    Notes: Rat's tongues were injured by cold and the reactive processes were observed by light and electron microscopy. Injured animals and untreated controls were injected with aurothiomalate and the cells in tongue muscles which contained gold were observed after increasing injection-sacrifice time intervals.In the very rapid regeneration, gold-containing macrophage-like cells fused with each other and with well preserved parts of myofibers. The selectivity of gold localization suggested, at least morphologically, a macrophage to myoblast development. The possible factors affecting the rate of muscle regeneration have been discussed with special reference to the nature of the injurious stimulus and the type of cellular response.

    Notes: Summary Mast cells from rat's tongue muscles, fixed with glutaraldehyde only, were examined with an electron microscopic microanalyzer — the EMMA-4. With the preparatory method used, high emissions of a number of elements were obtained in various intracellular spots. The granules of mast cells were found to contain strictly localized high concentrations of sulphur, calcium and zinc.

    Notes: Summary The effect of various steps in the preparation of muscle and blood cells for electron microscopic X-ray microanalysis, was studied with reference to calcium, zinc and copper levels. It was found that these elements are readily detectable in material fixed in glutaraldehyde and embedded directly in Epon 812. Alcohol dehydration, buffer rinses and post-osmication caused marked depletion of all elements examined.

    Description / Table of Contents: Zusammenfassung Die lokale Konzentration von Ca, P, Cl, S und Zn in ultradünnen Myokardschnitten wurden durch Röntgenmikroanalyse untersucht. Hamster mit Kardiomyopathie wurden mit einem gesunden Stamm gleichen Alters verglichen. Es wurde ein signifikanter Anstieg der Ca- und Cl-Konzentration bei den kranken Tieren gefunden, am ausgeprägtesten im hypertrophischen Stadium der Erkrankung. Die Konzentrationen von P und Zn waren unterschiedlicher. Es ergab sich, daß das Verhältnis zwischen den Elementen (besonders P/Ca) innerhalb der gleichen Zelle und das Verhältnis der gleichen Elemente innerhalb verschiedener Zelltypen quantitative Kriterien für die Bestimmung des Ausmaßes pathologischer Veränderungen bietet.
    Notes: Summary Localized elemental concentrations of Ca, P, Cl, S, and Zn were detected by X-ray microanalysis during viewing of ultrathin myocardial sections. Cardiomyopathic hamsters were compared with a healthy strain of the same age. Significant increases in calcium and chlorine concentrations were found in sick animals. These were highest in the hypertrophic stage of the disease. Phosphorus and zinc were more variable. Ratios between elements (especially P/Ca) in the same cells and of the same elements in different cell types were found to offer numerical criteria for estimating the degree of pathological change.

Nach Isoproterenolperfusion konnten Veränderungen der P-, Ca- und Zn-Konzentrationen und eine Beziehung dieser Änderungen zu hämodynamischen und morphologischen Parametern festgestellt werden. Das P/Ca-Verhältnis in den Kernen der Muskelfasern und die Calcium-Relation zwischen den Kernen der interstitiellen Zellen einerseits und der Myokardfasern andererseits zeigten eine direkte Beziehung zum Zustand des Herzens.
    Notes: Summary X-ray microanalysis was carried out on sections from isolated hearts perfused with a modifiedKrebs-Henseleit solution with or without isoproterenol. Changes in P, Ca and Zn concentrations could be detected after isoproterenol and some correlation with hemodynamic and morphological parameters was observed. Ratios between P/Ca in myonuclei and the ratios of interstitial cell to myofiber nucleus calcium were related directly to the state of the heart.

    Description / Table of Contents: Zusammenfassung Nach operativer Freilegung wurden beim Herzen von Ratten durch direkte Vereisung eine örtliche Schädigung gesetzt. Die Herzen wurden 2, 4, 20 und 48 Stunden sowie 7 und 12 Tage später untersucht. In den ersten 48 Stunden trat eine Infiltration mit Mastzellen und polymorphkernigen Granulozyten (hauptsächlich Eosinophilen) auf, gefolgt von einer Proliferation mononuklearer Phagozyten, fibroplastischen Zellen und neuer Kapillaren. Die benachbarten Muskelpartien zeigten eine Schädigung im Sinne eines Einfrier-Auftau-Rigors, segmentaler Muskelfaser-Nekrose, Phagozytose und fokaler Regeneration. Nach 7 Tagen erschienen zahlreiche Myofibroblasten, die enge Beziehungen zeigten zu Muskelfasern, die der Degeneration und Regeneration unterlagen. Nach 12 Tagen war eine verstärkte Bindegewebsbildung zu verzeichnen mit mesothelialen Deckzellen des Epikardiums, die Muskelfasern normalisierten sich. Es wird angenommen, daß Amine, die von den Mastzellen freigesetzt werden, an der Bildung der Myofibroblasten beteiligt sind.
    Notes: Summary Rat hearts were exposed and injured locally by direct cryoapplication. The hearts were examined 2, 4, 20 and 48 hours, 7 and 12 days later. In the first 48 hours mast cell and polymorphonuclear (mainly eosinophilic) granulocyte infiltration was followed by proliferation of mononuclear phagocytic and fibroblastic cells and new capillaries. The underlying muscle showed freezethaw-rigor damage, segmental intramyofiber necrosis, phagocytosis and focal regeneration. After 7 days numerous myofibroblasts appeared and they were intimately related to myofibers undergoing degeneration and repair. After 12 days there was increased fibrosis while mesothelial covering cells of the epicardium and myofibers returned to normal. It is suggested that amines released from mast cells are involved in the formation of myofibroblasts.
