Zusammenfassung Die Beeinflußbarkeit der hyperakuten xenogenen Abstoßungsreaktion (HXAR) in weit entfernt stammesverwandten Tierarten, Schaf— Hund, wurde untersucht. Extreme Vorimmunisierung der Empfänger mit Leberhomogenat vom Spender, zusammen mit Freundschem Adjuvans, verlängerte bei E vivo-Perfusion die Überlebenszeit der Schafnieren und verbesserte den renalen Blutfluß (RBF) geringfügig. Eine 12monatige Vorimmunisierung der Empfänger führte zu einer Provozierung von komplementbindenden und hämagglutinierenden Antikörpern, die die Werte der präformierten Antikörper dieser Klassen weit überstiegen. Die Wirkung dieser Antikörper auf die Oberflächenantigene der Nierenzellen kommt zum Ausdruck im Freiwerden von Zellenzymen, intracellulärem Kalium und einer Proteinurie in Kontroll- und Versuchsgruppen. Der verzögerte Abfall des RBF, die in Gang kommende Urinproduktion und die anfangs schwächere Ausschwemmung von Enzymen und Kalium in den Nieren, die durch immunisierte Tiere perfundiert wurden, läßt vermuten, daß xenogene „enhancing“-Antikörper in der Lage sind, wenigstens anfänglich die Zelloberflächenantigene zu blockieren und so vor dem Einwirken cytotoxischer Antikörper zu schützen. Übertragung von Hyperimmunserum auf unbehandelte Hunde führte zu gleichen Ergebnissen und stützt die Annahme, daß antigenmaskierende Antikörper die Ursache der Verlängerung der Überlebenszeit der Transplantate sein dürften.

Summary The possibility to modify the hyeracute xenogeneic rejection (HXR) in a system of widely divergent species, sheep—dog, was investigated. Pretreatment of the recipients over a 12month period with high amounts of liver homogenate from the donor species together with Freund's adjuvant, slightly prolonged the survival time of the xenogeneic e-vivo perfused kidneys and increased the renal blood flow (RBF). High amounts of complement-fixing and hemagglutinating antibodies, that exceeded the titers of preformed antibodies of these classes significantly, are due to the immunisation of the recipients. The action of these antibodies on the cell-surface antigens seems to be reflected in the release of cellular enzymes, and intracellular potassium and in proteinuria of control animals and treated animals. The delayed decrease of enzymes and potassium the kidneys, perfused by immunized animals, lead to the assumption, that xenogeneic enhancing antibodies are able to block the cell surface antigens initially and so protect them against cytotoxic antibodies. Transferring hyperimmunsera to untreated dogs reproduced similar effects. This supports the idea, that prolongation of the survival time of the kidneys is due to the antigen masking effect of enhancing antibodies.
