    Notes: Abstract Vacuoles isolated from storage root tissue of red beet (Beta vulgaris L.) do not leak significant quantities of betanin, sucrose, Na+ or K+ during isolation. This indicates that analysis of vacuoles in vitro gives meanigful information about the compartmentation of solutes in vivo. Preparations of vacouoles were used to determine the distribution of glycinebetaine and proline between vacuole and cytoplasm in beet cells. Both compounds were detected in preparations of isolated beet vacuoles. In the case of glycinebetaine it was shown that this solute was associated with the vacuoles, not with the small number of other organelles which contaminated the preparations. The vacuolar pool accounted for 26 to 84% of the total tissue glycinebetaine and 17 to 57% of the proline. Concentrations of these compounds in vacuole and cytoplasm were calculated and were always higher in the cytoplasm than in the vacuole. The concentration gradient across the tonoplast varied considerably. The significance of these results is discussed in relation to the hypothesis that glycinebetaine and proline function as benign cytoplasmic osmotica.

    Notes: Abstract The effects of hyper- and hypo-saline stresses on the levels of various inorganic and organic solutes inUlva lactuca have been recorded. Hypoosmotic stress decreased the tissue concentration of K+, Na+ and Cl- while hyper-osmotic stress caused a transient increase in Na+ and a stable accumulation of K+ and Cl-. The tissue content of β-dimethylsulphoniopropionate (β-dimethylpropiothetin) responded to changes in salinity. The time course of hypersaline stress showed the β-dimethylsulophoniopropionate concentration rose as the Na+ level fell. The levels of free sugars and amino acids, including proline, were relatively low in this alga and did not appear to be important in osmotic adjustment. The possibility that tertiary sulphonium dipolar ions have an analogous role in some algae to glycinebetaine and possibly other quaternary nitrogen compounds in higher plants as cytoplasmic osmotica is discussed briefly.

    Notes: Summary Advanced high sugar and acid breeding lines of tomatoes (Lycopersion esculentum Mill.) were rated higher in sweetness, sourness and overall flavor intensity than the standard cultivars Cal Ace or T3. Titratable acidity and soluble solids content were major contributors to differences in overall flavor intensity. The results demonstrate that significant improvement in tomato flavor can be attained by increasing sugar and acid contents in tomato fruits by genetic manipulation. Current evidence indicates that breeding for high soluble solids in horticulturally acceptable tomato cultivars is justified.

    Notes: Summary Low temperature germination responses were evaluated for 18 high altitude accessions representing five wild Lycopersicon species and 19 accessions of L. esculentum which have reputed ability to germinate in the cold. Survival analysis indicated that one accession of L. chilense germinates better at 10°C than PI 120256, the fastest-germinating L. esculentum genotype, and that PI 120256 germinates as well as PI 126435 (L. peruvianum). Additional wild ecotypes exhibiting rapid germination at 10°C were identified from L. peruvianum and L. hirsutum. These ecotypes may possess genetic potential for introgressing cold germination ability into L. esculentum cultivars.
