypanosoma cruzi showed many filopodium-like projections from the flagellar membrane when incubated in the presence of 100 ng/ml (a concentration which does not interfere with cell motility and viability) phorbol-12-myristate-13-acetate (PMA). PMA is a substance which binds to the membrane-associated protein kinase C. Few of these projections were observed in control parasites. PMA, when added to the interaction medium, significantly increased the attachment of epimastigotes to the surface and their ingestion by resident or activated mouse peritoneal macrophages maintained in vitro. PMA, however, did not interfere with the ingestion of trypomastigotes.

    Notes: Abstract NADH- or NADPH-oxidase activity was cytochemically detected at the ultrastructural level during the process of interaction betweenTrypanosoma cruzi and activated mouse peritoneal macrophages. The reaction product, indicative of enzyme activity, was found in the portion of the plasma membrane of the macrophages to which the parasites attached. It was also found in the membrane which surrounds endocytic vacuoles containing ingested parasites.

    Notes: Abstract When epimastigote forms ofTrypanosoma cruzi grown in a rich medium (LIT) are transferred to a simple, chemically defined medium (TAU3AAG, containing Ca2+, Mg2+, K+, Na+,l-proline,l-glutamate, andl-aspartate in phosphate buffer, they transform into trypomastigote forms. Morphometric analysis of transmission electron micrographs of thin sections of parasites collected at different steps of the transformation process showed that no changes occurred in the volume density of mitochondria and cytoplasmic vacuoles. However, a significant increase in the volume density of the kinetoplast DNA network as well as the lipid inclusions and a decrease in that of the reservosome (a special type of endosome) was observed. These observations suggest that during differentiation,T. cruzi accumulates lipids and uses molecules contained in the reservosome as its main energy source.

    Notes: The structure, relative density, and distribution of anionic sites on the surface of epididymal and ejaculated spermatozoa were studied using polycationic ferritin (CF), colloidal iron hydroxide (CIH), various enzymatic treatments, methylation, and de-acetylation. Macro-molecules containing sugar residues, probably sialic acid, are part of the sperm membrane and show a characteristic distribution and density that is dependent of the sperm region and of its origin. Unlike the spermatozoa of other eutheria examined, the exposure of the stallion spermatozoa to neuraminidase treatment did not produce significant changes in the density of the negative charge of the sperm surface. The ability of purified neuraminidase to act only after saponification suggests that sialic acid may be present in the acetylated form. When CIH was used it is seen that the density of the negative charge is rather uniform within a particular segment of the spermatozoa and abruptly changes at the junction of morphologically distinct segments (Between the acrosomal and post acrosomal region of the sperm head and between the post acrosomal region and middle piece of the flagellum). The acrosome presented more negative groups dissociated at pH 1.8 than the postacrosomal region. A greater concentration of anionic sites over the flagellum was also observed when CIH and CF were used. This assymetry probably represents different domains that may be related to specific functions.The cytochemical observations and the cellular electrophoretic mobility measurements did not show striking differences on the negative charge of sperm obtained from different regions of epididymis and ejaculates in contrast to previous results in other species. The spermatozoa collected from caput epididymidis bind CIH but not all population present equal response. In corpus and cauda region of epididymis the population displaying the capacity to bind CIH or CF significantly over the head and tail surface was the majority.This study corroborates that the distribution and density of terminal oligosaccharide residues on the sperm plasma membrane has species specific characteristics. The surface charge of the spermatozoa obtained either during the breeding or nonbreeding season, determined by measurements of cellular electrophoretic mobility and by the binding pattern of CIH and CF, does not show significant differences.
