    Notes: Abstract Pick’s disease (PD) is characterized by severe neuronal loss and gliosis in a frontotemporal lobar distribution, often associated with Pick bodies and ballooned neurons. Abnormal tau metabolism has been implicated in the pathogenesis of PD; however, the underlying mechanism of neuronal degeneration remains poorly understood. Evidence from other neurodegenerative diseases has suggested that DNA damage and apoptosis may play a major role in cellular degeneration and death. In the present study, an in situ nucleotidyl transferase assay (ISNTA) was used to identify DNA fragmentation in three cases of classical PD with Pick bodies and ballooned neurons, and two PD “variants”, one with ballooned neurons only and the other without Pick bodies or ballooned neurons. In all cases large numbers of ISNTA-positive neurons were present in anatomic regions having obvious degenerative changes (neuronal atrophy and loss, gliosis, cytoplasmic inclusions) by conventional histology. There was no clear association between neuronal DNA fragmentation and the presence of structural abnormalities such as Pick bodies or ballooned cytoplasm. ISNTA-positive glia were present in both cortex and subcortical white matter. Morphologic evidence of apoptosis was not detected in either neurons or glial cells. We suggest that DNA fragmentation in PD and probably other neurodegenerative disorders most likely specifies a population of potentially vulnerable cells in which both cell death and repair mechanisms have been activated. It is likely that only a very small number of these vulnerable cells at a given time will proceed to cell death; however, it is uncertain whether this occurs by apoptosis or some other mechanism.

    Notes: Summary The liquid crystalline gel phases of solutions of Pluronic F127, a triblock copolymer, were recently introduced as an alternative to disordered solutions of random coil polymers as replaceable media for capillary gel electrophoresis (CGE). Pluronic F127, from BASF, is a copolymer of poly(ethylene oxide) and poly(propylene oxide) with the approximate formula (EO)106 (PO)70 (EO)106. Polymer chains aggregate into spherical micelles in aqueous solutions, with poly(propylene oxide) chains creating a hydrophobic core surrounded by brushes of hydrated poly(ethylene oxide) tails. Crowding at high concentrations promotes ordering of micelles. Solutions in the range of about 14–24 % polymer are self-supporting, gel-like cubic liquid crystals at 25–30°C, but when cooled they become low viscosity liquids that are easily loaded into capillaries. This article reviews applications of Pluronic F127 media for capillary gel electrophoresis separations of nucleic acids of several types including oligonucleotides, double stranded DNA fragments, and supercoiled plasmid DNAs.

    Notes: A major cost consideration in the use of anaerobic digestion to convert biomass and waste to utility-grade gas is the expense of separating CO2 from the product gas. Anaerobic digestion has a number of inherent properties that can be exploited to increase the methane content of the gas directly produced by the digester, the most important of which is the high solubility of CO2(40-60 times that of methane) in water under digestion conditions. The methane enrichment concept examined in this study involved the recirculation of a liquid stream from the digester through a CO2 desorption process and the return of the liquid stream back to the digester for absorption of additional CO2 produced by the conversion of organic materials. A steady-state equilibrium model predicted that a digester gas methane content exceeding 94% could be achieved with this scheme using modest recirculation rates provided a desorption process could be designed to achieve a 60+% CO2 removal efficiency in the degassing of the liquid recycle stream. Using fixed-film laboratory digesters operated on synthetic feedstocks, the technique of methane enrichment was tested under pressurized and unpressurized conditions. A 93 + 2% methane gas stream was produced from a volatile-acid-fed bench-scale digester simulating the methanogenic stage of two-phase digestion under conditions of (1) a pH swing achieved without caustic addition that allowed digestion at pH 7. 5 and air stripping at pH 6. 5-7. 0, (2) digester pressurization to 30 psig, and (3) a recycle rate of 0. 33 L/L reactor/day. Significant but lower levels of methane enrichment were achieved with the single-stage digester at the low experimental recycle rate. However, the narrow range among all experiments of CO2 desorption efficiencies achieved in air stripping the recycle stream (35-60% CO2 removal) suggests that comparable methane enrichment-may be achieved with unpressurized single-stage digestion using greater recycle rates. A materials balance analysis of data from an unpressurized, single-stage digester employing no chemical addition and using laboratory degassing efficiencies indicated that 94% methane could be produced at recycle rates of less than 1. 4 L/L reactor/day with a methane loss of less than 2%.

    Notes: A physicochemical model for vapor-liquid equilibrium in multicomponent formaldehyde-containing mixtures (Maurer, 1986; Hasse et al., 1990; Hasse and Maurer, 1991a). is extended to describe enthalpy changes upon vaporization. Predicted enthalpy changes are compared with new experimental results. The measurements were carried out with a thin-film evaporator flow-calorimeter (Liu and Maurer, 1991). More than 80 experimental data points for mixtures of formaldehyde + water and formaldehyde + methanol + (small amounts of water) are reported. The data cover the temperature range from 323 to 363 K (formaldehyde + water) and 312 to 347 K (formaldehyde + methanol) at liquid phase formaldehyde mole fractions up to about 0.3 (formaldehyde + water) and 0.6 (formaldehyde + methanol). Comparisons between predicted and experimental results for the enthalpy change reveal deviations of only a few percent. Further improvements are achieved by fitting some model parameters, which originally were estimated from noncalorimetric data.

    Notes: High-gradient magnetic separation (HGMS) is an existing commercial technology which promises to be an effective means for the large-scale separation of both weakly magnetic and practically nonmagnetic, micron-sized particles. It utilizes the strong magnetic forces created typically by placing filamentary ferromagnetic packing materials inside a separator matrix magnetized by a uniform background magnetic field. In this paper, a practical mathematical model for HGMS is developed for quantitatively predicting the grade and recovery of the separated product and the capacity (concentration breakthrough) of the separator. Computer implementation and experimental verification of the new model for HGMS applied to pilot-scale coal beneficiation are described in Part III.

    Notes: A simple heuristic method for the systematic synthesis of initial sequences for multicomponent separations is proposed and applied to a number of synthesis problems which have been solved previously using other methods. Based on reported costs, it is shown that the initial sequences synthesized for the test problems by the new heuristic method are cheaper than those obtained by other ordered heuristic methods. These initial sequences are also either identical to or at most a few percents higher in costs than those optimum sequences obtained by other algorithmic, heuristic-algorithmic and heuristic-evolutionary methods. The new method is straightforward to apply by hand and it does not require any mathematical background and computational skill from the user.
