    Notes: Abstract  Guinea-pig ureteric smooth muscle is unusual in that intracellular acidification increases and alkalinization decreases force production. To help elucidate the mechanism underlying these effects on force we have investigated the effects of changing intracellular pH on both calcium and potassium currents in single cells isolated from the guinea-pig ureter to determine their possible role in force development. Depolarization to +40 mV resulted in a fast transient outward current which was inhibited by 4-aminopyridine but not tetraethylammonium. Intracellular alkalinization (20 mM trimethylamine) increased this current to 179 ± 24% of the control and resulted in the development of a slowly activating large outward current which was inhibited by tetraethylammonium and washout. Acidification (40 mM sodium butyrate) decreased the fast transient outward current to 58 ± 3% of the control and did not produce a slowly activating current. When potassium was replaced by caesium in the pipette solution, depolarization to 0 mV resulted in an inward calcium current which was abolished by nifedipine. Intracellular alkalinization increased this current to 126 ± 11% of the control whereas acidification had the opposite effect, decreasing it to 55 ± 10%. Furthermore, current-clamp experiments showed that intracellular alkalinization inhibited the amplitude of the action potential, therefore decreasing excitability of the cell. From our results, we suggest that the predominant effects of intracellular pH on force production in the guinea-pig ureter are mediated via the modulation of outward potassium currents (thereby reducing excitability of the tissue) rather than the effects on the inward calcium current.

    Notes: Summary The increase in in vitro adrenal tyrosine hydroxylase (TH) activity caused by drugs which produce an increase in preganglionic nerve activity as a result of actions outside the central nervous system was increased by the administration of diazepam. However, other central nervous system depressants such as pentobarbital, phenobarbital and diphenylhydantion were not found to share this property. Adrenal enzyme induction provoked by isoproterenol and 6-hydroxydopamine was increased by diazepam administration, whereas that produced by reserpine or insulin was not altered. The enhanced adrenal TH activity produced by diazepam pretreatment and isoproterenol administration could not be attributed to an increase in intensity or to an altered body temperature. Administration of 5-hydroxytryptophan can antagonize the induction of adrenal TH produced by isoproterenol, however, it did not block the TH induction produced by concurrent administration of diazepam and isoproterenol. The effect of diazepam on adrenal TH induction may be the result of its ability to interfere with central nervous system amine containing nerve functions which modulate preganglionic nerve activity, but the particular neurotransmitter system involved has not been determined.

    Notes: Capillary supercritical fluid chromatography (SFC) is proving to be a viable and useful separation method for thermally labile and nonvolatile materials. As with other capillary chromatographic techniques, very fast separations can be accomplished by sacrificing total efficiency and optimizing the conditions for rapid analysis. This is achieved using short, small-bore capillary columns, increased mobile phase linear velocities and very fast pressure programming rates. These principles are demonstrated for the rapid separation of selected component systems.

    Notes: Capillary supercritical fluid chromatography and its combination with mass spectrometry (SFC-MS) is an important analytical methodology for the analysis of thermally labile and high molecular weight compounds. The mass spectrometer provides sensitive and highly selective detection for the chromatographic effluent. The same physical-chemical properties of supercritical fluids that provide important chromatographic advantages are also important for the transport and gas phase introduction of analyte molecules into the mass spectrometer. The low mobile phase flow rates of small diameter (〈 100 μm i.d.) capillary columns allow the total chromatographic effluent to be introduced with conventional, but often slightly modified, ion source configurations for quadrupole mass spectrometers with either electron impact (EI) or chemical ionization (CI) detection. The full range of CI reagents can also be used to obtain additional selectivity and complementary structural data. The instrumentation and operating parameters for capillary SFC-MS methods are described. The applicability and usefulness of various SFC-MS analysis methods are demonstrated using a variety of samples.

    Notes: Vinylidendicarbonitril (Vinylidencyanid) zeigt bei radikalisch gestarteter Copolymerisation eine viel stärkere Neigung zur Bildung von alternierend aufgebauten 1:1-Copolymeren als Acrylnitril. Während Polyvinylidencyanid trotz der molekularen Symmetrie nicht kristallisiert, sind alternierende Copolymere, vor allem mit Butadien, Isopren, Isobutylen und Vinylidenchlorid, leicht kristallisierbar. Aus höherschmelzenden Copolymeren wurden Fasern hergestellt und auf ihre physikalischen Eigenschaften geprüft. Besonders Fasern aus den Copolymeren mit Vinylacetat und Vinylchlorid zeigen im nassen und im trockenen Zustand ein hervorragendes elastisches Verhalten. Die Faser aus dem alternierend aufgebauten Copolymeren von Vinylidencyanid und Vinylacetat (Darvan®-Nytrilfaser) ist nur mäßig orientiert und zeichnet sich durch Weichheit, gute elastische Erholung und Sprungelastizität im trockenen und nassen Zustand aus.

    Notes: A method is described whereby, under software control, the scan function employed to collisionally excite ions in an ion-trap mass spectrometer is regulated so as to obtain parent ion (fixed product) tandem mass spectral data. At the same time, constant neutral-loss information is also provided. Key features are the automatic ‘intelligent’ selection of parent ions from observation of the initial mass spectrum and the scanning of the applied ‘tickle’ frequency in order to locate the precise resonance points for excitation of these ions. Results for the model compounds perfluorotributylamine (‘FC43’) and n-butylbenzene are reported and complicating effects such as the unimolecular decay of the precursor ion species and charge exchange between product ions and the original sample material are discussed.

    Notes: The electrospray ionization (ESI) and plasma desorption (PD) mass spectra of over 20 peptides and proteins, with molecular weights (Mr) ranging between 1182 and 143000, have been directly compared. Both techniques produced molecular ions for the majority of materials studied; however, neither approach proved to be universally applicable. PD failed for a number of proteins that were successfully analyzed by ESI, including some of very high Mr. On the other hand, ESI failed for proteins that apparently could not acquire a sufficient number of positive charges to allow transmission through the quadrupole mass filter. A non-covalently bound adduct, ribonuclease S, did not survive either method intact and a simple glycoprotein, ribonuclease B, did not yield the expected molecular ion with either approach. The mass measurement accuracy of quadrupole ESI is five to tenfold better than obtained with a commercial time-of-flight PD mass spectrometer. Furthermore, ESI's superior mass resolution (with quadrupole mass spectrometers) will prove to be particularly helpful for the characterization of mixtures of closely related materials. Sensitivity was only compared qualitatively but is highly compound dependent with both techniques. In favorable cases, ESI spectra can be obtained on low femtomolar quantities of proteins while PD typically requires several hundred femtomoles to high picomoles, depending on a number of factors including Mr.
