    Description / Table of Contents: Zusammenfassung Es wurden der Stoffwechsel und die Organverteilung von doppelt-markiertem Vitamin D3 (1,2-3H-4-14C-Cholecalciferol) an Ratten untersucht, die entweder mit Phenobarbital (PB) oder Diphenylhydantoin (DPH) behandelt waren. Während die DPH-behandelten Ratten ein Stoffwechselmuster und eine Organverteilung ähnlich den Normaltieren aufwiesen, hatten die mit PB behandelten Ratten am 3. Untersuchungstag eine höhere Konzentration von 25-Hydroxycholecalciferol im Serum und am 10. Tag der Untersuchung eine im Vergleich zu den Normaltieren und DPH-Ratten erniedrigte Radioaktivität in der Leber und Niere. Da jedoch die Calcium-Retention nicht nur in der PB-Gruppe, sondern auch in den DPH-Tieren erniedrigt gefunden wurde, kann derzeit noch nicht entschieden werden, ob die bei Epilepsiekranken nachweisbaren vorwiegend osteomalacischen Knochenveränderungen durch eine Störung des Vitamin D-Stoffwechsels verursacht werden.
    Notes: Summary The metabolism and the organ distribution of double-labelled vitamin D3 (1,2-3H-4-14C-cholecalciferol) has been investigated in rats receiving either phenobarbital (PB) or diphenylhydantoin (DPH). Whereas the DPH-treated rats showed a metabolic pattern and organ distribution comparable to the control animals, the PB-treated rats had a greater amount of radioactivity (mainly as 25-Hydroxycholecalciferol) in their serum at 3 days, and at 10 days they showed a reduced concentration of radioactivity in their liver and kidney. As the retention of47calcium was not only decreased in the PB-group but as well in the DPH-group, which showed an almost normal handling of the injected vitamin D3, it remains speculative, as to whether or not the PB-induced alterations in the metabolism of vitamin D are responsible for the reported occurence of osteomalacic bone disease in patients treated by anticonvulsants.

    Notes: Summary Investigations have been performed on the metabolism and organ distribution of 5,6-trans-25(OH)D3, which represents an analog of vitamin D3 being active in uremia. This analog is rapidly metabolized into at least two more metabolites. The main storage organ for this compound seems to be fat, whereas liver and intestinal mucosa showed at 24 hrs the highest concentration of polar metabolites. In contrast to 25(OH)D3 5,6-trans-25 (OH)D3 is much less absorbed from the intestinal tract, which should be considered if this compound is applied for treatment of uremic bone diseases.

    Description / Table of Contents: Summary Elimination of ampicillin and gentamicin by hemofiltration was measured in 5 and 4 patients, respectively, being treated for end stage renal failure. Serum half-life time of ampicillin after a single I.V. dose of 2 g, was found to be 2.97±0.73 h, and the filtrate concentrations declined parallel to the serum ampicillin levels, as was to be expected. The serum half-life time after a single I.V. dose of 1 mg/kg body weight gentamicin was 3.26±0.42 h, and considerably shorter than T/2 of gentamicin in conventional hemodialysis, while the filtrate concentrations increased paradoxically with declining serum levels. This could be explained by binding of this aminoglycoside to the polyacrylnitril membrane of the RP 6® dialyser used for hemofiltration.
    Notes: Zusammenfassung Bei 5 bzw. 4 Patienten mit terminaler Niereninsuffizienz wurde die Elimination von Ampicillin bzw. Gentamicin durch die Hämofiltration gemessen. Die Serum-Halbwertzeit von Ampicillin betrug nach einer Einzeldosis von 2 g i.v. 2,97±0,73 h und die Filtratkonzentration nahm, wie zu erwarten war, parallel den Serum-Ampicillin-Spiegeln ab. Die Serum-Halbwertzeit von Gentamicin betrug nach einer Einzeldosis von 1 mg/kg Körpergewicht i.v. 3,26±0,42 h und war damit beträchtlich kürzer als T/2 von Gentamicin bei der konventionellen Hämodialyse, während die Filtratkonzentrationen paradoxerweise mit abfallenden Serum-Spiegeln anstiegen. Dieses Verhalten könnte durch eine Bindung des Aminoglykosids an die Polyacrylnitrilmembran des RP 6® Dialysators, wie er für die Hämofiltration verwendet wurde, erklärt werden.

    Notes: Summary The plant Solanum malacoxylon (S.M.) is known to cause severe soft tissue calcifications in cattle and sheep and has recently become of special, interest since it exerts biological actions which resemble those of vitamin D. In order to investigate whether S.M. is capable to improve the rachitic bone changes in vitamin d and phosphate deficient rats, a watery extract of 50, 100 and 200 mg S.M. was fed daily to these animals over a period of 10 days. The width of epiphyseal plates was compared after that time with those of rats treated with 0, 0.225, 0.45 and 0.9 IU of vitamin D3 daily. There was a dose related curative effect of S.M. on the epiphyseal lesions very similar to that of vitamin D3. The hypercalcemic and hyperphosphatemic effect of S.M. was identical to that of vitamin D3. The 25-hydroxy-vitamin D serum levels remained almost undetectable, in the S.M. treated rats as well as in the vitamin D3 treated animals.
