    Notes: Summary The pharmacokinetics of theophylline (200 mg i.v.) were determined in 20 geriatric patients with multiple diseases. Serum concentrations were fitted to an open 2-compartment model. Percent changes in venous pCO2 and pO2 were used as parameters of the pharmacodynamic action. Total clearance was decreased and elimination half-life of theophylline was found to be prolonged in the elderly patients compared with data of a study with young healthy volunteers. The stronger the pharmacodynamic action of theophylline (percent change of venous pCO2 after 30 min), the faster was its elimination and the lower were measured concentrations after 2, 6, and 12 h.

    Description / Table of Contents: Zusammenfassung Bei 10 alten (77,2 Jahren) Patientinnen und 10 jungen (20,1 Jahren) weiblichen Probanden wurde die Pharmakokinetik von Canrenon nach mehrfacher täglicher oraler Gabe von 100 mg Spironolcaton unter den Bedingungen des steady-state untersucht. Die Konzentrationsbestimmungen erfolgten simultan mit einer spezifischen hochdruckflüssigkeits-chromatographischen und mit einer weniger spezifischen fluorimetrischen Methode. Maximale und mittlere Serumkonzentrationen von Canrenon waren bei den älteren Patientinnen etwa doppelt so hoch wie bei den jungen Versuchspersonen. Dies war die Folge einer verminderten Eliminationskapazität für Spironolacton bei den alten Patientinnen. Dabei war der Anteil von fluorogenen Metaboliten zu Carenon bei den alten Patientinnen höher als bei den jungen Probanden, so daß offenbar auch Verschiebungen in den Stoffwechselwegen von Spironolacton im Alter auftreten.
    Notes: Summary The pharmacokinetics of canrenone were compared in 10 elderly (77.2 years) patients and 10 young (20.1 years) female persons after multiple oral dosing of 100 mg Spironolactone during steady-state. The concentrations were determined using both a specific HPLC-assay and a nonspecific fluorometric assay. Maximum as well as mean concentrations of canrenone in serum of the elderly subjects were approximately twice as high as those in the young. This was the consequence of an impaired capacity for elimination of spironolactone in the elderly subject. In addition the ratio of the other fluorigenic metabolites and of canrenone were higher in the elderly. Thus also shifts in the metabolic pathways of spironolactone occure with progressing age.

    Description / Table of Contents: Zusammenfassung In der vorliegenden Arbeit sollte der Einfluß von Furosemid auf das Aktivitätsverhalten lysosomaler Enzyme der Leber untersucht werden. 10 Wochen und 1 Jahr alte männliche Albinoratten wurden an 4 aufeinanderfolgenden Tagen mit jeweils 40 mg Furosemid behandelt. In Anlehnung an die Methode von de Duve wurde durch fraktionierte Zentrifugation eine Lysosomenfraktion und ein entsprechender Überstand gewonnen, in dem die Aktivitäten der lysosomalen Enzyme β-Glucuronidase, β-Acetylglucosaminidase, Kathepsin D und eines „kollagenolytischen“ Enzyms bestimmt wurden. Der Proteingehalt diente als Bezugsgröße der Enzymaktivitäten. Außerdem wurden im Serum die cytoplasmatischen Enzyme GOT, GPT, Gamma-GT und alkalische Phosphatase bestimmt. Die Enzymaktivitäten zeigten ein vom Alter der Tiere abhängiges Verhalten. Bei den 1 Jahr alten Tieren kam es unter Furosemidgaben zu einer Aktivitätszunahme der β-Glucuronidase und von Kathepsin D. im Lysosomensedimentüberstand und im Lysosomensediment, ebenso des „kollagenolytischen“ Enzyms im Lysosomensediment. Bei den 10 Wochen alten Tieren wurde — bei konstantem Proteingehalt — eine Aktivitätsabnahme der β-Glucuronidase, der β-Acetylglucosaminidase und von Kathepsin D im Lysosomensediment ermittelt. Die Ergebnisse wurden denen in der Literatur bekannten Daten gegenübergestellt und diskutiert.
    Notes: Summary The present study investigated the activities of lysosomal enzymes of the liver after administration of Furosemid. 10 weeks and 1-year old male albino rats were treated with 40 mg Furosemid for 4 subsequent days. According to the method devised by de Duve a sediment rich in lysosomes was produced by fractionated centrifugation and subsequently the enzyme activity of β-glucuronidase, β-acetylglucosaminidase, cathepsin D and a collagenolytic enzyme was measured in the sediment as well as in the corresponding lysosomal supernatant. The protein content served as a reference for the enzyme activities. In addition, we investigated the activities of cytoplasmic enzymes such as GOT, GPT, gamma-GT and the alkaline phosphatase. The enzyme activity changes were age-dependent. With Furosemid treatment the activities of β-glucuronidase and cathepsin D increased in the lysosomal supernatant and the lysosomal sediment of the 1-year old rats, whereas the activities of the collagenolytic enzyme increased in the lysosomal sediment of the same group. In the lysosomal sediment of the 10-weeks old rats a decrease of β-glucuronidase, β-acetylglucosaminidase and cathepsin D was observed. These results are discussed in the light of reports from the literature.
