Notes: Summary Studies in anorectic tumor-bearing rats indicate that anorexia is correlated to imbalances of neutral amino acids in blood and CNS. Consequently plasma amino acids of patients with neoplastic and non-neoplastic internal diseases were studied during phases of anorexia; special regard was given to the precursors of dopamine and serotonin. Anorectic patients were compared to non-anorectic patients with neoplasia. During anorexia, plasma levels of valine and leucine and hence the ratio of the molar concentrations of Val+Leu+Ile/Phe+Tyr were significantly decreased in each anorectic patient as compared to non-anorectic patients whose ratios were always within the normal ranges. As aromatic and branched-chain amino acids compete for penetration of the blood brain barrier, the decrease of the amino acid ratio may induce a raised flux of phenylalanine and tyrosine into the CNS which results in an increased activation of dopaminergic neurons — known to cause anorexia.

Notes: Abstract The effect of tunicamycin, an inhibitor of N-glycosylation of proteins, on growth and on synthesis of DNA and protein was studied in suspension cultures from Nicotiana tabacum and Catharanthus rosea. In the presence of 0.1–1 μg · ml-1 tunicamycin, cell division and DNA synthesis stopped in cells which had been proliferating logarithmically, but protein formation continued. Cytophotometric determination of the nuclear DNA content in Catharanthus cells showed that a cell-cycle arrest had occurred in G1 phase. Metabolic labelling of cells with the glycoprotein precursors glucosamine or mannose was inhibited, too. The results indicate that one or more glycoproteins are needed for the cell to pass through the G1 phase, as was recently postulated for animal and yeast cells.

Notes: Highly Selective pH-Solid-State-Sensors Based on Proton-Carrier-PVC-Membranes with Polymeric PlasticizersBasing on membrane chemical experiments, the systematic development of a highly selective proton carrier PVC membrance for solid state contact sensors with trioctylamine as ligand and a phthalic acid polyester as polymeric plasticizer is described. Its resistance to saponification leads, compared to monomeric plasticizers, to higher stability of measured values of alkaline solutions. The incorporation of lipophilic anions is indispensable for the repulsion of elements of negative charge. The membrane systems' selectivity is strongly influenced by the ligand's exolipophilic periphery. Several membrane systems attractive for analysis are specified.

Notes: The lifetime of cation selective carrier-PVC-membranes partially depends on the components' remaining in the membrane. An exchange of monomeric by polymeric plasticizers with low tendency to migrate lengthens the function time drastically. Other than for Na+ - and NH4+-selective membranes, it is essential for K+ - and Ca2+-selective membranes and optional for H+-selective membranes to incorporate lipophilic anions in order to make the phase transfer catalysis more efficient. The resistence to saponification of phthalic acid polyester gives H+-selective membranes a high stability of measured values even in the alkaline range. For anion selective PVC-membranes, instead of cation selective plasticizers the plasticizing qualities of a liquid charged ligand should be used. The tubular carrier-PVC-membranes of our ion selective flow through measuring systems are diffusion welded to the ends of two PVC-tubes [1] so that they are absolutely tight with no risk of potential leakage. Migration of the membrane components plasticizer and ionophore across this border as well as their extraction [2, 3] into the measuring solution [4] will naturally reduce the membrane's functionning time [5].

Notes: Biosensors Based on Catalase Membranes with Utilization of Enzymatic Bifunctionality for Molecular-selective Carrier-Analysis of EthanolA robust bioelectrochemical measuring system for amperometric electroanalysis of ethanol is presented, which allows measurements between 0.01 and 99.8 vol.% of ethanol without preceeding dilution. It is distinguished by long-lived biosensors based on bifunctional catalase membranes. Besides the O2-sensitive enzymatic biosensor the design includes components for intermediate carrier analysis such as a microdialysator for analyte uptake into buffered, H2O2 containing carrier solution out of the sample solution both with tangential countercurrent fluxes, an oxigenating pump for constant pO2 in the carrier stream, a pressure adjusting chamber as protection from variing pressure on the enzyme membrane resulting from the roller pump's pulsations. Utilization of the membrane's catalatic and peroxidatic bifunctionality is the basis of ethanol analysis: peroxidatically acting catalase molecules cannot release oxygen by catalatic reaction of H2O2 at the same time.

Notes: Bioelectrochemical 2-Propanol Biosensors2-Propanol biosensors for O2-detectors based on bienzyme membranes with sequentially installed catalase and alcohol oxidase are described. Measurements were performed in undiluted media up to 20 vol.-% 2-propanol according to the principle of intermediate carrier analysis by use of a micro-dialysator, an oxygenating pump and a pressure adjusting chamber. For the extension of the measuring scale in the enzyme membrane O2 is released out of hydrogen peroxide within the phosphate buffered carrierstream by catalase. Therefore, oxygen is available to the serially installed alcohol oxidase for the enzymatic substrate transformation beyond the physically soluted part of oxygen in the measuring medium. Molecular selectivities of bienzyme membranes are described for normally used moisturizing concentrates in printeries.

Notes: Summary The measurement of chloride in heparinized blood by means of an ion-selective disc electrode in a flow-optimized flow-through system is compared with other established methods in clinical chemistry for Cl− measurements in serum, e.g. coulometry and mercurimetry. The methods are shown to correlate well. The direct blood measurement of Cl− prevents falsification of data due to sample preparation or due to long retention periods such as may occur in serum samples with measurements using ion-selective crystal membrane electrodes. Calibration is effected by means of aqueous solutions of NaCl under addition of hydrogencarbonate. Graphs prove the stability of potential.

Notes: Electroanalytical flow-through measurements with L-lactate- and β-D-glucose-biosensors using reference sera showed that only by an ion-impermeable but gas-permeable PTFE membrane separating enzyme membrane and electrochemical measuring cell, the high selectivity of oxidases can fully be utilized. For clinical-chemical serum analysis biosensors with O2-detectors are to be preferred which are at least equivalent to photometric comparing methods. Too high values found in measurements using H2O2-detectors are caused by undesired reactands being anodically converted. At O2-sensitive enzymatic L-lactate- and β-D-glucose-biosensors, inevitable biological fouling by O2-consuming microorganisms can be controlled by the use of glutardialdehyde.

Notes: Ion-Selective Flow-Through Sensors with Solid-State Contacts Insensitive to Light and Oxygen for Plant Physiological On-Line Electroanalysis of Transmembraneous Ionic Transports During PhotosynthesisA bioelectrochemical multiple measurement system with ion-selective flow-through sensors for determination of pH, ionic activities and concentrations of transmembraneous ionic transports is described. Non-invasive electroanalysis during photosynthesis was realized by development of new solid-state contacts insensitive to light and oxygen. Continuous measurement of Ca2+, K+, Cl- and pH demonstrates the known intake of potassium ions by Vicia faba L. guard cells under the influence of light. Glucose intensifies the effect.
