    Notes: The pathway of bacteriophage λ head formation has been well characterized in vivo and in vitro (Hohn, Katsura & Hohn, 1977). First a petit λ particle consisting of a shell protein pE, a protein pNu3 and minor proteins, but lacking DNA, is formed. The pNu3-containing structure is then converted to a structure which does not contain pNu3, and which is capable of taking up and cutting the viral DNA. The purpose of this work is to determine the scaffold structure, formed by pNu3, by small-angle scattering techniques based on the known physico-chemical properties of pNu3-containing and pNu3-lacking particles (Künzler & Hohn, 1978). The scaffold is found to be inside the protein shell but less concentrated than the 130 Å radius core reported from electron micrographs.

Membrane and vesicle fractions of skeletal muscle contain like those of cardiac muscle a Na+ + K+ activated ATPase which is inhibited by k-strophanthine. 2. Contrary to the cardiac muscle there is a maximum of activity of the Na+ + K+ stimulated ATPase in the membrane fraction of the skeletal muscle not only at high Na+ and low K+ concentrations, but also at the reversal of the ion relations, that is at high K+ and low Na+ concentrations. 3. Investigations of the effectiveness of strophanthine on the membrane fractions of cardiac and skeletal muscle of rabbits, guinea-pigs and rats resulted in a significant species specifity in sensitiveness: a half maximal inhibition of the Na+ + K+ activated ATPase of the heart or skeletal muscle was found in rabbits at 0,7 or 700 μM/l, in guinea-pigs at 1,0 or 500 μM/l and in rats at 50 or 300 μM/l respectively. The Na+ and K+ activated membrane ATPase of the skeletal muscle is accordingly in rabbits 1000 times, in guinea-pigs 500 times but in rats only 6 times less sensitive than that of the cardiac muscle. The Na+ and K+ activated membrane ATPase of the rat hearts is 50–70 times less sensitive to strophanthine than that of guinea-pig or rabbit hearts. 4. The sensitiveness of the vesicle fraction of heart- and skeletal muscle of the rabbit to strophanthine corresponds to that of the membrane fraction. Addition of sodiumacide diminishes the sensitiveness of the Na+ and K+ activated vesicle fraction and results in an activation by low strophanthine concentrations (10−6–10−8).
    Notes: Zusammenfassung 1. Membran- und Vesikelfraktionen des Skeletmuskels enthalten ebenso wie die des Herzmuskels eine durch Na+ + K+-aktivierbare und durch k-Strophanthin hemmbare ATPase. 2. Im Gegensatz zum Herzmuskel findet sich in Membranfraktionen des Skeletmuskels eine maximale Aktivität der Na+ + K+-stimulierten ATPase nicht nur bei hohen Na+- und niedrigen K+-Konzentrationen, sondern auch bei der Umkehr des Ionenverhältnisses, d. h. bei hohen K+- und niedrigen Na+-Konzentrationen. 3. Untersuchungen über die Wirksamkeit des Strophanthins an Membranfraktionen des Herz- und Skeletmuskels von Kaninchen, Meerschweinchen und Ratten ergaben deutliche artspezifische Empfindlichkeitsunterschiede: Eine halbmaximale Hemmung der Na+ + K+-aktivierten ATPase des Herz- bzw. Skeletmuskels fand sich an Kaninchen bei 0,7 bzw. 700 μM/l, an Meerschweinchen bei 1,0 bzw. 500 μM/l und an Ratten bei 50 bzw. 300 μM/l. Die Na+ + K+-aktivierte Membran-ATPase des Skeletmuskels ist demnach bei Kaninchen 1000 mal, bei Meerschweinchen 500 mal und bei Ratten sechsmal unempfindlicher als diejenige des Herzmuskels. Die Na+ + K+-aktivierte Membran-ATPase des Rattenherzens ist gegen k-Strophanthin 50–70 mal unempfindlicher als die des Meerschweinchen-bzw. Kaninchenherzens. 4. Die Empfindlichkeit der Vesikelfraktion des Herz- und Skeletmuskels des Kaninchens gegenüber Strophanthin entspricht derjenigen der Membranfraktion. Der Zusatz von Na-Azid vermindert die Empfindlichkeit der Na+ + K+-aktivierten Vesikel-ATPase des Skeletmuskels gegen Strophanthin und hat zur Folge, daß niedrige Strophanthinkonzentrationen (10−6–10−8) eine Aktivierung verursachen.

    Description / Table of Contents: Zusammenfassung An einem auslesefreien Material von 275 Basaliomexcidaten wurde die Neubildung elastischer Fasern im Basaliomstroma mit Hilfe der Elastica-Färbungen (Orcein, Resorcin-Fuchsin) histologisch untersucht. In nahezu einem Drittel der Basaliome konnten neugebildete elastische Fasern im Stroma beobachtet werden. An Stellen, an denen eine Neubildung elastischer Fasern nachweisbar war, fanden sich ungewöhnlich zahlreich Fibroblasten und Mastzellen (Toluidin-Blaufärbung). Offensichtlich kommt es unter dem Einfluß des auswachsenden Basaliomstranges im Sinne einer dermo-epidermalen Interaktion 1. zu Mastzellvermehrung 2. zur Fibroblastenstimulierung 3. zur Neubildung elastischer Fasern. Die Beobachtungen legen den Schluß nahe, daß zwischen den genannten Befunden eine Wechselwirkung besteht. Als Folge der dermo-epidermalen Interaktion können die Fibroblasten (auch der Altershaut!) im Basaliomstroma so weit stimuliert werden, daß sie erneut die Fähigkeit erlangen, elastische Fasern zu bilden — eine Funktion, die sonst üblicherweise mit zunehmendem Alter weitgehend eingeschränkt ist. Die Befunde scheinen von Bedeutung unter dem Aspekt der “Verjüngung” menschlicher Altershaut — sie sind bisher jedoch nur von rein theoretischem Interesse.
    Notes: Summary From 275 specimens of all sorts of basal cell epitheliomas the new formation of elastic fibers was histologically investigated by means of selective elastic tissue stains such as orcein and resorcinfuchsin. In nearly one third of the basal cell epitheliomas newly formed elastic fibers could be observed in the stroma. In places where a new formation of elastic fibers was demonstrated fibroblasts and mast cells (toluidine blue stain) were unusually numerous. Apparently the influence of the growing tumor masses of basal cell epitheliomas leads to a dermo-epidermal interaction in the form of 1. increase of mast cells 2. stimulation of fibroblasts 3. new formation of elastic fibers. The observation suggest that there exists a relationship between the above noted findings. Due to the dermo-epithelial interaction (even in senile skin) in the stroma of basal cell epithelioma fibroblasts can be stimulated to such a degree that they regain the ability to produce elastic fibers—a function which usually is greatly diminished with increasing age. The findings seem to be important from the view point of rejuvenation of aging human skin.
