    Notes: Summary. Peripheral blood lymphocyte subsets in normal and preeclamptic pregnancies have been studied by using the monoclonal antibodies OKT3 (T cells), OKT4 (helper/inducer T cells) and OKT8 (suppressor/cytotoxic T cells). In addition the numbers of mononuclear cells bearing la and monocyte antigens have been assessed by using the monoclonal antibodies OKIal and OKM1. No significant differences were found between 10 normal pregnant and 10 non-pregnant subjects. Ten preeclamptic patients were studied and showed an increase in OKT4-positive helper cells. This was significant in terms of percentage of mononuclear cells but not the absolute numbers or the helper/suppressor (OKT4/OKT8) ratio.

    Notes: Lymphocytes isolated from human tonsils are capable of lysing cells or the natural killer (NK)-susceptible line K562. Although weak compared with that of autochthonous peripheral lymphocytes, cytotoxicity is invariably manifest at high effector-to-target ratios. Like peripheral NK cells, tonsillar cytotoxic lymphocytes possess a low buoyant density, which makes possible their partial enrichment from non-cytotoxic cells by centrifugation on discontinuous Percoll gradients. However, examination of cytotoxic fractions indicates that, unlike blood lymphocyte-mediated cytotoxicity, effector function is not associated with the presence of large granular lymphocytes. Furthermore, a functional distinction from classical NK cells is apparent since, although tonsillar cytotoxicity is significantly enhanced after exposure to supematants from polyclonally activated allogeneic tonsils, pretreatment with lymphobtastoid (Namalva) interferon (IFN-α) at doses shown to potentiate maximally the cytotoxicity of peripheral blood lymphocytes fails to influence reactivity. These data provide preliminary evidence for the existence of, at least limited, heterogeneity among human NK cells.

    Notes: The influence of type I interferon (1FN) on leucocyte-mediated antibody-dependent cellular cytotoxicity (ADCC) has been examined in a system in which differential alloantibody sensitization of human erythrocyte target cells allows discrimination between lymphocyte and monocyte effector function. ADCC mediated by unfractionated mononuclear leucocyte populations was regularly enhanced by interferon pretreatment over a range of concentrations of the sensitizing antibody. The capacity for IFN-augmented reactivity was, however, removed with adherent cell depletion, suggesting that lymphocyte (K-cell)effectors are not modulated by IFN, even though such population demonstrate IFN-potentiated natural killer (NK) reactivity against K562. These results suggest that monocyte and NK cell function but not K-cell activity arc influenced by interferon.

    Notes: Diffraction contrast phenomena on X-ray topographs taken with continuous-spectrum synchrotron radiation have been studied at wavelengths of 0.057, 0.064, 0.071, 0.100, 0.154, 0.206 and 0.250 nm. The specimen was a polished plate of natural diamond with surfaces parallel to (110), ½ mm thick. Using the {\bar 1}11 reflection and a stored electron beam energy of 1.8 GeV all topographs (except that taken with λ = 0.25 nm) were harmonic free. The specimen exhibited mixed-habit growth, containing sectors of normal faceted {111} growth and sectors of non-faceted `cuboid' growth in which growth-surface orientation was variable and only approximately parallel to {100}. Prior to X-ray topography the specimen had received localized damage from implantation with fluorine ions of 17 MeV energy. Features whose variation with wavelength was studied included (1) the relative strengths of integrated reflections from {111} and `cuboid' growth sectors, (2) the intensity of `spike' disorder diffuse reflections relative to sharp Bragg reflections, (3) contrast from inclusions, polishing striae and fracture damage and (4) lattice bending and diffraction contrast at the sites of fluorine ion implantation. Theoretical predictions of the wavelength variation of the intensity of the diffuse reflection images and of contrast due to resolved defects showed good agreement with the observations.

    Notes: Abstract Components of a xenobiotic detoxication/toxication system involving mixed function oxygenases are present inMytilus edulis. Our paper critically reviews the recent literature on this topic which reported the apparent absence of such a system in bivalve molluscs and attempts to reconcile this viewpoint with our own findings on NADPH neotetrazolium reductase, glucose-6-phosphate dehydrogenase, aldrin epoxidation and other reports of the presence of mixed function oxygenases. New experimental data are presented which indicate that some elements of the detoxication/toxication system inM. edulis can be induced by aromatic hydrocarbons derived from crude oil. This includes a brief review of the results of long-term experiments in which mussels were exposed to low concentrations of the water accommodated fraction of North Sea crude oil (7.7–68 µg 1−1) in which general stress responses such as reduced physiological scope for growth, cytotoxic damage to lysosomal integrity and cellular damage are considered as characteristics of the general stress syndrome induced by the toxic action of the xenobiotics. In addition, induction in the blood cells of microsomal NADPH neotetrazolium reductase (associated with mixed function oxygenases) and the NADPH generating enzyme glucose-6-phosphate dehydrogenase are considered to be specific biological responses to the presence of aromatic hydrocarbons. The consequences of this detoxication/toxication system forMytilus edulis are discussed in terms of the formation of toxic electrophilic intermediate metabolites which are highly reactive and can combine with DNA, RNA and proteins with subsequent damage to these cellular constituents. Implications for neoplasms associated with the blood cells are also discussed. Finally, in view of the increased use of mussel species in pollutant monitoring programmes, the induction phenomenon which is associated with microsomal enzymes in the blood cells is considered as a possible tool for the detection of the biological effects of environmental contamination by low concentrations of certain groups of organic xenobiotics.

    Notes: Abstract Individuals of Mytilus edulis L., collected from the Erme estuary (S.W. England) in 1978, were exposed to low concentrations (7 to 68 μg l-1) of the water-accommodated fraction (WAF) of North Sea crude oil. The pattern of accumulation of petroleum hydrocarbons in the body tissues was affected by the presence of algal food cells, the period of exposure, the hydrocarbon concentration in seawater, the type of body tissue and the nature of the hydrocarbon. Many physiological responses (e.g. rates of oxygen consumption, feeding, excretion, and scope for growth), cellular responses (e.g. lysosomal latency and digestive cell size) and biochemical responses (e.g. specific activities of several enzymes) were significantly altered by short-term (4 wk) and/or long-term (5 mo) exposure to WAF. Stress indices such as scope for growth and lysosomal latency were negatively correlated with tissue aromatic hydrocarbons.

    Notes: Summary Lymphocytes isolated from axillary nodes draining breast carcinoma possess variable natural cytotoxic capacity. Augmentation of lymph node cell (LNC) cytotoxicity by interferon (IFN) is also variable, with only some populations displaying potentiated lysis following exposure to either IFN-α or gene-cloned IFN-α2. Where present the IFN-induced augmentation of LNC cytotoxicity was invariably weaker than that observed following similar treatment of autochthonous peripheral blood mononuclear cells (PBMC). Irrespective of their responsiveness to IFN the cytotoxic activity of all LNC preparations examined was significantly increased following pre-incubation with either staphylococcal enterotoxin A (SEA) or factors elaborated by lectin-pulsed allogeneic LNC. The induction or amplification of LNC-mediated natural cytotoxicity by lymphokines may provide a local potentiation of natural immune function at the host : tumour interface.
