Summary In comparison with dialysis and the methods of protein precipitation, gel filtration offers considerable advantages in the removal of salts from protein solutions. The behavior of inorganic ions (milligram- to submicrogram amounts) was studied on Sephadex micro-columns with the aid of radioactive isotopes. Inorganic ions were found to behave diversely depending on the concentration chosen and the swelling- and elution liquid. In 0.9% sodium chloride solution, submicrogram amounts of cations and anions also enter the gel phase; they can be separated quantitatively and reproducibly from the protein, which remains excluded from the gel phase. On the basis of the experimental data furnished, the gel filtration with Sephadex G-25 can be used for the isolation of protein-bounded serum iodine (PBI) from inorganic iodine. In contrast to the previous methods of separation, it is likewise useful for blood serums with extremely augmented iodide content. The saving in time and work is a further advantage.

Résumé La filtration sur gel offre dans le relargage des solutions de protéines des avantages importants, par comparaison avec la dialyse et les méthodes de précipitation des protéines. On a étudié, au moyen d'isotopes radioactifs le comportement des ions minéraux (en quantités de l'ordre du milligramme jusqu'aux quantités submicro) sur microcolonnes Sephadex. D'après cela, les ions minéraux se comportent différemment suivant leur concentration, la solution mère et le liquide d'élution. Dans une solution de chlorure de sodium à 0,9%, on trouve des cations et des anions dans la phase gel en quantités submicro; on peut les séparer quantitativement et de façon reproductible à partir de la protéine, qui reste hors de la phase gel. D'après les résultats expérimentaux communiqués, la filtration sur gel, avec Sephadex G-25 est applicable à l'isolement de l'iode du sérum lié aux protéines (PBI) de l'iode minéral. Cette méthode convient, à l'opposé des méthodes de séparation connues jusqu'ici, également au sérum sanguin à teneur en iode extrêmement élevée. Les gains de temps et de travail obtenus fournissent un avantage supplémentaire.

Zusammenfassung Die Gel-Filtration bietet bei der Entsalzung von Proteinlösungen im Vergleich zu Dialyse und den Methoden der Eiweißfällung erhebliche Vorteile. Mit Hilfe radioaktiver Isotope wurde das Verhalten anorganischer Ionen (Milligramm- bis Submikrogrammengen) an Sephadexmikrosäulen untersucht. Danach verhalten sich anorganische Ionen je nach Wahl ihrer Konzentration und der Quell- und Elutionsflüssigkeit unterschiedlich. In 0,9%iger Natriumchloridlösung treten auch Submikrogrammengen von Kationen und Anionen in die Gel-Phase ein; sie sind quantitativ und reproduzierbar vom Eiweiß, das von der Gel-Phase ausgeschlossen bleibt, abtrennbar. Auf Grund der mitgeteilten Versuchsergebnisse ist die Gel-Filtration mit Sephadex G-25 auf die Isolierung des proteingebundenen Serumjods (PBI) von anorganischem Jod anwendbar. Sie bewährt sich im Gegensatz zu den bisher bekannten Trennmethoden auch bei Blutserum mit extrem erhöhtem Jodidgehalt. Einen weiteren Vorteil bildet die erzielte Arbeits- und Zeitersparnis.
