    Notes: Summary The demonstration of glycogen by PAS-method and the reaction of phosphorylase by Takeuchi et al. were performed in the hearts of rats 30 min after experimental coronary occlusion. While the cardiac muscle cells in the center of the ischemic region still contained glycogen, the cells in the border zone of the infarction did not. The phosphorylase activity was demonstrable only in those cardiac muscle cells which contained glycogen. As well known that the demonstration of phosphorylase activity is possible only if glycogen as starter is present, it is concluded that the lack of phosphorylase activity by the histochemical reaction in cells which do not contain glycogen doesn't prove the absence of the enzyme.

    Description / Table of Contents: Summary By means of the dynamic elastometer ofkrug, it could be shown that the white matter of 31 brains from diabetic patients was significantly firmer than that of 60 control brains. The gray matter of 28 brains from uremic patients revealed a significant reduction in firmness. In the diabetic patients a close relationship existed between the individual values for the rigidity of the brain and the blood sugar values. Only a slight reduction of the dry substance was found in the brains from diabetic patients. Many factors play a role in the pathogenesis of changes in diabetes, such as disturbances of cellular and capillary membranes, and changes of the colloidal state by polymerization of the substances in the white matter. Here either increased amounts of polymerizable substances are deposited and polymerized by catalyzers within the brain, or polymerizable substances of the brain are polymerized by pathologic products arising out of the intermediate metabolism.
    Notes: Zusammenfassung Gegenüber 60 Vergleichsfällen konnte mittels des dynamischen Elastometers nachKrug bei31 Diabetikergehirnen bestätigt werden, daß die weiße Substanz signifikant fester ist. Die graue Substanz von 28 Urämiegehirnen zeigte eine signifikante Verringerung der Starre. Zwischen den Einzelwerten der Starre und dem Blutzucker Hegt bei den Diabetesfällen eine straffe Korrelation vor. Eine Verminderung der Trockensubstanz fand sich zwar bei den Diabetikergehirnen, aber nur mit geringer Signifikanz. Bei der Pathogenese der Veränderungen beim Diabetes spielen wohl viele Faktoren eine Rolle, wie Störungen an der Zell- und Capillarmembran, sowie Änderungen des kolloidalen Zustandes durch Polymerisation von Stoffen im Mark, wobei entweder polymerisationsfähige Stoffe vermehrt abgelagert und durch hirneigene Katalysatoren polymerisiert oder hirneigene polymerisationsfähige Stoffe durch pathologische Produkte aus dem Zwischenstoffwechsel polymerisiert werden.

    Description / Table of Contents: Summary A simple method is described for the early detection of the myocardial infarct, in which the hydrogen ion concentration of sections of the heart prepared by the cryostat are measured with pH indicator paper. An ischemie region in the myocardium of rats and cats, produced by ligating a coronary artery, may be demonstrated within five minutes after the ligation. The change in the hydrogen ion concentration in a necrosis, as described in the literature, appears also in the myocardial infarct and serves as proof of it until occurence of light-microscopically recognizable signs of necrosis.
    Notes: Zusammenfassung Es wird eine einfache Methode zum Frühnachweis des Herzinfarktes beschrieben. Durch Messung der Wasserstoffionenkonzentration an Kryostatschnitten vom Herzen mit pH-Indicatorpapier wird ein bei Ratten und Katzen erzeugter Ischämiebezirk bereits 5 min nach Unterbindung einer Coronararterie im Herzmuskel nachweisbar. Die in der Literatur beschriebene Änderung der Wasserstoffionenkonzentration in einer Nekrose wird für den Herzinfarkt bestätigt und dient zu seinem Nachweis bis zum Auftreten von lichtmikroskopisch erkennbaren Nekrosezeichen.

    Description / Table of Contents: Zusammenfassung Bei 3 Patienten, die unmittelbar nach herzchirurgischen Eingriffen verstarben, konnte durch Bestimmung der Wasserstoffionenkonzentration [H+] mit Indicatorpapier am Gefrierschnitt des Herzens 1–2 Std nach dem Tod jeweils eine umschriebene ischämische Schädigung nachgewiesen und das Alter des frischen Herzinfarktes anhand der veränderten [H+] festgestellt werden. Innerhalb der ersten 1–2 Std nach Beginn der Herzmuskelischämie war die [H+] in dem ischämisch geschädigten Bereich erhöht (pH〈6.0). Anschließend wies der Infarktbereich, im Randbereich beginnend, eine erniedrigte Wasserstoffionenkonzentration (pH ∼ 7.4–7.5) auf. Im Zusammenhang mit der erniedrigten [H+] ließ sich mit der Perjodsäure Schiff-(PAS-) Reaktion sogenanntes PAS-positives diastaseresistentes Material im Myokard nachweisen. Beim enzymhistochemischen Succinodehydrogenasenachweis fand sich in einem 7–8 Std alten Infarkt eine verstärkte Enzymreaktion.
    Notes: Summary In three patients who died immediately after surgical procedures on the heart it was possible one—two hours after death to demonstrate with indicator paper on frozen sections of myocardium a circumscribed ischemic lesion and to determine the age of the infarction from the changed hydrogen ion concentration. Within the first one—two hours after the onset of ischemia the hydrogen ion concentration in the area of ischemic damage is increased (pH〈6.0). Subsequently the infarcted area, beginning in the border zone, shows a decreased hydrogen ion concentration (pH appr. 7.4–7.5). As to the decreased hydrogen ion concentration, it was possible with the periodic-acid-Schiff (PAS) reaction to demonstrate so-called PAS-positive diastase-resistant material in the myocardium. The enzymhistochemical succinodehydrogenase test showed an increased enzyme reaction in an infarction which was 7–8 hours old.
