    Notizen: A phenomenological model of biocide action against microbial biofilms was derived. Processes incorporated in the model include bulk flow in and out of a well-mixed reactor, transport of dissolved species into the biofilm, substrate consumption by bacterial metabolism, bacterial growth, advection of cell mass within the biofilm, cell detachment from the biofilm, cell death, and biocide concentration-dependent disinfection. Simulations were performed to analyze the general behavior of the model and to perform preliminary sensitivity analysis to identify key input parameters. The model captured several general features of antimicrobial agent action against biofilms that have been observed widely by experimenters and practitioners. These included (1) rapid disinfection followed by biofilm regrowth, (2) slower detachment than disinfection, and (3) reduced susceptibility of microorganisms in biofilms. The results support the plausibility of a mechanism of biofilm resistance in which the biocide is neutralized by reaction with biofilm constituents, leading to a reduction in the bulk biocide concentration and, more significantly, biocide concentration gradients within the biofilm. Sensitivity experiments and analyses identified which input parameters influence key response variables. Each of three response variables was sensitive to each of the five input parameters, but they were most sensitive to the initial biofilm thickness and next most sensitive to the biocide disinfection rate coefficient. Statistical regression modeling produced simple equations for approximating the response variables for situations within the range of conditions covered by the sensitivity experiment. The model should be useful as a tool for studying alternative biocide control strategies. For example, the simulations suggested that a good interval between pulses of biocide is the time to minimum thickness. © 1996 John Wiley & Sons, Inc.

    Notizen: Plasmid gene product accumulation in a cell population depends on the fraction of plasmid-containing cells and the distribution of single-cell plasmid content. These important population properties have been related to plasmid replication regulation and kinetics and to plasmid segregation rules at the single-cell level using population balance mathematical models. Budding yeast populations are considered in detail because of the practical potential of yeast host-vector systems and because of the model complications introduced by the asymmetric division pattern observed for Saccharomyces cerevisiae at all but the largest growth rates. Solutions are presented for several different reasonable models of plasmid replication and segregation. The results offer potential for identification of important qualitative features of yeast plasmid replication and of model parameter values from average and segregated experimental data on yeast populations.

    Notizen: Periodic environmental shifts have been used to induce synchrony in many different microbial populations. In this article, the induction synchrony phenomenon is analyzed using an age distribution model in which the age at which the cells divide is subjected to periodic forcing. It is found that synchrony will occur whenever the period of the forcing lies in the interval between the youngest and the oldest division age that occur in the population during the forcing. The analysis also predicts that under certain conditions it should be possible to obtain a multimodal synchrony in which cells in the population are distributed among a set of discrete, synchronized cell lines. The behavior of the age distribution when the conditions for synchrony are not satisfied is briefly explored. It is found that the age distribution model is able to exhibit a very rich spectrum of possible dynamic behavior. Many of the phenomena observed can be thought of in terms that are familiar from nonlinear analysis, such as stable and unstable limit cycles, period doubling, halving, and chaos. The richness of dynamic behavior opens the possibility that environmental shifts or periodic forcing could be used as a powerful tool in discriminating models of microbial kinetics and cell cycle control.

    Notizen: An inducible β-galactosidase from an extremely thermophilic organism, Thermus strain 4-1A, has been isolated and partially purified. There were significant dissimilarities to T. aquaticus β-galactosidase. It had a pl of 4.5, was inhibited by sulphydryl inhibitors and a number of transition metals, and was activated by EDTA and SH-containing reagents. The β-galactosidase showed strong product inhibition, and weaker inhibition by some other mono- and disaccharides. It was very stable up to 90°C at pH 8. On immobilization by diazonium linkage to porous glass, the pH optimum (6.0), the KM with ONPG (5mM) and the product inhibition were not altered.

    Notizen: A new method for manipulating the steady-state behavior of a mixed culture is introduced. The method makes use of differences in adherence properties between competing populations to maintain a desired population ratio. The very specific nature of some ligand-to-cell interactions allows precise manipulation of even closely related populations. The control method is illustrated by analysis and simulations of models of a competitive mixed culture and a culture of an unstable recombinant organism. In both cases, retention of the disadvantaged population via cell adhesion results in creation of a stable coexistence steady state over a range of operating conditions.

    Notizen: A chemostat with population specific recycle was employed to alter the dynamics of a competitive mixed culture of Escherichia coli. Based on differential expression of a functional maltoporin, the two populations were separated by specific adhesion on starch-Sepharose. The slower growing population was the recycled to the reactor. The specific recycle was successful in maintaining the slower growing population at a high level than in comparable reactors without recycle.

    Notizen: By means of a multiplex polymerase chain reaction (PCR) we amplified rearranged T-cell receptor γ chain genes to detect monoclonality in 370 formalin-fixed skin biopsy specimens, showing histological features of parapsoriasis or mycosis fungoides. PCr products were analyzed by temperature gradient gel electrophoresis (TGGE). We selected 20 positive cases for use in a comparison of this technique with conventional agarose and polyacrylamide gel electrophoresis (PAGE). With TGGE the T-cells had shown monoclonality in 272 of the 370 cases; with agarose electrophoresis they did so in only 5 of the 20 selected cases and with PAGE in 16. Where multiple biopsy specimens from the same patient were analyzed, PCR products showed identical rearrangement patterns in TGGE. TGGE is an efficient technique that works on routine material and can help to verify a histological diagnosis of cutaneous T-cell lymphoma.

    Notizen: Various chemometric experimental designs have been employed for the optimisation of capillary electrophoresis (CE) methods. Similar designs have been utilised in the assessment of the robustness of CE methods. The designs employed include central composites, fractional factorials, Plackett-Burman, simplex and overlapping-resolution mapping. Optimisation studies have largely concentrated on the use of these designs on selection of the optimal electrolyte composition. The robustness testing studies performed have involved the use of screening designs to identify the critical parameters affecting responses such as migration times and resolution. Further designs such as central composites have then been employed to set method limits following robustness studies. It is concluded that the use of experimental designs and statistical data evaluation in conjunction with personal computer-controlled CE autosamplers and instruments are of great benefit in the optimisation and robustness evaluation of CE methods.
