Zusammenfassung Nach einmaliger Injektion von 3H-Thymidin wurden die Teilphasen des Generationszyklus von Leberepithelzellen ausgewachsener Ratten nach 2/3 Teilhepatektomie mit Hilfe der „Prozent markierte Mitose-Methode“ geprüft. Die autoradiographischen Untersuchungen erfassen dabei die erste Regenerationswelle. Alle Tiere wurden 38 Std nach der Operation zu unterschiedlichen Tages- und Nachtzeiten getötet. Folgende Ergebnisse zeichnen sich ab: 1. Nach der Teilhepatektomie wird die G 1-Phase für sehr viele Leberepithelien durch ihren Eintritt in die S-Phase vorzeitig unterbrochen und somit beträchtlich abgekürzt. Eine zweite postoperative G 1-Phase geht jedoch nicht ein. Die erste parenchymatöse Regenerationswelle wird somit von einer proliferierenden Zellpopulation getragen, die nur einmal S, G 2 und M passiert. 2. Die mittlere S-Phasenlänge beläuft sich bei einer sehr geringen Variationsbreite auf 7,2 Std. Die minimale Länge von G 2 beträgt etwas weniger als 2, die maximale 4,5–5 Std. Die Mitose beansprucht einen mittleren Zeitraum von 1–1,5 Std. Nur im Einzelfall nimmt die Kernteilung maximal 3–4 Std in Anspruch. Der nur vergleichsweise langsam erfolgende Anstieg von Silberkornzahlen über stark markierten Mitosen ist als Folge einer beträchtlichen Variationsbreite der G 2-Phase zu interpretieren. 3. Sehr frühzeitig treten unter den gegebenen experimentellen Bedingungen auch außerordentlich schwach markierte (um den Faktor 30–40 geringer geschwärzte) Mitosen in einem sehr hohen Prozentsatz auf. In „überexponierten“ Autoradiogrammen steigt dieser Wert noch an und erreicht nahezu 100%. Die Existenz dieser extrem schwach markierten Mitosen spricht nicht allein für eine typische Wiederverwendung, d. h. Reutilisation von bereits in die DNS anderer Zellen eingebauter Metaboliten, sondern weitaus mehr für eine Utilisation 3H-Thymidin-haltigen, noch nicht in die DNS inkorporierten Vorläufers. Dieser „Pool“ 3H-Thymidin-haltigen Materials ist sicherlich im Vergleich zum Vorläuferangebot während der typischen Verfügbarkeitszeit von 3H-Thymidin sehr klein, aber offensichtlich in einem relativ langen Zeitraum wirksam.

Summary After a single injection of 3H-thymidine autoradiographic experiments to determine the different phases from the generation cycle of parenchymal liver cells of rats following a partial hepatectomy (2/3) had been carried out with the “percentage of labeled mitoses method”. The studies are extended on the first wave of regeneration in liver cells. All animals were sacrificed 38 hrs after the operation. Following results had been obtained: 1. After partial hepatectomy the G 1-phase for many parenchymal liver cells is precociously interrupted and shortened as a consequence of their entrance into the S-phase. A second post-operative G 1-phase was not observed. Thus the first wave of parenchymal regeneration consists of a proliferating cell population, which passes S, G 2 and M only once. 2. The medial value for the length of S amounts to 7.2 hrs, and there is only a very small variation in the duration of this phase. The minimal length of G 2 is a little shorter than 2 hrs, the maximal value reaches 4.5–5 hrs. Mitosis takes place within a medial time of 1–1.5 hrs, only very seldom the maximal duration of this phase ranges up to 3–4 hrs. The number of silver grains over intensively labeled mitoses increases relatively slowly as a function of the radioactive time. This must be interpreted as a consequence of a considerable variation in the G 2-phase. 3. Under the experimental conditions investigated also extremely weak labeled mitoses can be observed in a high percentage. The number of silver granules are reduced about the factor 30–40 in comparison with intensively labeled mitoses. In autoradiograms with a very long time of exposition the percentage of such mitoses reaches nearly the 100% value. The appearance of these extremely weak labeled mitoses may have two reasons. Their existence can, by all means, be due to a reutilization of 3H-thymidine of or 3H-thymidine metabolites from the DNA of other cell populations with a rapid proliferation. But it is more likely that these labeled mitoses are due to a delayed utilization of 3H-thymidine containing precursors, which had not yet been incorporated into DNA of other cells. This “pool” of 3H-thymidine must be very small as compared with the offer of this tritiated precursor during typical availability time of 3H-thymidine. But probably this “pool” is effective over a relatively long period.
