However, this compound shows neither diuretic nor hyperglycemic effect, being different from those of hydrochlorothiazide or diazoxide.

    Notes: Summary The effect of four exogenous prostaglandins, PGA1, PG802, PGE2 and PGF2α, upon the lower urinary tract was investigated in female mongrel dogs without neurogenic lesions in vivo. The urethral resistance was studied by means of a urethral pressure profile, and the bladder function by evaluating whether or not the micturition was triggered. The reduction of urethral resistance in terms of the maximum urethral closure pressure was most significant with PGE2 given intraarterially. Micturition was most frequently provoked by the intravenous administration of PG802, a derivative of PGE1. PGE series seemed to be the most potent for the evacuation of urine in female dogs.

    Notes: Summary The accelerated transport of the blood neutral amino acids into the brain in encephalopathic patients with fulminant hepatitis and advanced liver cirrhosis was demonstrated not only by determining the cerebrospinal fluid (CSF) aminogram but also by calculating the predicted velocity of the amino acid transport through the blood-brain barrier. Significant elevation in CSF aromatic amino acid (AAA) and methionine levels was observed in the encephalopathic patients. Arousal from hepatic encephalopathy by drip infusion of a branched chain amino acid (BCAA)-enriched solution was obtained coincidentally with the elevated BCAA levels and diminished concentrations of AAA and methionine in CSF. These clinical observations were confirmed experimentally in rats treated with carbon tetrachloride (CCl4) andd-galactosamine by obtaining the elevation of neutral amino acid contents in the brain and the slight increase in the brain uptake index (BUI) of a radiolabeled amino acid.

    Notes: Summary A new animal model of retinoblastoma was developed in newborn inbred CDF rats by intravitreous inoculation of retinal tumor cells (5×104/5 μl) derived from the cultured tumor cell line EXP-5. The retinal tumor from which the cell line originated was induced by a single intravitreous inoculation of human adenovirus serotype 12 (5 μl of 108 TCID 50/0.1 ml) in syngeneic rats. Within 1 month after intravitreous moculation of EXP-5 cells, a clinically recognizable ocular tumor was obtained in all 39 rats. Ad 12-specific T-antigens were demonstrated in the cultured tumor cells using immunofluorescent techniques. Morphologically these tumor cells closely resembled retinoblastuma, with poorly differentiated intracytoplasmic organelles, solitary cilia with a 9+0 tubule pattern, and abnormal nuclear membrane associated with a set of basal bodies. The significance of this highly manipulable retinal tumor cell line is the capability of providing a full-fledged intravitreous tumor in 1-month-old CDF rats, whose actual life span is known to be 42 months. Transplantable retinal tumors described to date are reviewed breifly and compared with the presently reported cell line.

    Notes: Summary The purification of a substance which protects mitochondrial activity against its decay in association with various cerebral pathologies, and the effect of this substancein vitro andin vivo have been mentioned. Defatted egg albumin was hydrolyzed with pronase, and diafiltrated through mesh, to obtain fractions of molecular weight less than 5,000. The diafiltrate was further fractionated using Sephadex G-25 column chromatography, and a fraction which had a protective action against the decay of mitochondrial DNP-ATPase activity was gathered. This digested egg albumin (DEA 5,000 S) showed no immunological reaction and seemingly penetrated well through the cell membrane. DEA 5,000 S prevented the decay of DNP-ATPase activity and swelling of brain mitochondria during agingin vitro. Also, the administration of DEA 5,000 Sin vivo shortened the duration of unconsciousnees and reduced EEG abnormalities in rats subjected to hypoxia in a special chamber filled with N2 gas. It is suggested that this Digested Egg Albumin has a marked action in restoring the function of metabolically impaired brain.

    Notes: Summary Glucose oxidation and lipogenesis were studied in isolated adipocytes from control and nonobese, experimentally hyperinsulinaemic rats. In cells from the hyperinsulinaemic animals oxidation of either [1-14C]- or [6-14C] glucose was increased in the presence or absence of insulin, at substrate concentrations from 0.1 to 20 mmol/l. Glucose incorporation into total triglycerides and fatty acids was also increased. These enhanced rates of glucose metabolism were due to increased activity of the glucose transport system in addition to increased activity of intracellular glucose metabolism. Therefore, these data indicate that insulin can influence long term glucose homoeostasis by augmenting the overall cellular capacity for glucose metabolism at several loci.

    Notes: Summary The ability of insulin and the S-sulphonate A and B-chain derivatives to bind to a receptor on cultured human lymphocytes was evaluated. A receptor site for the S-sulphonate A-chain was identified and was strongly influenced by the intact insulin molecule. S-sulphonate A-chain weakly interfered with insulin binding. S-sulphonate B-chain showed no evidence of significant binding and did not interfere with insulin or S-sulphonate A chain binding.14CO2 production from14C-1-glucose was stimulated by insulin in cultured lymphocytes and this effect was blunted by S-sulphonate A-chain. The sulphhydryl blocking agent used in the production of insulin A-chain appears to be of critical importance.

    Notes: Summary Trypsin-catalyzed coupling of porcine desoctapeptide-insulin with synthetic octapeptides produced the [LeuB24]-(I), [LeuB25]- (II) and [LeuB24, LeuB25]- (III)analogues of human insulin. I, II and III displayed respectively 20–30%, 1–2% and 0.5% of the receptor binding activity of the normal hormone. Biological activities of these analogues seemed to be proportional to their binding potencies when assayed in vitro, while in an in vivo assay analogue I was fully active and II exhibited 10–20% of normal activity. III was less active than II in all assays tested.

    Notes: Summary The authors have developed a new method of magnification cerebral angioautotomography which is performed by the use of a special head-swing apparatus. From the results of experiences of 24 intracranial aneurysms in 15 patients, the method has proved useful for the intracranial aneurysms in detecting the shape and the stalk of aneurysms which were obscured or hidden on conventional magnification cerebral angiography.

    Abstract: Zusammenfassung Die Verfasser berichten über eine neue Methode der Autotomographie für die zerebrale Angiographie. Mit dem besondern Apparat für Autotomographie, gewinnt man die willkürlichen tomographischen Bilder des Angiograms. Obgleich dieser Apparat einfach und handlich ist, sind die Tomogrambilder klar und gut. Dieser Bericht beschreibt das Prinzip und die Gebrauchsmethoden dieses Apparats.
    Notes: Summary A new method of autotomography for cerebral angiography, accomplished by means of a simple and handy device, has made it possible to take arbitrary tomographic planes of angiograms which result in clear cut views of the tomogram. The principle of the device is described, the method of use explained and representative films, produced by this new method, are demonstrated.
