Notes: Abstract Homogenates of rat hypothalamic tissue were fractionated by means of discontinuous sucrose density gradient centrifugation. Immunoreactive luteinizing hormone releasing hormone (LHRH), thyrotropin releasing hormone (TRH), and α-melanocyte stimulating hormone (α-MSH) were found to be concentrated in the synaptosome-enriched fraction. This fraction was suspended in 0.32 M sucrose and the release of the three peptides was investigated. After incubation, the synaptosomes were re-isolated by ultrafiltration, and the concentration of each peptide in the ultrafiltrate was determined by radioimmunoassay. When the synaptosomal fraction was incubated at 30° C in 0.32 M sucrose containing either 60 mM K+-2 mM Ca2+ or 140 mM Na+ alone a release of LHRH, TRH, and α-MSH occurred. Of the total content 30–50% of LHRH but only about 10% of TRH and α-MSH was releasable. When the synaptosome preparation was preincubated for 30 min at 30°C with 10−4 M vinblastine. K+- as well as Na+-induced release of LHRH, THR, and α-MSH was inhibited, and the stimulatory effect of each cation was almost totally blocked by preincubation with 5×10−4 and 10−3 M vinblastine. The inhibitory action of vinblastine (5×10−4 M) did not affect the oxidation of glucose to CO2 by the synaptosomes. The results of the present investigation demonstrate that synaptosome-enriched fractions of hypothalamic origin are more stable with respect to LHRH, TRH, and α-MSH release during incubation in isotonic sucrose than they are in ionic solutions, and that the peptides are released by a vinblastine-sensitive mechanism.

Notes: Summary Three independent isolates ofChlamydomonas, selected for caffeine resistance, were found to arrest in G1 phase, as determined by quantitative fluorescence measurements of DNA, when grown at a non-permissive temperature. This cell cycle arrest correlated with lowered levels of cAMP and of adenylate cyclase activity. The arrested cells could be rescued by added cAMP but not AMP, hence the defect was not one of general purine metabolism. Back-crosses to wild type revealed that the phenotypes observed result from a combination of three separable mutations. It is clear that the mutations define functions that are more stringently required for cell division than for growth since the mutant strains are able to grow up to fifteen times normal size while blocked at the non-permissive temperature. The possible interaction of cAMP dependent events with division is discussed.

Notes: A series of 17 cysteine, N-acetyl cysteine, glutathione, and N-trifluoroacetyl glutathione conjugates have been prepared, and their thermospray (TSP) spectra have been recorded in the positive and negative ion modes. The compounds undergo extensive fragmentation, which primarily occurs at the carbon-sulfur bonds. For most of the compounds, positive ion TSP is more sensitive than negative ion thermospray. Probably due to the thermal lability of these adducts, the quality of the spectra obtained are dependent on source conditions, requiring fine control of the vaporization/desolvation process.

Notes: Metabolism of 4,4′-thio-bis-(2-t-butyl-5-methylphenol)(TBBC) in rats resulted in the formation of a glucuronide conjugate of TBBC. This conjugate was identified by a combination of high-performance liquid chromatography (HPLC) and mass spectrometry and a tandem mass spectrometric method employing a fast particle ionization technique. A comparison of mass spectral data from the in-vivo metabolite of TBBC and an enzymically synthesized glucuronide conjugate of TBBC showed the metabolite to be the monoglucuronide.

Notes: A series of Senecio alkaloid and alkaloid N-oxide standards has been analyzed using positive and negative ion thermospray liquid chromatography/mass spectrometry (LC/MS) with an ammonium acetate-containing mobile phase. On-line separations of pyrrolizidine alkaloids from extracts of Senecio jacobaea (tansy ragwort) and Senecio vulgaris (common groundsel) were done using an ammonium hydroxide-containing mobile phase. All of the alkaloids known to be present in the extracts were detected by ammonium hydroxide thermospray LC/MS, as well as many other components which may be as-yet-unidentified alkaloids.

Notes: The utility of combined open-tubular liquid chromatography/negative ion chemical ionization (NCI) mass spectrometry for the separation and analysis of trifluralin, a commonly used pre-emergence herbicide, and seven of its metabolites has been demonstrated. The NCI mass spectra for these compounds are reported. Some of the NCI mass spectra were obtained from as little as 50 pg of analyte.

Notes: The following indole compounds have been unequivocally identified and semi-quantitated in culture supernatants of Rhizobium strains by gas chromatography mass spectrometry: indole-3-aldehyde, trytophol, indole-3-carboxylic acid, indole-3-glycollc acid, indole-3-actic acid, indole-3-glyoxylic acid, N-acethyl-L-tryptophan and indole-3-pyruvic acid, as well as indole-3-acetic acid. The strains included a number of chosen Rhizobium mutants defective in various stages of nodule formation, and the biological implications of the findings are discussed. Two alternative approaches to sample purification were taken, one based primarily on solvent partitioning and the other on high-performance liquid chromatography. The advantages of each method are discussed. Gas chromatographic retention times and mass spectra are given for the TMS derivatives of 17 authentic indole standards.
