    Notes: An azurin, a small respiratory copper protein from the bacterium Pseudomonas fluorescens biotype G, has been studied by mass spectrometry to determine sequence information. The study of homologously related proteins by mass spectiometry is particularly attractive, since the correct nature of major parts of the deduced sequences can be confirmed by comparison with the sequences of the protein from related organisms. An oxidized tryptohan residue has been identified amongst the products from a cyanogen bromide digest of this wild type azurin. In the same digest, a product is also found to arise from cleavage of the peptide chain at the C-terminal side of the same tryptophan residue. These results are rationalized.

    Notes: The effect of changes in crystallinity on the β-transition in poly(vinyl chloride) was studied by dynamic mechanical measurements of loss tangent (tan δ) and storage modulus (E′) over the temperature range -160° to 85°C. Four frequencies were covered, 3.5, 11, 35, and 110 Hz. The data presented demonstrate that crystallinity has a significant influence on the relaxation processes involved in the β-transition. The restrictions on segmental mobility imposed by crystallinity are most pronounced in the region between Tβ and Tg, with Tβ tending to shift to lower temperatures, Tg shifting to slightly higher temperatures, the magnitude of tan δ decreasing, and engineering tensile strength increasing. Our data also show that individual PVC resins have mechanical and physical properties which differ enough so that comparisons which are often made between them may be inconclusive. Moreover, the properties of one resin may be more sensitive to thermal treatment than those of another. We are continuing this study to establish correlations between the observed trends and other important mechanical properties such as impact strength, toughness, tensile, and compressive strength.

    Notes: A vacuome (‘Golgi apparatus’) consisting of small globular inclusions has been demonstrated in Chlamydomonas sp. These inclusions may be seen in the living, unstained organism; they are stainable vitally with neutral red; they have been stained vitally with neutral red and then blackened with osmic vapor under direct observation, and they have been impregnated by osmic and silver methods without previous treatment with neutral red.The reaction of these inclusions to the iodin test for starch suggests that they may play some rǒle, possibly one of storage, in the cycle of starch metabolism.

    Notes: Viscoelastic properties of the BBL ladder polymer were determined by a dynamic mechanical technique for the temperature range -150°C to 250°C and at frequencies of 3.5, 11, and 110 Hz. A secondary loss dispersion centered at -50°C (at 110 Hz) was discovered which probably arises from an in-chain relaxation mechanism. The loss dispersion resembles β-transitions found in more conventional polymers in intensity (maximum tan δ = 0.035), activation energy( E = 19,000 cal/mole), and in its close relation to the high-temperature mechanical properties of the polymers. Only slight changes in the low-temperature loss peak cause significant increases in brittleness. It is proposed that such changes, produced by a variety of environments, result from random chain scission at imperfection points in the ladder structure. For this reason we conclude that improving the environmental stability of BBL is of prime importance in its further development for specific applications.

    Notes: The fine structure of corpora lutea from 14 white-tailed deer was studied from early through near-term pregnancy. The corpora contained both thecal and granulosal lutein cells. The small, elongate and cylindrical thecal lutein cells contained many lipid droplets, juxtanuclear Golgi elements, abundant agranular endoplasmic reticulum, and other cytoplasmic organelles. These cells were observed throughout pregnancy. Some of the thecal lutein cells became modified during early to midpregnancy. The modified thecal lutein cells possessed many lipid droplets, several lysosomes, packets of PAS-positive glycogen granules, and numerous small membranous whorls of agranular endoplasmic reticulum. The granulosal lutein cells had highly folded and/or ruffled plasma membranes, abundant tortuous tubular and cisternal agranular endoplasmic reticulum, numerous rod-shaped to round mitochondria with tubular and lamellar cristae, Golgi elements, and a few packets of granular endoplasmic reticulum. Many granulosal lutein cells became modified during mid- to near-term pregnancy by the addition of numerous small membrane-bound osmiophilic droplets and variable numbers of large nonmembrane-bound lipid droplets. The granulosal and thecal lutein cells appeared more active in steroid biosynthesis than the modified granulosal and thecal lutein cells.

    Notes: Serum iron transfer from mother to fetus was measured in near-term pregnant females from 16 species representing 15 families and 7 orders of eutherian mammals. The amount of transfer at 2 hr was 5% or greater in species with hemochorial placentation and less than 0.1% at 3 hr in species with epitheliochorial or endotheliochorial placentation.

    Notes: In Trichamoeba sp two types of inclusions are recognized on the basis of reaction to vital dyes and tomethods of osmic and silver impregnation. Globular inclusions, which are stained selectively with neutral red, may be blackened under direct observation by exposure to osmic vapor in hanging-drop preparations and demonstrated by osmic and silver impregnation. Rod-like and granular mitochondria, stainable vitally with Janus green, may be distinguished from the neutral-red globules in preparations stained with a mixture of Janus green and neutral red, and are demonstrated by Regaud's chondriosome method.

    Notes: Acetylacetone (Hacac) admitted as a gas to an ion source containing CoCl2 adsorbed on a field ionization/field desorption emitter produces the ion [Co(acac)Cl]+. Similar ions arising from interaction of adsorbed salt and a ligand admitted as a gas were found for CoCl2 and trifluoroacetyl acetone, and for FeCl2 and acetylacetone. The chelation process represents a new kind of reaction of chemisorbed species in field ionization mass spectrometry.
