    Notizen: Abstract  This paper describes a methodology to calculate methotrexate (MTX) pharmacokinetic parameters after intramuscular administration using two samples and the population parameters. Total and free MTX were measured over a 36-h period in 56 rheumatoid arthritis patients; 14 patients were studied after a two-dose scheme at 15-day intervals. The Hill equation was used to relate the free MTX to the total MTX changes in plasma concentrations, and a two-compartment open model was used to fit the total MTX plasma concentrations. A non-linear mixed effect procedure was used to estimate the population parameters and to explore the interindividual variability in relation to the following covariables: age, weight, height, haemoglobin, erythrocyte sedimentation rate, platelet count, creatinine clearance, rheumatoid factor, C-reactive protein, swelling joint count, and Ritchie’s articular index. Population parameters were evaluated for 40 patients using a three-step approach. The population average parameters and the interindividual variabilities expressed as coefficients of variation (CV%) were: CL, 6.94 l ⋅ h−1 (20.5%); V, 34.8 l (32.2%); k12, 0.0838 h−1 (47.7%); k21, 0.0769 h−1 (61.6%); ka, 4.31 h−1 (58%); Emax, 1.12 μmol ⋅ l−1 (19.7%); γ, 0.932 (12.3%); and EC50, 2.14 μmol ⋅ l−1 (27.3%). Thirty additional data sets (16 new patients and 14 patients of the previous population but treated on a separate occasion) were used to evaluate the predictive performance of the population parameters. Twelve blood samples were collected from each individual in order to calculate individual parameters using standard fitting procedures. These values were compared to the ones estimated using a Bayesian approach with population parameters as a priori information together with two samples, selected from the individual observations. The results show that the bias was not statistically different from zero and the precision of these parameters was excellent.

    Notizen: Abstract Hydroxamic acids have been shown to be toxic to many pest insects and pathogens. In this study, the behavioral responses of western corn rootworm larvae to naturally occurring and synthetic hydroxamic acids were investigated. In a choice test between corn roots treated with hydroxamic acids and roots treated with distilled water (control), western corn rootworm larvae chose to burrow into the control roots significantly more often than compoundtreated roots. In addition, when corn roots were treated with different hydroxamic acids in a designed searching-behavior test, neonate larvae of western corn rootworm responded by significantly reducing the number of turns, while the area searched and locomotor rate significantly increased. The responses were dependent on the concentrations of the test compounds. These results suggested that hydroxamic acids were acting as behavior-modifying and possibly feeding-deterrent chemicals.

    Notizen: The practical value of a wave function derives from its ability to estimate or predict the chemical and physical properties of the electronic structure which it describes. Reliability is defined as a property to be evaluated on the basis of (i) the magnitude of the difference between the measured and calculated values of the observable together with (ii) the sign of the difference, (ii) the number and nature of observables correctly estimated, and (iv) the number and type of electronic structures that are correctly estimated. Systematic statistical comparison implies the existence of both internally consistent sets of wave functions for homologous series of molecules and their corresponding experimental values together with reliable error estimates. The most complete data base currently available for comparison is the spectroscopic constants for the first and second row diatomic hydrides. Utilizing appropriate statistical comparison techniques, four approximations (CEPA, PNO-CI, GTO-SCF, and STO-SCF) are compared among themselves and against experimentally measured values. The CEPA approximation yields differences from experiment that approximate a normal error distribution, while the other approximations show systematic departures from experiment. Two values, ωe and ωeχe, for SH exhibit differences large enough to cast doubt upon the calculated value, the experimental value, or both.

    Notizen: Power moments for all of the reduced hydrogenic Green's functions have been computed. These moments have the same form as the corresponding moments of the free-particle Green's functions. Green's functions of the second kind are defined, and uses for these objects in model potential theory and the theory of many-body Green's functions are pointed out. In the case of the ground state of the hydrogenic atom, the Green's function of the second kind has been given.

    Notizen: A theory of isoelectronic molecules which describes stable and metastable members of a sequence has been developed. To achieve this synthesis, it has been necessary to require that the total electronic energy surface E(R,Z,Z') for the sequence contain critical points - that is, values of the nuclear charges Z and Z' at which energy minimum, maximum [at Re(min) and Re(max)], and horizontal inflection points occur. For ground state sequences a primary physical source of these extreme points is the screened, coulombic repulsion of like-charged atomic centers in the diatomics. With this realization, we can write analytical forms that have the correct scaling behavior and which properly model the screened, coulombic repulsion for E along certain straight-line trajectories in the (Z,Z') plane. This leads to the observation that Re(max) values diverge logarithmically in λ-like transitions wherever the screened coulombic repulsion becomes small as the nuclear charges vary along those trajectories.At the horizontal inflection points the E surface contains A2 folds, as required by Thom's theorem for analytical surfaces containing one control parameter. Within the isoelectronic sequence molecular subgroups have been noted and explained in terms of the underlying atomic shell structures of the molecules' constituent atoms. Using input data for seven stable molecules together with the analytical surface selected for study, we have estimated the equilibrium bond distances Re(min), dissociation energies De(min), and harmonic force constants E(2)(min) for 18 other neutral and charged species.

    Notizen: Dialysis was attempted as a means to alleviate the product-activated controls presumed to limit the formation of threonine by an auxotrophic mutant of Escherichia coli strain W. The occurrence of inhibition rather than enhancement of yields by dialysis was traced to the fact that threonine actually was not inhibiting its own synthesis. Instead, α,ε-diaminopimelic acid became depleted, but the imbalance could not be corrected by exogenous replacement.
