Notes: Localization of IgA, secretory component (SC) and J chain was investigated immunocytochemically in minor salivary glands of the lip and palate to define the mechanism involved in the transport of immunoglobulin A (slgA) into the saliva from the minor salivary glands. SC synthesis was detected in mucous acinar cells and ductal epithelial cells. Free SC is secreted into the saliva through secretory granules in the mucous acinar cells. Dimeric IgA containing J chain is translocated through these cells as slgA by a SC-medialed transport mechanism involving cytoplasmic vesicles.

Notes: Summary A highly sensitive and specific method for staining exogenous chromium and iron in tissues is described. This method is superior to conventional complex-forming methods with regard to its sensitivity and specificity for these metals. The staining reaction is based on the metalcatalysed oxidation of phenylamines. Tissue sections were incubated in a medium containing hydrogen peroxide and phenylamines, p-phenylenediamine or phenylhydrazine. Results obtained from test-tube experiments concerning the catalytic activities of metals indicated that the staining reactions depends on the activities of metals in tissues.

Notes: Summary We describe a new autoradiographic technique for differentially demonstrating the presence of trace amounts of cadmium that remains unbound to metallothionein in tissue sections. This procedure involves [14C-1,4,5,8--naphthol]-1-(2-thiazolylazo)-2-naphthol (TAN) instead of a metal radioisotope. Tissue sections were stained using labeled TAN, and were then subjected to routine autoradiographic procedures. The results obtained indicated that the distribution of photographic grains truly reflected the distribution of metallothionein-unbound Cd in the sections. This demonstrates the suitability of this procedure for the differential demonstration of cadmium present in different chemical forms in tissues.

Notes: Abstract The thumb-nail shape of a crack front has been observed in a compact tension (CT) specimen, when a fatigue crack is formed ahead of the machined notch in the course of preparation for the fracture toughness testing. Assuming that the distribution of the stress intensity factor K I along the crack front line is equal to that corresponding to the original straight crack front, a slightly curved crack front is obtained with the aid of Paris' law for the fatigue crack growth rate. Comparing the calculated results with the experimental observation in the 10 T CT specimens, whose thickness is 247 mm, it is found that the effect of the stress singularities at the intersection of the crack front line and the free surface can be disregarded and that they show extremely good agreement with each other.
