    Description / Table of Contents: Zusammenfassung Die verallgemeinerte Methode der konjugierten Gradienten (GCG) von Concus und Golub [1] und Widlund [2] und ihre angepaßte Form (AGCG) werden bei der numerischen Lösung von Least-Squares-Problemen untersucht. Einige numerische Vergleiche mit dem SOR-Verfahren [3] und dem CG-Verfahren [4] werden gezogen und deuten an, daß die GCG-Methode stets besser als die SOR-Methode ist und daß das AGCG-Schema allen anderen erwähnten Methoden vorzuziehen ist.
    Notes: Abstract The Generalized Conjugate Gradient (GCG) method of Concus and Golub [1] and Widlund [2] and its adjusted form (AGCG) are considered for the numerical solution of the least squares problems. Some numerical comparisons with the SOR [3] and CG [4] method are also given indicate that the GCG method is always better than the SOR [3] and that the adjusted GCG (AGCG) scheme is preferable to the CG, GCG and SOR methods.

    Notes: A dichotomy of hair-cell types has been found in the bullfrog sacculus, and considerable evidence supports the view that one type (“peripheral”) is transformed during macular growth to the other type (“central”). Between the periphery and the center of the macula, one finds a gradation of form from “peripheral” to “central” type. Occasionally in adults and more often in stage-26 tadpoles one finds the presumably younger peripheral type of hair cell occurring well beyond the limits of the macula proper. The apparent morphogenic sequence for saccular hair cells is (1) development of a kinocillum on an endolymphatic epithelial cell, (2) gradual transformation of microvilli into stereocilia, (3) growth of the stereocilia and development of kinociliary bulb, (4) achievement of final size and form.

    Notes: The use of a laser-desorption mass spectrometer to examine a coal liquefaction extract known to contain high-molecular-mass material (from size-exclusion chromatography) has shown the presence of components of molecular mass in excess of 104Da for the first time. The molecules detected differ from each other in molecular mass by a few hundred or a few thousand Da. These preliminary results indicate that the basic coal-structure molecules probably consist of aromatic clusters on a chin, in reasonable agreement with current theories of coal structure.

    Notes: Molecular masses up to 270 000 u have been observed during matrix-assisted laser desorption mass spectroscopy on a sample of bituminous coal; the major fraction of desorbed ions appeared between m/z values of 800 and 5000. Similar mass distributions have been found in a coal-liquefaction extract and a pyrolysis tar. The highest molecular masses identified in this work far exceed previously reported high molecular masses found in coal-derived products (12 000 u), which were also detected by laser-desorption mass spectrometry.

    Notes: The production of carbon clusters (but not fullerenes) from coal-derived material by the use of laser desorption mass spectrometry is described. Benzo(e)pyrene, a standard polynuclear hydrocarbon found in coal tars, also gave carbon clusters despite using low laser power to avoid pyrolysis of target molecules. Positive- and negative-ion spectra of the standard included carbon clusters. These clusters are formed by the laser energy and can be confused with aromatic hydrocarbons of the same nominal masses. They have no relevance for studies of coal structure.

    Notes: By repeated selection for longer survival in an isotonic solution of glycerol, a stable subline of Novikoff rat hepatoma cells has been isolated. The cells exhibit markedly increased resistances to osmotic lysis in isotonic solutions of glycerl. They are twice as large and have twice as many chromosomes as cells of the parental line. It is suggested that the osmotic stress procedure can be extended for the selection of numerous kinds of mutants and can be used as a method of analysis of membrane properties.

    Notes: The cells responsible for the long-term in vitro generation of murine mast cells have been examined. Sequential analysis of all colony types obtained from cultures of spleen or bone marrow cells showed that only colonies derived from multipotential cells (mixed-erythroid colonies) or mast cell progenitors, contained cells responsible for mast cell generation in liquid cultures. Primary colony growth and subsequent maintenance of mast cells in liquid cultures was dependent upon pokeweed mitogen-stimulated spleen cell-conditioned medium (SCM). Mixed-erythroid colonies from 14-day cultures of spleen cells had the greatest capacity for mast cell generation. Analysis by clone splitting and transfer to high (20%) and low (2.5%) concentrations of SCM showed that the concentration of SCM used in either the primary colony culture or subsequent liquid culture phase altered both the proliferative capacity of the mast cells generated and the frequency of mast cell progenitors within individual mixed-erythroid colonies. Thus, mixed-erythroid colonies stimulated with 2.5% SCM contained the highest proportion of mast cell progenitors (34% of colonies) and when stimulated with 20% SCM, approximately fourfold higher numbers of mast cells were produced at weekly intervals from liquid cultures maintained in 2.5% SCM compared to parallel liquid cultures containing 20% SCM. These studies confirm the hemopoietic origin of mast cells and demonstrate that a factor(s) in SCM is able to modulate their proliferative potential.

    Notes: The helical molecule 1-fluorobenzo[ c] phenanthrene shows a large (17.4 Hz) 13C-19F coupling resulting primarily from direct through-space overlap between a fluorine lone pair and the sterically opposed C—H bond. After application of corrections for estimated through-bond contributions, comparison of the magnitude of this coupling with the analogous couplings in 4-fluorophenanthrene, 10-fluorobenzo[ a] pyrene, 11-fluoro-5-methylchrysene and 7-fluorobenzo[b] fluoranthene reveals an exponential decrease with increasing H … F internuclear distance, consistent with theoretical predictions. No evidence was found for an angular dependence of the coupling. HETCOR experiments showed the signs of n+1J(HF) in 1-fluorobenzo[ c] phenanthrene, 4-fluorophenanthrene, 10-fluorobenzo[ a] pyrene and 11-fluoro-5-methylchrysene to be opposite to those of the corresponding nJ(CF), hence negative. Substituents at both positions 4 (para to fluorine) and 9 (para to the sterically opposed C—H bond) exert only a minor influence on 5J(CF), the couplings in the latter series responding largely to sterically induced distortions of the molecular framework, and in the former series to both steric distortions and an electronic effect. The difference in 5J(CF) between isomers is a measure of the electronic effect of the substituent. Electron-withdrawing substituents para to fluorine reduce the magnitude of the through-space coupling whereas electron-releasing substituents increase the coupling.

    Notes: We show that high-resolution transmission electron microscopy can be used directly to reveal the structure of the metal framework and its defects in Ba2YCu3O7. The use of dynamical effects under carefully selected and controlled conditions allows the detection of long-range order in the disposition of the oxygen vacancies, but inevitable misalignments make the quantification of the order parameter difficult. Specimen preparation methods are compared, and a wide-angle convergent-beam pattern is demonstrated.
