    Notes: Summary It has been demonstrated by injections of H3-Thymidine into adult and juvenile snails (Helix pomatia) that the mitotic activity in the central nervous system, the connective tissue and epidermis was generally higher in juvenile animals than in adult ones. Data are given for the amount of triated thymidine labelled nuclei in the tissues, which are investigated.

    Description / Table of Contents: Zusammenfassung Da Milch aus sekretionsgestörten Eutern in ihrer chemischen Zusammensetzung gegenüber Milch aus gesunden Eutern stark verändert sein kann, wurde der Einfluß von Euterentrzündungen auf die Aktivität der Lactoperoxidase untersucht. Dazu wurde in über 100 Viertelgemelksproben von Kühen in normaler Lactation mit positivem Schalmtestbefund, der durch exakte elektronische Zellzählung abgesichert wurde, die Enzymaktivität gemessen. Die statistische Auswertung der Ergebnisse ergab eine mit 95,0% gesicherte positive Korrelation zwischen Zellgehalt und Enzymaktivität, die mitr = 0,24+ jedoch relativ niedrig ist, so daß sich der Peroxidase-Test zwar nicht zum Nachweis von Milch mit erhöhtem Zellgehalt eignet, jedoch als wertvolles Indiz in der Rechtssprechung dienen kann.
    Notes: Summary Since milk of disturbed udders shows a chemical composition different from normal milk, the influence of raised leucocyte counts on the activity of Lactoperoxidase was investigated. The enzyme activity in milk of more than 100 individual udder quarters was determined, the cows being in normal lactation but having positive CMT tests, affirmed by electronic cell determination. There was a significant positive correlation between cell count and enzyme activity of r = + 0,24 (p 95%), which is relatively low. Therefore the peroxidase test is not suitable for detection of milk with high cell counts, however, it can be used for legislative reasons.

    Notes: Summary Dilution and incubation of two batches of β-acetyldigoxin tablets (Novodigal®) in a weakly alkaline solution at 37° C yields amounts of α-acetyldigoxin increasing with time. Thus, within 60 min the percentage of α-acetyldigoxin was approximately 60 and 70% for lot A and lot B, respectively. The rate of isomerisation does not control the rate at which acetyldigoxin is absorbed in the intestinal tract.

    Description / Table of Contents: Zusammenfassung Im Binde- und Nervengewebe von Gastropoden, welche in Kunstharze eingebettet wurden, lassen sich nach Entfernung des jeweiligen Einbettungsmediums mit licht- und fluoreszenzmikroskopischen Färbeverfahren (Paraldehydfuchsin, Aldehydthionin, Alcianblau, Pseudoisocyanin, RF 500) mit hoher histochemischer Spezifität Zellen und Strukturen darstellen. Neben verschiedenen Zelltypen mit unterschiedlichen Funktionsstadien im Bindegewebe können vor allem die neurosekretorischen Zellen und ihre Fortsätze in den Cerebralganglien der untersuchten Schneckenarten bis in ihre Neurohämalbereiche differenziert angefärbt werden. Mit AT und PAF konnten dabei neurosekrethaltige Axone in der Cerebralkommissur von Planorbarius corneus erstmals sicher lichtmikroskopisch nachgewiesen werden. Die von uns verwendeten Verfahren erlauben eine gezielte elektronenmikroskopische Analyse der im Semidünnschnitt licht- und fluoreszenzoptisch selektiv histochemisch identifizierten Zellen und Strukturen.
    Notes: Summary In the present study specific cells and structures in the connective-and nervous tissues of some gastropods were demonstrated by light- and fluorescence techniques, performed on semithin sections of plastic embedded material. After dissolving the embedding medium (Epon or Styrene-methacrylate), the semithin sections are stainable by Gomori's Paraldehyde-Fuchsin, Aldehyde-Thionin, Alcian Blue and the fluorochromes Pseudoisocyanin and RF 500. Best results are obtained optically by staining with Pseudoisocyanin, because all fluorescing substances (i. a. the neurosecretory cells and fibres in the ganglia) appear on a practically dark background. For the first time, with Aldehyde-Thionin and Paraldehyde Fuchsin—but not with Pseudoisocyanin—neurosecretory fibres has been demonstrated in the cerebral commissure of Planorbarius corneus. The histochemical identifying of specific cells and structures in semithin sections by light- and especially fluorescence microscopic highly selective reactions gives the possibilities to analyse these target cells by electron microscopic investigations.

    Notes: Summary 3H-digoxigenin-mono-digitoxoside 1 mg was swallowed by 6 healthy subjects. Maximum plasma levels of radioactivity were reached within 1–2 h; in two subjects there was a second peak at 8–12 h. No definite half lives could be determined because the falls in plasma activity were not exponential. 3.9–39% and 34.5–76.6% of the dose were eliminated in urine and faeces, respectively. 75–90% of the total radioactivity in plasma was CHCl3-insoluble, there was less of this fraction in urine, and the major portion in faceces was CHCl3-soluble. The CHCl3-insoluble fraction in urine was separated into 3 components by chromatography on an Al2O3-column and consisted mainly of conjugates of the monoglycoside and 3-epidigoxigenin. TLC-separation of the lipophilic fraction in urine also revealed unchanged monoglycoside and 3-epidigoxigenin, as well as traces of digoxigenin, 3-ketodigoxigenin and 2 unidentified, more polar metabolites. In faeces, the main excretion product was the unchanged compound, and traces of digoxigenin, 3-epidigoxigenin, 3-ketodigoxigenin and one of the more polar metabolites detected in urine. Two patients with surgical T-tube bile-duct drainage showed significantly greater biliary excretion after oral administration of the digoxigenin-mono-digitoxoside than after digoxin. Almost all the radioactivity excreted in bile was CHCl3-insoluble and the monoglycoside was shown to be the only conjugation partner present by incubation with arylsulfatase and β-glucuronidase. The results show that digoxigenin-mono-digitoxoside has such a rapid metabolic inactivation and biliary clearance in man that it is unlikely to be of any therapeutic value.
