    Notes: Summary 1. A preparation of the isolated stomach and vagus of the guinea-pig is described which reacts to direct and indirect electrical stimulation with consistent contractions during several hours. 2. Pharmacological investigations on this preparation had the following results: a) ganglionic blocking drugs (hexamethonium, Pendiomid et al.) abolished the contractions produced by nicotine and vagal stimulation, whereas those produced by direct electrical stimulation, acetylcholine, histamine and barium remained intact. b) Atropine abolished the effect of nicotine and vagal stimulation as well as the effect of direct electrical stimulation. However, this paralysing action appeared only after a considerably longer presence of atropine in the bath than was necessary to abolish acetylcholine contractions. 3. In the presence of atropine inhibitory effects were seen on direct electrical stimulation. They were somewhat reduced by ganglionic blocking drugs, but could be abolished by sympatholytic drugs.

On the isolated preparation of the vagus and stomach of guineapigs, rabbits and rats atropine abolishes immediately the action of acetylcholine whereas it takes several minutes to abolish the effect of electrical vagal stimulation. 2. On this preparation electrical stimulation of the vagus sets free a substance which contracts an isolated test-ileum suspended in the same Tyrode-bath. The effect of the transmitter is abolished by atropine, enhanced by eserine. 3. The results indicate that also in the digestive tract acetylcholine is the transmitter of parasympathetic-motor nerve impulses; they do not support the view of liberation of an atropine-resistant transmitter. — High doses of tryptamine which on the intestine inhibit the action of 5-oxytryptamine do not influence the effect of electrical vagal stimulation.

    Notes: Zusammenfassung Untersuchungen über den Mechanismus der nach hohen Strophanthindosen (150–300 γ/100 g s.c.) an Ratten auftretenden Lähmungen haben zu folgenden Ergebnissen geführt: 1. Von der Hemmungswirkung am M. gastrocnemius ist zuerst die indirekte, später auch die direkte Erregbarkeit betroffen. 2. Diese Hemmung der Muskelerregbarkeit durch Strophanthin läßt sich am isolierten Phrenicus-Zwerchfellpräparat der Ratte nur zeigen, wenn man den Kaliumgehalt der Tyrodelösung auf 30 mg-% erhöht. 3. Nach lähmenden Strophanthindosen kommt es am intakten Tier zu einer Erhöhung des Serumkaliumspiegels und zu einer Abnahme des Kaliumgehaltes der Muskulatur (M. gastrocnemius, Zwerchfell, Herz). 4. Für einen ursächlichen Zusammenhang zwischen der muskellähmenden Wirkung des Strophanthins und einer Abnahme des Kaliumgradienten spricht ferner, daß am Ganztier die Lähmung zum Zeitpunkt der stärksten Kaliämie auftritt. Am Zwerchfellpräparat wird die Strophanthinlähmung in gleicher Weise wie die durch hohe Kaliumkonzentrationen (50 mg-%) verursachte durch Adrenalin und Calcium-Ionen antagonistisch beeinflußt. 5. Die lähmende — und temperatursenkende — sowie die Kaliämie verursachende Wirkung ist beim k-Strophanthin fast doppelt so stark wie beim g-Strophanthin.

The excretion of sodium is increased only with high doses (50 bis 200 μg/100 g); with lower doses (12,5 μg/100 g) it may be diminished. 2. K-strophanthidine has less influence on the excretion of potassium. — G-strophanthine (Oubain) and Scillaren have only about half the activity of k-strophanthine, whereas Convallatoxin is about equipotent.

    Notes: Zusammenfassung 1. An narkotisierten Katzen und Ratten ist die tödliche Wirkung hoher intravenöser Cholindosen wesentlich durch eine Lähmung der Atmung bedingt. Dieser liegt eine Blockierung der neuromuskulären Erregungsübertragung zugrunde. Bei künstlicher Beatmung werden sonst tödliche Dosen überlebt. 2. Die blutdrucksteigernde Wirkung höherer Cholindosen läßt sich durch Atropin abschwächen oder aufheben. Das spricht aber nicht gegen den nicotinartigen Charakter dieser Wirkung, die auf der Erregung sympathischer Ganglien und einer Adrenalinausschüttung aus dem Nebennierenmark beruht, sondern für eine ganglionär blockierende — Antinicotinwirkung — des Atropins. 3. In Versuchen an weißen Mäusen ist die DL 50 des Cholins nach Vorbehandlung mit Atropin nicht signifikant erhöht. Regitin, Buscopan, am deutlichsten aber Hexamethonium vermindern die Giftigkeit.
