    Description / Table of Contents: Zusammenfassung Durch Wiederfindungsmessungen mit Hilfe von i.p. injiziertem3H-Digoxin und125-markiertem Digoxin konnte gezeigt werden, daß Standardkonzentrationen von Digoxin, die arzneistoffreiem Gewebe zugesetzt werden, zu falsch negativen Bestimmungen führen können. Der Vergleich der Radioaktivität aus verschiedenen Geweben vor und nach Extraktion mit Amberlite XAD-2 ergab, daß nur um 50% der gesamten Glykosidmenge extrahiert wird: BD9-(Berlin)-Ratten wurden 15mg/kg Körpergewicht β-Methyldigoxin i.p. injiziert. Aus Extrakten mit Amberlite XAD-2 von Blut, Niere, Herz und Leber konnten außer β-Methyldigoxin dessen Metaboliten Digoxin und Digoxinbidigitoxose ab 60 ng/Fleck in prozentualen Anteilen als Fluoreszenzflecke sichtbar identifiziert werden, durch die Anwendung von HPTLC- Platten mit Fluoreszenzzusatz. Radioimmunologisch bestimmte Konzentrationen fanden sich in guter Übereinstimmung mit dünnschichtchromatographischen Ergebnissen.
    Notes: Summary The recovery measurements in rat tissues performed via i.p. injected radioactive digoxin derivates (3H-digoxin,125-digoxin derivative) showed that approximately 50% of the total glycoside content will be extracted. Thus, an addition of digoxin standards to drug-free tissues may lead to false negative determinations. By comparison of the radioactivity before and after extraction the following results were obtained: After i.p. application of 15 mg/kg of β-methyldigoxin to BD 9 (Berlin)-rats the resulting tissue concentrations were extracted by Amberlite XAD-2. β- Methyldigoxin and its metabolites digoxin and digoxinbisdigitoxide could be separated and distinguished from artifacts by fluorescence detection on HPTLC-plates with a detection limit of 60 ng/spot. Concentrations determined by radioimmunoassay are in satisfactory agreement with HPTLC results.

    Notes: Summary During the clinical course of a secondary plasma cell leukemia the cellular protein synthesis and immunoglobulin synthesis were measured in a cell-free system prepared from peripheral plasma cells. After treatment with cyclophosphamide and a combination of cyclophosphamide with melphalan and vincristine no change in the activity of total protein synthesis was found. In contrast, a decrease of total protein synthesis could be observed after treatment with BCNU combined with cyclophosphamide and Ara-C. The relative amount of 10–15% Ig-synthesis remained unchanged under the different chemotherapeutic regimes. These findings suggest a primary effect of BCNU on cellular protein synthesis in myeloma cells similar to that reported in HeLa cell cultures.

    Notes: Abstract The experiments presented here deal with the effects of the inhalation of dichlorvos [dimethyl-(2,2 dichlorvinyl)-phosphate, DDVP] vapor on acetyl-cholinesterase (ACHE) activity in rat bronchial tissue. Exposure to DDVP concentrations of 0.8 and 1.8 μg/l for 3 days reduced ACHE activity in the bronchial tissue (62.8±0.8 and 51.6±1.6% of the control), but did not elicit any changes in blood ACHE activity (101±4.5% of the control each). Higher concentrations (4.3 μg/l) induced a decline in ACHE activity also in the blood (38.2±1.1% of the control). In the histochemical preparations used to demonstrate CHE activity in bronchial tissue (thiolacetic acid method), a staining of the bronchial glands and smooth muscles characteristic of the enzyme activity was strongly reduced after exposure of the animals to even the lowest dose applied (0.2 μg/l). The question of whether localized inhibition of ACHE in the bronchial tissue might cause increases in airway resistance due to activation of a broncho-bronchial reflex is discussed. This efferent cholinergic mechanism has been found to be at least partly responsible for maintenance of bronchospasm and hypersecretion in chronic obstructive deseases of the respiratory system.
