    Notizen: The rate constants for [1O2] [MCLA] and [1O2][NaN3] were measured by quenching the near-infrared emission (1Δg→3∑g) in steady state with MCLA and NaN3, respectively. 1O2 was constantly generated by energy transfer to O2 from Ar laser-excited Rose Bengal. The Stern - Volmer plots yielded the second-order rate constants of 2.94 × 109 M-1 S-1 and 3.83 × 108 M-1 S-1 for quenching 1O2 with MCLA and NaN3 in water at pH 5.4, respectively. The 1O2 + MCLA reaction emitted light with maximum at 465 nm at pD 4.5 identical to the O2- + MCLA reaction.

    Notizen: Continuous ultrafiltration of synthetic waste water containing peptone was carried out by using positively and negatively charged polyacrylonitrile membranes. The filtration experiments were operated in cross-flow mode under 10 kPa of applied pressure. The filtration properties of the charged membranes were compared with those of uncharged polyacrylonitrile ultrafiltration membranes having similar molecular sieve characteristics and membrane structure to the charged ultrafiltration membranes. During the continuous filtration, the filtration rate decreased and the operation pressure increased because of the formation of a peptone gel layer on the membrane surface. It was found that, for the positively charged membrane, the decrease in the filtration rate of the charged membranes was smaller than that of the uncharged membrane. In addition, the positively charged membrane maintained the initial operation pressure during the filtration. The reduction of fouling in the positively charged membrane was discussed by analyzing the gel components on the membrane surface.

    Notizen: Glycosaminoglycans including heparin, heparan sulfate, chondroitin sulfate, dermatan sulfate and hyaluronic acid were analyzed by reversed polarity capillary electrophoresis. Detection was achieved at 240 nm based on the formation of a copper (II) complex in copper sulfate solution at low pH. Glycosaminoglycans having a high ratio of iduronic acid to glucuronic acid, as well as ones having lower molecular weight, gave the highest detection sensitivity using this method. Detection of the copper (II)-heparin complex is extremely sensitive, permitting the analysis of as little as 10-9 g. This method was also successfully applied to the analysis of heparin oligosaccharides that lacked a chromophore prepared from heparin using controlled, low pH nitrous acid depolymerization.

    Notizen: Mitochondrial inner membrane proteins extracted from beef heart tissue were examined for reactivity to antimitochondrial antibody (AMA) present in sera of patients with primary biliary cirrhosis (PBC) by an immunoblotting technique. Four proteins, which reacted with AMA, had molecular masses of 70 kDa, 50 kDa, 47 kDa and 40 kDa, as defined by their relative mobility (Rf) in sodium dodecyl sulfate-polyacrylamide gel electrophoresis. All sera of 114 PBC patients were positive with at least one and as many as four of the mitochondrial proteins. The major antigenic proteins of mitochondrial inner membrane to which AMA reacts were the 70 kDa and 47 kDa proteins. All PBC sera containing antibodies to the 50 kDa and/or 40 kDa proteins reacted with 70 kDa as well. The isolation of antigen reacting with AMA of PBC is important to warrant further study of AMA and the cause of the disease. The isolation of responsible antigens had been difficult because the four antigens were insoluble. However, the antigen newly found by us, the 36 kDa fragment, obtained by partial trypsin digestion, is soluble. Using several procedures, the antigenic protein target of AMA was purified from mitochondria for the first time. We determined the N-terminal sequence of the soluble 36 kDa fragment, 25 residues in length. Until now the N-terminal sequence of the 36 kDa protein has not shown significant homology with any known protein. The present results of antigen purification would contribute to the elucidation of the epitopes of AMA antigen.
