    Notes: Atypical antipsychotic drugs (APDs), all of which are relatively more potent as serotonin (5-HT)2A than dopamine D2 antagonists, may improve negative symptoms and cognitive dysfunction in schizophrenia, in part, via increasing cortical dopamine release. 5-HT1A agonism has been also suggested to contribute to the ability to increase cortical dopamine release. The present study tested the hypothesis that clozapine, olanzapine, risperidone, and perhaps other atypical APDs, increase dopamine release in rat medial prefrontal cortex (mPFC) via 5-HT1A receptor activation, as a result of the blockade of 5-HT2A and D2 receptors. M100907 (0.1 mg/kg), a 5-HT2A antagonist, significantly increased the ability of both S(–)-sulpiride (10 mg/kg), a D2 antagonist devoid of 5-HT1A affinity, and R(+)-8-OH-DPAT (0.05 mg/kg), a 5-HT1A agonist, to increase mPFC dopamine release. These effects of M100907 were abolished by WAY100635 (0.05 mg/kg), a 5-HT1A antagonist, which by itself has no effect on mPFC dopamine release. WAY100635 (0.2 mg/kg) also reversed the ability of clozapine (20 mg/kg), olanzapine (1 mg/kg), risperidone (1 mg/kg), and the R(+)-8-OH-DPAT (0.2 mg/kg) to increase mPFC dopamine release. Clozapine is a direct acting 5-HT1A partial agonist, whereas olanzapine and risperidone are not. These results suggest that the atypical APDs via 5-HT2A and D2 receptor blockade, regardless of intrinsic 5-HT1A affinity, may promote the ability of 5-HT1A receptor stimulation to increase mPFC DA release, and provide additional evidence that coadministration of 5-HT2A antagonists and typical APDs, which are D2 antagonists, may facilitate 5-HT1A agonist activity.

    Notes: Changes in the proportion of benzimidazole-resistant strains in populations of Venturia nashicola were monitored in pear orchards. Successive applications of thiophanate-methyl increased the level of resistance in the populations. When the application of benzimidazoles was stopped and other fungicides were applied, the proportion of highly resistant strains gradually decreased and the proportion of intermediately resistant, weakly resistant and sensitive strains increased. It is suggested that this phenomenon is an example of genetic homeostasis within microbial populations.

    Notes: Experiments are reported on two techniques for melt-texture processing Ba2YCu3O6.5 by directional solidification from a semisolid melt containing particles of BaY2CuO5 and a copper-rich liquid. One of these employs an electric resistance furnace with ambient or oxygen enriched atmosphere; the other is a laser-heated furnace operating at 1.3 atm oxygen. Solidification interface morphologies and other structural features were examined in quenched specimens. Depending on growth rate and temperature gradient, three different types of growth morphologies of the growing 123 phase were observed: "faceted plane front,'' "cellular dendritic'' or "equiaxed blocky.'' The interface temperature decreased markedly with increasing growth rate for the faceted plane front specimens. In the remaining specimens, solidification took place over a range of temperatures. The temperature of the "root'' of the solidification front dropped, but temperature of the solidification front "tip'' did not. A solidification model is developed and employed to interpret experimental observations. The model assumes limited diffusion of solute in the liquid during the growth of the superconducting phase. The model shows, in agreement with experiment, that growth rate of the low temperature solid phase has a strong effect on ability to obtain the desirable faceted plane front, and that thermal gradient has only a small effect. Interparticle spacing of the high temperature phase, BaY2CuO5, is also predicted to have a strong effect.

    Notes: Abstract Antifungal activity of the novel compound acibenzolar-S-methyl (CGA245704: benzo[1,2,3]thiadiazole-7-carbothioic acid S-methyl ester) was examined in vitro. No remarkable activity was observed on mycelial growth and conidial germination of almost all fungi tested. Only melon isolates of Didymella bryoniae were sensitive to this compound. On potted plants, acibenzolar-S-methyl showed control efficacy on anthracnose and scab of cucumber and rust of Japanese pear but not on Fusarium wilt of cucumber. In field trials, the occurrence of both rust and scab on Japanese pear was suppressed with this compound. Based on these experiments, it was suggested that acibenzolar-S-methyl induced resistance to some but not all diseases on cucumber and Japanese pear. Induction of disease resistance in cucumber was rapidly triggered after treatment with acibenzolar-S-methyl.

    Notes: Mitochondrial inner membrane proteins extracted from beef heart tissue were examined for reactivity to antimitochondrial antibody (AMA) present in sera of patients with primary biliary cirrhosis (PBC) by an immunoblotting technique. Four proteins, which reacted with AMA, had molecular masses of 70 kDa, 50 kDa, 47 kDa and 40 kDa, as defined by their relative mobility (Rf) in sodium dodecyl sulfate-polyacrylamide gel electrophoresis. All sera of 114 PBC patients were positive with at least one and as many as four of the mitochondrial proteins. The major antigenic proteins of mitochondrial inner membrane to which AMA reacts were the 70 kDa and 47 kDa proteins. All PBC sera containing antibodies to the 50 kDa and/or 40 kDa proteins reacted with 70 kDa as well. The isolation of antigen reacting with AMA of PBC is important to warrant further study of AMA and the cause of the disease. The isolation of responsible antigens had been difficult because the four antigens were insoluble. However, the antigen newly found by us, the 36 kDa fragment, obtained by partial trypsin digestion, is soluble. Using several procedures, the antigenic protein target of AMA was purified from mitochondria for the first time. We determined the N-terminal sequence of the soluble 36 kDa fragment, 25 residues in length. Until now the N-terminal sequence of the 36 kDa protein has not shown significant homology with any known protein. The present results of antigen purification would contribute to the elucidation of the epitopes of AMA antigen.
